        Development of the Beamline Front End at the Pohang Light Source(PLS)-II

        The third-generation synchrotron radiation accelerator has been providing beams to users since 1995 and have been made to upgrade the performances of the accelerators and the experimental devices. For that reason, the PLS-II was started in 2009 as a three-year project. An important feature is the extension of the ID (insertion device) beamline and the ecient placement of the storage ring magnet structure. The overall structure of the beamline as well as the length and structure of the front end had to be changed to expand the ID beamline. Also, the safety of many vacuum parts that would be exposed to the large heat due to the increase in the beam energy was rstly considered in the design, and an analysis was performed in parallel. This paper describes the overall structure of the front end, including the design and analysis of the vacuum components in the Pohang Light Source(PLS)-II beamline.

      • Comparing characteristics of agricultural and physiological of ‘Iksan370’ and ‘Keumkang’

        Characteristics of agronomy, especially good yield productivity is very important in wheat (Triticum aestivum L.) as human food crop. Development of elite cultivar having good traits is required for food stability. In 2013, a new wheat line called Iksan370 has been developed by the National Institute of Crop Science, RDA. Iksan370 has hard red winter wheat characteristics and better features like resistance to biotic, abiotic stress and higher yield ability than Keumkang. Heading and maturing date were delayed about ten days and period to flowering date was shorter three or four days than Keumkang. Culm and spike length of Iksan370 were 95cm and 15.3cm, averagely. This means that culm and spike length were longer than Keumkang (79cm and 7cm, respectively). It showed higher milling rate (75.5%), protein content (13.5%) and SDS-sedimentation volume (77.5ml) than Keumkang. Genotypes of six genes, GBSSA, B, D, PPO18, Pina and Pinb, were represented that were similar to Keumkang, respectively. Average yield of Iksan370 in the preliminary trial test was 6.61 tons/ha in upland. We expect that yield productivity and farm income were increased by Iksan370.

      • Development of Molecular Marker for Korean Wheat Cultivar Identification using AFLP Technique

        Amplified fragment length polymorphism (AFLP) is one of molecular marker technique based on DNA and is extremely useful in detection of high polymorphism between closely related genotypes like Korean wheat cultivars. Six Korean wheat cultivar specific marker sets have been developed from inter simple sequence repeat (ISSR) analysis and we can identify the 13 Koran wheat cultivars form other cultivars using six that (Son et al., 2013). We used four combinations of primer sets in our AFLP analysis for developing additional cultivar specific markers in Korean wheat. Twenty-one of the AFLP bands were isolated from ACG/M-CAC primer combination and 19 bands were isolated from E-AGC/M-CTG primer combination, respectively. We used forty bands to design sequence characterized amplified region (SCAR) primer pairs for Korean wheat cultivar identification. Only one of 40 amplified primer pairs, C2, were able to use for wheat cultivar identification. The DNA band of 215bp length was amplified by C2 primer pairs in ten cultivars, Eunpa, Olgeuru, Gobun, Saeol, Milsung, Sinmichal, Jokyung, Sugang, Goso, and Joah. Then C2 primer was applied to these primer sets as newly SCAR marker, six cultivars are identifying from other cultivars, additionally. Finally, to use the C2 and six primer sets, 19 Korean wheat cultivars are identified.

        저층 RC 건물의 내진성능 보강에 관한 실험적 연구

        Nowaday, most of the low-rise concrete structures which have less than five stories were built before the intensified seismic code was established 2005. According to the fact that our country is not a safety zone ay more, studies are need to reinforce the seismic performance of that structures. The basic frame of low-rise structure are consist of beams and columns with partition walls, therefore that are very weak about secondary wave of earthquake because of the high stiffness. The partition wall are consist of open channel for sunlight or ventilation and intermediate wall. The intermediate walls will enhance the stiffness of columns, but will cause shear failure with short column effects because of the reduced effective depth. But we don't have studies and adequate design code for partition wall effects, therefore some more studies are need for these facts. 현재 우리나라에서 5층 이하로 건축된 저층 철근콘크리트 건물의 대부분은 2005년 국내건축구조물의 내진설계기준이 강화되기 이전에 설계 및 시공이 이루어졌음으로, 더 이상 지진의 안전지대가 아닌 것으로 인식된 우리나라도 이들에 대한 내진성능 보강방안에 대한 연구가 필요한 실정이다. 저층 구조물의 기본 골조는 대부분 보와 기둥에 칸막이 벽으로 이루어져 있으므로 강성이 커서 지진의 횡파에 매우 취약하다. 칸막이 벽은 채광 과 환기를 위한 개구부와 그 아래 허리벽으로 구성되어 있다. 허리벽은 기둥의 강성을 증가시키지만 유효길이를 감소시켜 단주효과를 유발시키며 지진발생 시 기둥의 전단파괴를 야기할 수 있다. 그러나 현재 국내에서는 칸막이 벽에 대한 연구가 많지 않고 적합한 설계기준도 없는 실정이므로 이에 대한 연구가 필요하다고 사료된다.

