In this study the antimicrobial spectrum and sensitivity of a liquid formulation of enrofloxacin against the major six pathogenic bacteria isolated from the diseased poultry in Korea were evaluated in comparison with ciprfloxacin (CFX), gentamicin (GM) and penicillin (PC). A liquid form of enrofloxacin, a test material and ciprofloxacin showed at least 83% antibacterial activity against all of six test pathogenic microorganisms at the fixed concentration of 10 ㎍/㎖. Gentamicin and penicillin revealed much lower antibacterial activity at the same condition. We evaluated the liquid form of enrofloxacin as having widespread antibacterial spectrum against poultry infections. The liquid form of enrofloxacin showed potent antibacterial activity, with the MIC range being 0.12∼2 ㎍/㎖ against Gram positives, 0.06∼2 ㎍/㎖ against Gram negatives and 0.5∼2 ㎍/㎖ against Mycoplasma spp. These values were equivalent to or somewhat higher than those of ciprofloxacin, but much lower than those of gentamicn and penicillin. This new liquid form of enrofloxacin would find its way in application of mixing with drinking water for the prevention and treatment of diverse infections in chickens.

We evaluated the efficacy of the liquid form of enrofloxacin against poultry respiratory and digestive infections, the major problems in the poultry industry, from diverse diagnosis viewpoints. Results obtained through this study were as follows: 1. The liquid form of enrofloxacin showed good acceptability to poultry owing to no abhorrence to the drug by chickens, which indicated its suitability for mixing in drinking water. 2. The application of the enrofloxacin formulation improved the productivity of the chicken farm to a great extent by lowering the death toll of chicken raised in the farm. 3. The use of the drug shortened the days to the market, and increased the body weight at the time of marketing. 4. Preventive and therapeutic effects of the liquid enrofloxacin were observed higher than ampicillin, a major antibiotic used in poultry farming. From above results, the liquid form of enrofloxacin would find its use for the prevention and treatment of digestive and respiratory infections, let alone the growth promotion and feed efficiency, especially in the chicken farming practice.
한국인 소화불량 환자 중 어느환자에서 내시경 검사를 할 것인가?

Background/Aims: Dyspepsia is remarkably common in the general population. Although upper gastrointestinal endoscopy is the investigation of choice for dyspepsia, the selection of the patients who need endoscopy is very difficult. This study was aimed to investigate the usefulness of the various parameters for the selection of the patients who need endoscopy in Korean population, Methods: An analysis of the endoscopic findings was carried out in 141 patients with dyspepsia according to parameters including H. pylori IgG seropositivity, age and alarm signs, such as dysphagia, weight loss, vomiting, anemia, family history of upper gastrointestinal malignancy, hematemesis or melena, abdominal mass and anorexia. Results: The positive rates of stomach cancer and peptic ulcer in patients above 35 years of age were 3% (4/118) and 30% (35/118), respectively. The positive rates of stomach cancer and peptic ulcer in patients with alarm signs were 2% (2/94), 36% (34/94) and the rates in patients with H. pylori IgG seropositivity were 4% (3/70), 37% (26/70), respectively. However, no parameter was useful in the selection of patients with stomach cancer or peptic ulcer. In patients with alarm signs, the positive rates of peptic ulcer in patients with and without H. pylori IgG seropositivity were 46% (22/48) and 26% (12/46), respectively (p=0.04). In patients under 35 years of age, the positive rates of peptic ulcer in patients with and without H. pylori IgG seropositivity were 58% (7/12) and 18% (2/11), respectively (p=0.048). Conclusions: The overall positive rates of stomach cancer or peptic ulcer were significantly higher in patient group with both H. pylori IgG seropositivity and alarm signs, and that under 35 years of age with H. pylori IgG seropositivity. However, there were also considerable number of peptic ulcer in patients who were excluded from those groups. We recommend that those parameters should be used only as a decision aid in selecting the candidates for endoscopy.

