      • 게르마늄 내성효모에 의한 게르마늄의 세포내 축적

        게르마늄 내성 효모를 개발하여, 유기게르마늄을 생합성할 수 있는 최적의 조건을 알아보고, 게르마늄의 효모균체내에서의 생합성을 확인하였다. 1. 게르마늄 내성효모의 성장정도가 일반효모보다 약간 높은 것으로 나타났으며, 내성효모 배양액의 pH는 시간이 경과함에 따라 일반효모의 배양액보다 pH의 상승정도가 큰 것으로 나타났다. 2. 내성효모의 성장정도는 GeO2 의 농도가 90ppm까지는 점차 상승하다가 그 이상에서는 성장정도가 점차 감소하는 추세를 보였으나 일반효모보다는 감소하는 정도가 적은 것으로 나타났다. 3. 배지의 GeO2 농도를 80ppm으로 하고 pH를 8.0으로 조절한 후 30℃ 24시간동안 배양한 효모 중의 게르마늄의 농도가 0.818g/mg으로 가장 높게 나타났다. 4. 효모세포내의 게르마늄의 분포상태를 측정한 결과, 대부분의 게르마늄이 효모세포벽을 구성하는 고분자 다당체나 단백질에 결합된 것으로 추측된다. 또한 게르마늄의 일부는 수용성 단백질과 결합되어 있는 것을 확인할 수 있었다. A mutant of Saccharomyces cerevisiae ACTC 4105 was derived by a series of subcultures in the media containing increasing GeO_2 concentrations. The growth of tolerant yeast cells and the yield of organic germanium was studied as a function of GeO_2 concentrations, temperature and pH of media. The distribution of organic germanium in the tolerant yeast cells incubated in the media containing GeO_2 was also studied.

        The purpose of this study was to investigated exercise type for body composition and cardiovascular function in middle aged womens. Twenty housewives selected as subject and then divided into three group; The health group with 7 persons, swimming group with 7 persons, control group with 6 persons. The results were as follows; 1. Body composition 1) Non significant different was appeared on body density, %fat, fat in among groups. 2) Significant different was appeared on LBM in health group higher than swimming group and control group. 2. Cardiovascular function 1) Significant different was appeared on restHR in health group higher than swimming group and control group. 2) Non significant different was appeared on VO2/kg in among groups. 3) Significant different was appeared on VEmax in health group and swimming group higher than control group. 4) Significant different was appeared on all-out time in health group higher than swimming group and control group.

        Inhibitory Effect of Cordycepin on Human Platelet Aggregation

        Cordycepin separated from Cordyceps militaris is a major physiologic active component in Cordyceps militaris. The platelet aggregation is stimulated by Ca^(2+), which is either mobilized from intracellular endoplasmic reticulum or transported from extracellular space. cGMP antagonizes the actions of Ca^(2+). Based on these facts, we have investigated the effects of cordycepin on the mobilization of Ca^(2+) and the production of cGMP on collagen (10 ㎍/ml)-induced human platelet aggregation. Cordycepin potently stimulated the human platelet aggregation induced by collagen (10 ㎍/ml) in a dose-dependent manner. Cordycepin (500 μM) inhibited also the collagen-induced human platelet aggregation in the presence both 1 mM and 2 mM of CaCl_(2). These are in accord with the results that cordycepin inhibited the Ca^(2+)-influx on collagen-induced human platelet aggregation. These results suggest that cordycepin decrease the intracellular Ca^(2+) concentration to inhibit collagen-induced human platelet aggregation. Besides, cordycepin increased the level of cGMP on collagen-induced human platelet aggregation. This result is related with the decrease of intracellular Ca^(2+) concentration, because cGMP inhibits the mobilization of Ca^(2+). In addition, cordycepin inhibited the human platelet aggregation induced by LY-83583, inhibitor of guanylate cyclase. This result suggested that cordycepin inhibit the platelet aggregation by stimulating the activity of guanylate cyclase. In conclusion, we demonstrated that cordycepin might have the antiplatelet function by inhibiting Ca^(2+)-mobilization via the stimulation of the production of cGMP.