        내진설계를 위한 공동주택의 고유주기 측정에 관한 연구

        Recently, apartment designs are tend to be changed from the standard pattern by many causes and the beam-column structures are getting popular instead of wall structure system. Therefore, for the effective use of planed space the heights of apartment are tend to be higher and higher. According to Korea Building Code, earthquake resistance designs or seismic design for those high rise apartments must be more attentive and accurate, especially, dynamic periods of structures must be exactly measured, because those are very important for equivalent static analysis. The important subject of this study is to investigate the safety factors and seismic performance for natural period of high rise buildings by comparing the natural periods getting from ambient vibration method with those of Korea Building Code. 최근 공동주택은 여러 가지 원인에 의해 그 형태가 변화되고 있으며, 구조물도 벽식 형태의 구조물에서 보-기둥 형태의 구조물로 선호도가 바뀌고 있다. 따라서, 계획된 공간을 효율적으로 이용하기 위하여 구조물은 점점 더 고층화 되어가는 경향이 있다. 한국건축구조기준(KBC)에서는 이러한 고층 공동주택의 경우 내진설계 시 지진하중을 산정할 때, 구조물의 진동특성은 매우 신중하고 정밀하게 측정해야 하며, 특히 동적주기는 동적 주기는 이에 상응하는 정적 해석 시 매우 중요하므로 정밀하게 측정해야 한다. 본 연구의 주 목적은 상시 미진동 실험으로부터 얻어진 기타골조식과 전단벽식을 고려하는 경우 고층빌딩의 고유주기를 한국건축구조기준의 값과 비교하여 안전계수를 산정하고 내진성능을 평가하여, 보다 경제적인 내진설계방안을 제시하는 것이다. 또한, 연구결과 기타 골조 식은 매우 보수적으로 주기를 산정하는 것으로 나타나므로 내진설계 시 이러한 사항도 고려해야 할 것으로 사료된다.

      • 사람의 제대혈관 장력에 미치는 DMSO(Dimethyl Sulfoxide)의 효과

        Starting from 1960s through to 1980s, dimethyl sulfoxide (DMSO) earned fame as a valuable agent that could be used for many purposes including patient therapy. DMSO is dipolar aprotic compound endowed with unique physical and chemical properties. It is a potent solvent for a broad spectrum of solutes. DMSO has been known to exert varions biological effects such as diuretic, vasodilatory, analgesic, anti-inflammatory and cardiovascular effects. Also DMSO has been reported to reduce intracranial pressure and improve cerebral blood flow in experimentally induced ischemia and vasospasm. Some studies presented a possibility that may alter vascular smooth muscle tension. On the other hand, human umbilical vessels are ideal for the study of vasoactive substances because of poor innervation of autonomic nerves to umbilical vessels. This study was undertaken to investigate effects of DMSO on the vascular tension of human umbilical vein (HUV) and artery (HUA). DMSO(1-10%) relaxed HUV and HUA preparations precontracted with KCl (3×10^-2M) in a concentration-dependent manner. At all concentrations, vasodilating effect of DMSO was greater in HUV than HUA. Pretreatment with methylene blue (5×10^-6M), soluble guanylate cyclase inhibitor and L-NAME (10^-4M), NO synthase inhibitor, did not alter relaxing action of DMSO. After pretreament with indomethacin, cyclooxygenase inhibiotor, the DMSO-induced relaxing response was partially suppressed only in HUA (41 ± 5.7%). Following chemical denudation with saponin (0.03mg/ml, 45min), the DMSO-induced relaxing responses of both umbilical vessels were not changed. These results suggest that the DMSO induces an endothelium-independent relaxation both in the human umbilical artery and vein, and that the DMSO-induced relaxing response of HUA results, at least in part, from the mediation of prostanoids.