DGCR8 is a RNA-binding protein working with DROSHA to produce pre-microRNA in the nucleus, while DICER does not only mature microRNA but also endogenous siRNAs in the cytoplasm. Here, we have produced Dgcr8 conditional knock-out mice using progesterone receptor (PR)-Cre (Dgcr8flox/flox; PRcre/+ mice, Dgcr8d/d) and demonstrated that canonical microRNAs dependent of DROSHA-DGCR8 complex are required for uterine development as well as female fertility in mice. Adult Dgcr8d/d females did not undergo regular reproductive cycle and produce any pups when housed with fertile males, whereas administration of exogenous gonadotropins induced normal ovulation with corpus luteal formation in these mice. Ovulated oocytes from Dgcr8d/d mice had comparable fertilization potentials and were normally developed to the blastocyst after fertilization as compared to those in control Dgcr8f/f mice. Interestingly, PR-Cre-dependent Dgcr8 deletion showed aberrant infiltration of acute inflammatory immune cells to female reproductive organs only when Dgcr8d/d mice were mated with male mice. With respect to uterine development, gross morphology, histology, and weight of Dgcr8d/d uterus were similar to those of control at 3-week-old age. However, multiple uterine abnormalities were noticeable at 4-week-old age when PR expression is significantly increased, and these deformities became severe onwards. Gland formation and myometrial layers were significantly reduced, and stromal cell compartment did not expand and became atrophic during uterine development in these mice. These results were consistent with aberrantly reduced cell proliferation in stromal cell compartments of Dgcr8d/d mice. Collectively, our results suggest that DGCR8 dependent-canonical microRNAs are essential for development and physiology of the uterus with respect to morphogenesis, proper immune modulation, reproductive cycle, and steroid hormone responsiveness in mice.
MicroRNA is required for uterine stromal cell proliferation in mice

DGCR8 is a RNA-binding protein working with DROSHA involved in critical processes for microRNA production in the nucleus. To understand function of miRNAs in the uterus, we have produced uterus-specific Dgcr8 conditional knock-out mice using two well-known Cre mouse models, anti-Mullerian hormone receptor 2 (Amhr2)-Cre and progesterone receptor (PR)-Cre. Dgcr8flox/flox;PRcre/+ mice were mainly analyzed and considered as uDgcr8 KO in this study unless otherwise indicated as Dgcr8flox/flox;Amhr2cre/+ mice. Morphological and histological analyses, embryo cultures, genomic DNA PCR, realtime RT-PCR and Western blotting were performed. uDgcr8 KO females bred with fertile males did not produce any offspring, suggesting that these mice are infertile. Vaginal smear analyses showed that these mice do not undergo estrous cycle, whereas Dgcr8flox/flox;Amhr2cre/+ mice exhibited regular estrous cyclicity. In vitro culture of 2-cell stage embryos and histological analyses for CL in uDgcr8 KO demonstrated that they can respond to gonadotrophins to ovulate healthy oocytes with comparable fertilization potentials as compared to those in Dgcr8flox/flox mice (Control). Gross morphology, histology, and weight of uteri of uDgcr8 KO mice were similar to those of control at 3-week-old stage. However, uterus become extremely thinner and shorter from 4-week-old stage onward. Histological examination showed significant reduction in gland numbers and stromal area from 4-week-old stage. Interestingly, this phenotype is reflected by significant increase of PR expression in the uteri of 4-week-old mice. In addition, stromal cell proliferation of uDgcr8 KO is severely impaired. BrdU incorporation experiments showed that while epithelial cells undergo proliferation by E2 treatment, stromal cells do not incorporate BrdU under the uterine conditions provided with E2+P4. Collectively, these results conclude that microRNAs are essential for uterine stromal cell proliferation in mice.

Backaround : From medical care perspective regarding HIV, early detection of HIV is critical in effectively managing its sequelae. People who are aware of the fact that they are infected with HIV significantly reduce high risk behaviors, thus limiting transmission to others compared to individuals who are unaware of their HIV serostatus. Materials and Methods : HIV rapid test validation study was conducted at Asan medical center using OraQuick Advance Rapid HIV-1/2 Antibody Test which uses oral fluid specimens for the diagnosis of HIV infection. A total of 57 subjects were tested; 53 individuals were confirmed to have HIV infection by the Western blot analysis and 4 individuals were negative for HIV. Results : The oral fluid specimens from 52 of 53 infected individuals showed positive reaction to the OraQuick Advance Rapid HIV-1/2 Antibody Test. The test results were negative for the 4 subjects who were negative for HIV. OraQuick Advance Rapid HIV-1/2 Antibody Test's sensitivity was 98.1%, specificity was 100%, positive predictive value was 100%, and negative predictive value was 80%. Conclusion : The recently approved OraQuick Advance Rapid HIV-1/2 Antibody Test has the ability to detect HIV antibodies comparable to that of confirmatory tests such as Western blot analysis.