      • Biallelic Deletion of Pxdn in Mice Leads to Anophthalmia and Severe Eye Malformation

        <P>Peroxidasin (PXDN) is a unique peroxidase containing extracellular matrix motifs and stabilizes collagen IV networks by forming sulfilimine crosslinks. PXDN gene knockout in Caenorhabditis elegans (C. elegans) and Drosophila results in the demise at the embryonic and larval stages. PXDN mutations lead to severe eye disorders, including microphthalmia, cataract, glaucoma, and anterior segment dysgenesis in humans and mice. To investigate how PXDN loss of function affects organ development, we generated Pxdn knockout mice by deletion of exon 1 and its 5′ upstream sequences of the Pxdn gene using the CRISPR/Cas9 system. Loss of both PXDN expression and collagen IV sulfilimine cross-links was detected only in the homozygous mice, which showed completely or almost closed eyelids with small eyes, having no apparent external morphological defects in other organs. In histological analysis of eye tissues, the homozygous mice had extreme defects in eye development, including no eyeballs or drastically disorganized eye structures, whereas the heterozygous mice showed normal eye structure. Visual function tests also revealed no obvious functional abnormalities in the eyes between heterozygous mice and wild-type mice. Thus, these results suggest that PXDN activity is essential in eye development, and also indicate that a single allele of Pxdn gene is sufficient for eye-structure formation and normal visual function.</P>

        Effects of Gelatin Hydrolysates Addition on Technological Properties and Lipid Oxidation of Cooked Sausage

        This study investigated the impacts of gelatin hydrolysate addition on the technological properties and lipid oxidation stability of cooked sausage. Gelatin hydrolysate was prepared from pork and duck skin gelatin, through stepwise hydrolysis using collagenase and pepsin. The cooked sausages were formulated without gelatin (control) or with 1% pork skin gelatin, 1% duck skin gelatin, 1% pork skin gelatin hydrolysate, and 1% duck skin gelatin hydrolysate. The pH, color characteristics, protein solubility, cooking loss, and textural properties of cooked sausages were evaluated, and the 2-thiobarbituric acid reactive substances (TBARS) value was measured weekly to determine lipid oxidation stability during 4 wk of refrigerated storage. Enzymatic hydrolysis of gelatin decreased protein content and CIE L* but increased redness and yellowness (p<0.05). When 1% gelatin or gelatin hydrolysate was incorporated in cooked sausage, however, little to no impacts on pH value, moisture content, protein content, color characteristics, protein solubility, and cooking loss were found (p>0.05). The addition of 1% duck skin gelatin hydrolysate increased the cohesiveness and chewiness of cooked sausages. The inclusion of 1% duck skin gelatin accelerated lipid oxidation of cooked sausages during refrigerated storage (p<0.05), whereas duck skin gelatin hydrolysate caused a lower TBARS value in cooked sausage compared to duck skin gelatin. The results show comparable effects of gelatin and gelatin hydrolysate addition on the technological properties of cooked sausages; however, the oxidative stability of raw materials for gelatin extraction should be evaluated clearly in further studies.

      • 아이스하키선수들의 심폐지구력에 관한 비교 연구

        The purpose of this study was to investigate differences of the physical fitness and cardiovascular function between high school Ice-hockey players(HSIP) and university Ice-hockey players(UIP) The subjects are 16 persons respectly. I adopted the physical test and used treadmill to understand the effect of aerobic exercise and to know the physical fitness and developing states. The result of this study are as follows ; 1. Cardiovascular was significant difference appeared in terms of V emax between HSIP 128.25 ± 23.89 ℓ /min, UIP 157.12 ± 28.98 ℓ /min(p$lt;.05), RR between HSIP 52.88 ± 6.15f/min, UIP 64.75 ± 9.44f/min(p$lt;.05). 2. Non significant difference appeared in terms of rest HR between two group, significant difference appeared in terms of HRmax between HSIP 188.25 ± 8.31beats/min, UIP 200.25 ± 7.98beats/min(p$lt;.05). 3. Non significant difference appeared in terms of V˙O₂/HR, V˙O₂max, V˙O₂/kg, V˙CO₂ between HSIP and UIP.