        자원봉사자의 기능적 동기 및 자원봉사센터의 관리관행이 조직성과에 미치는 영향

        사회적 서비스에 대한 수요가 늘고 비영리 조직의 운영이 자원봉사자의 활동에 의존하고 있기때문에 자원봉사자들을 활용하는 기관의 입장에서는 동기가 유발된 자원봉사자들을 모집하고 이들이 지속적으로 활동하도록 할 방안을 모색하는 일이 중요한 과제로 부상하였다. 이제까지 자원봉사자들의 이탈을 막고 자원봉사자로서 오래 체류하도록 할 방안에 관한 연구가 많이 이루어져 왔지만 효과적인 요인을 찾아내는 데 미비하였다. 따라서 자원봉사자들의 활동지속과 친사회적 행위를 강화할 방안의 마련이 학계에서나 실무계에서 중요한 과제가 되고 있다. 이 논문의 목적은 자원봉사 활동에 참여하려는 자원봉사자들의 기능적 동기수준과 자원봉사 센터의 관리관행이 자원봉사자들의 활동지속의지와 친사회적 행위를 하위요인으로 한 조직성과에 미치는 영향을 조사하고, 자원봉사자들의 봉사활동만족이 이들 사이에 매개역할을 하는 지를 검증하는 일이다. 이러한 연구 목적을 수행하기 위해 이 연구는 기능적 접근(functional approach)에 근거하여 자원봉사자들의 행위를 분석한다. 본 연구의 가설 검증을 위해 국내에서 활동하고 있는 자원봉사자들로부터 회수한 유효 설문지 1,227 개를 분석한 결과 발견한 중요한 연구결과는 다음과 같다. 첫째, 자원봉사자들의 기능적 동기 수준 및 자원봉사센터의 관리관행에서 자원봉사자 지원적 경향이 자원봉사자들의 조직성과에 유의한 긍정적 영향을 미친다. 둘째, 위와 같은 영향에 자원봉사자들의 활동만족이 유의한 매개역할을 하는 것으로 나타났다. 아울러 본 연구의 함축적 의미와 연구의 한계점이 제시되었다. As the needs for social service are increasing and nonprofit organizations depend on volunteers, it becomes very important to prepare some ways to recruit the well motivated volunteers and to enhance their intentions to remain and their prosocial behaviors. Particularly there have been many studies concerning volunteer retention and many factors have been identified. However, literature survey indicates that many of these earlier studies did not explain sustained volunteerism, which means there must be other factors important in influencing the prediction of durability as a volunteer and the level of pro-social behavior toward other volunteers and their organizations. From this respect the present study intends to explore more effective factors in the prediction of volunteers’s retentions and their pro-social behaviors. Another purpose is to analyse the effect of volunteers’s satisfaction as mediating variable on the relationship between the independent variables(volunteers’s functional motivation and managerial practices of volunteer center) and dependent variable. This study is based on a functional approach to understanding the motivations that prompt people to become volunteers and that sustain their efforts overtime and pro-social behaviors toward other volunteers. In order to address the hypotheses and research questions, a questionnaire survey was conducted and the effective 1,227 cases responded by volunteers from diverse regions in South Korea were used for a statistical analysis. The research findings are summarized as followings: First, functional motivators had a significantly positive influence on organizational effectiveness comprised of volunteers’ intention to remain as volunteers and on volunteers’s pro-social behaviors. The supportive managerial practices of volunteers administration centers influenced on the organizational effectiveness. Second, volunteers’ satisfaction was influenced by the levels of functional motivators. Third, volunteers’ satisfaction affected directly the organizational effectiveness and mediated the relationship between the independent variables(functional motivators and supportive managerial practices) and dependent variable. The Implications and limitations of the study were suggested.

      • Voltae-activated CI ̄ Conductance in Cultured Human Melanocytes

        Using the whole-ell patch-clamp technique we recorded Cl^(-) currents in cultured human melanocytes. With high symmetrical Cl^(-) solutions melanocytes expressed time-dependent Cl^(-) currents that did not run down for approximately 30 min. The currents showed voltage-dependent activation and reached a steady-state at about 500 msec during hyperpotarizing pulses. Current vs. voltage relation indicates that the current was activated by more hyperpotarizing voltage Steps, showing inward rectification. Based on conductance vs. voltage relation the half activation voltage of the current was determined tobe -60 mV. When tail current amplitudes were plotted as a function of test pulses, tail current reversed at -10 mV, dose to ECI of 0 mV in this condition, indicating that the observed current was specific Cl^(-) current.