        간편조리세트 원재료의 미생물 오염도 조사

        본 연구에서는 경기도 내 유통 중인 간편조리세트 55건 내 농·축·수산물 원재료의 미생물 오염도를 조사하였다. 55건의 간편조리세트 중 농산물이 원재료로 들어가는 제품은 48건, 축산물이 원재료로 들어가는 제품은 43건, 수산물이 원재료로 들어가는 제품은 16건이었다. 농·축·수산물에서 일반세균은 100%의 검출률을 보였으며, 일반세균 평균 검출량은 농산물 6.57 log CFU/g, 축산물 4.60 log CFU/g, 수산물 5.47 log CFU/g으로 나타났다. 농·축·수산물에서 대장균군은 각각 81.25%, 69.77%, 43.75%의 검출률을 보였고, 대장균군 평균 검출량은 농산물 2.83 log CFU/g, 축산물 1.34 log CFU/g, 수산물 1.12 log CFU/g으로 나타났다. 대장균은 13건(30.23%)의 축산물에서 0.70-2.36 log CFU/g 범위로 검출된 반면, 수산물에서는 1건(6.25%)만 검출되었고, 농산물에서는 검출되지 않았다. 농·축·수산물에서 진균은 각각 97.92%, 93.02%, 93.75%의 검출률을 보였고, 진균 평균 검출량은 농산물 3.82 log CFU/g, 축산물 2.92 log CFU/g, 수산물 2.82 log CFU/g으로 나타났다. 농·축·수산물에서 식중독균은 각각 35.42%, 37.21%, 31.25%의 분리율을 보였고, 바실루스 세레우스, 살모넬라균 등 7종의 45개 식중독균을 분리하였다. 간편조리세트로 인한 식중독 사고 예방을 위하여 세척, 충분한 가열 섭취 및 조리과정 중 교차오염에 대한 주의가 필요하다. This study investigated the microbial contamination of agricultural, livestock, and marine ingredients in 55 meal kits distributed across Gyeonggi-do, South Korea. Of the 55 meal kits, 48 contained agricultural ingredients, 43 contained livestock ingredients, and 16 contained marine ingredients. The detection rate of the total aerobic bacteria in the agricultural, livestock, and marine products was 100%. The average numbers of the total aerobic bacteria were 6.57 log colony-forming units (CFU)/g in the agricultural products, 4.60 log CFU/g in the livestock products, and 5.47 log CFU/g in the marine products. The coliform detection rates in the agricultural, livestock, and marine products were 81.25%, 69.77%, and 43.75%, respectively. The average numbers of coliforms were 2.83 log CFU/g in the agricultural products, 1.34 log CFU/g in the livestock products, and 1.12 log CFU/g in the marine products. Escherichia coli was detected in 13 livestock products (30.23%), with levels ranging from 0.70 to 2.36 log CFU/g. Contrastingly, E. coli was detected in only one marine product (6.25%) and was not detected in any agricultural products. The detection rates of fungi in agricultural, livestock, and marine products were 97.92%, 93.02%, and 93.75%, respectively. The average numbers of fungi were 3.82 log CFU/g for the agricultural products, 2.92 log CFU/g for the livestock products, and 2.82 log CFU/g for the marine products. The isolation rates of foodborne pathogens from the agricultural, livestock, and marine products were 35.42%, 37.21%, and 31.25%, respectively. Forty-five foodborne pathogens of seven species, including Bacillus cereus and Salmonella spp., were isolated from the raw materials of the agricultural, livestock, and marine products in 55 meal kits. To prevent foodborne diseases caused by meal kits, it is necessary to focus on washing, heating, and preventing cross-contamination during cooking.
