어린 해삼(Apostichopus japonicus)용 사료 개발을 위한 실험사료의 사육효능 평가

Six experimental diets for juvenile sea cucumbers Apostichopus japonicus were evaluated and compared with current commercial diets. Sea cucumbers (50 per tank; initial mean weight $2.3{\pm}0.1g$) in three replicates of seven groups were fed one of six experimental diets (ED1, 20% wild-harvested Sargassum thunbergii powder; ED2, 20% commercial S. thunbergii powder; ED3, 20% Undaria pinnatifida and Ascophyllum nodosum powders; ED4, 40% U. pinnatifida and A. nodosum powders; ED5, 10% brewer's yeast; and ED6, 10% sea shell powder) or a commercial diet (CD) for 28 weeks. The survival rate in all groups was 85-94%, with no significant difference among the experimental groups. The mean body weight of the sea cucumbers was significantly different among the experimental groups 6 weeks after the feeding trial. The final mean weight of the sea cucumbers was highest in ED1, ED3, and ED4, followed by ED6, ED5, ED2, and CD, in decreasing order. The results of this study suggest that U. pinnatifida and A. nodosum powders could provide a substitute for the S. thunbergii powder commonly used in commercial sea cucumber diets. Thus, the EP4 diet formulation, which contains no S. thunbergii, could be used as a practical feed for juvenile sea cucumbers.

Penaeidins은 새우류에 존재하는 항미생물성펩티드의 한 종류로서 새우의 외부 병원체에 대한 방어기작을 구성하는 중요한 인자이다. 본 연구에서는 대하 혈구세포의 cDNA library로부터 분리된 EST 클론을 분리 동정하였다. 아미노산 염기서열 분석과 계통수 분석을 통하여 본 연구에서 분리한 EST 클론이 대하의 Penaeidin 3-2 유전자와 아미노산 수준에서 동일함을 밝혔다. 대하의 Penaeidin 3-2는 signal peptides로 예상되는 19개 아미노산 잔기를 포함하여 전체 71개의 아미노산 잔기로 이루어져 있고, C-말단에는 3개의 이중황화결합을 형성할 수 있는 6개의 cysteine가 존재하였다. 대하 Penaeidin 3-2 전사체는 혈구세포, 간췌장, 근육 조직에서 발현되었으며, 특히 혈구세포에서 주로 많이 발현되었다. 흰반점바이러스의 인위감염 실험에서 바이러스 감염 후 대하 Penaeidin 3-2의 발현이 증가하였다. 이상의 결과들로부터 대하 Penaeidin 3-2가 자체 방어 작용에서 중요한 역할을 할 것으로 예상된다. Penaeidins are members of a special family of antimicrobial peptides existing in several species of shrimp and play a crucial role in the immunological defense of shrimp. In this study, we isolated and characterized one EST clone (penaeidin) from cDNA library of fleshy prawn Fenneropenaeus chinensis hemocytes. Amino acids sequence comparison and phylogenetic analysis with other known penaeidins revealed that our clone was completely identical to F. chinensis Penaeidin 3-2 (Accession no. ABC33920), which composed of 71 amino acids with a putative signal peptide (1-19) and a cysteine-rich domain (C-terminal part). The expression and distribution of Penaeidin 3-2 transcripts in shrimp were detected in hemocytes, hepatopancreas, and muscles, and that Penaeidin 3-2 was constitutively expressed mainly in hemocytes. The artificial infection of white spot syndrome virus to F. chinensis resulted in Penaeidin 3-2 mRNA up-regulation in hemocytes, suggesting that the possible role of Penaeidin 3-2 in host defense system of F. chinensis.

A 3×2 factorial experiment was conducted to investigate effects of water temperature and feeding rate on growth and body composition of olive flounder. Triplicate groups of fish (initial body weight of 118 g) fed a extruded pellet (55% protein and 4708 ㎈/g) to satiation and at restricted feeding rates of 0.25 and 0.4% body weight per day (BW/d) at the different water temperatures (13 and 18℃) for 9 weeks. Weight gain increased significantly with increase in feeding rates at each temperature. Weight gain of fish fed to satiation was significantly higher at 18 than 13℃, whereas, that of fish fed at 0.25 and 0.4% BW/d were significantly or slightly lower at 18 than 13℃. Feed efficiency and protein efficiency ratio of fish fed to satiation were not significantly different between 13 and 18℃, but those of fish fed at 0.25 and 0.4% BW/d were significantly higher at 13 than 18℃. The major finding of this study is that satiation feeding is efficient for optimal growth and feed efficiency of grower oliver flounder (116-164 g) in suboptimal water temperatures. The maintenance feeding ration which is zero growth performance, were 0.30 and 0.41% BW/day at 13 and 18℃, respectively. In the restricted feeding regime, compromised growth of fish were worsen in higher water temperature (18℃ vs. 13℃). It might be related to high metabolic rate of fish that spend more energy for maintenance metabolism. Based on these results, we suggest that a satiation feeding regime is recommended for a productive growth of grower olive flounder in the suboptimal temperature.
양식산 참조기 및 수조기 미성어의 계절별 성장 경향과 저수온 내성 특성

본 연구는 조기류(참조기, 수조기)의 양식생물학적 기초 자료를 확보하고자 육상수조에서의 성장 특성 및 저수온 노출에 대한 영향을 조사하였다. 1년 사육기간동안 수조기 가 참조기에 비해 성장이 빨랐고, CF는 모두 9월에 가장 높게 나타났으며, DFI는 모두 9~10월에 최대값을 보인 후 낮아졌다. 사료계수는 참조기가 2.20, 수조기가 1.34였다. 완 만한 수온 하강에 대한 영향은 참조기와 수조기 미성어 모 두 4.7˚C 이하에서 폐사가 발생하였으며, 조기류 월동의 임 계수온은 9˚C로 나타났다. In the present study, we investigated the characteristic of growth and the tolerance of cold temperature in small yellow croaker and yellow drum, the results shows that the growth of yellow drum were faster than those of small yellow croaker with high food convergency efficiency, In condition factor (CF), the results shows that the values of yellow croaker and yellow drum were significantly higher in September, respectively. In mortality according to cold shock, two fishes was died under 4.7˚C in slow temperature descending test, but was both died under 9˚C in acute temperature descending test.

We evaluated the dietary black soldier fly meal (BSFM) as a fish meal (FM) replacer based on growth, biological indices and composition of whole body, viscera and liver in Korean catfish, Silurus asotus. Three experimental diets contained the protein sources as 40% FM as control (FM), 35% FM + 5% BSFM (FM + BSFM) and 20% FM + 35% dehulled soybean meal (FM + SBM). Each group of 112 fish with an average initial body weight of 85.2 ± 5.0 g (mean ± SD) were randomly distributed into 9 tanks (1,500-L each) in triplicates and fish were fed twice a day with one of the 3 experimental diets for 10 weeks. At the end of feeding trial, weight gain and specific growth rate of fish fed the FM+BSFM diet were significantly higher than that of fish fed the FM+SBM, but not significantly different from that of fish fed the FM diet. The composition of whole body, viscera and liver and biological indices were not significantly different between FM and FM + BSFM. The results of this study suggest that BSFM can be a good animal protein source and can be used substitute for fish meal up to 12.5% in diet.
적수온(21℃)에서 사육한 성장기 넙치 (Paralichthys olivaceus)의 배합사료 적정 공급률

This study was conducted to investigate the effects of feeding rate on growth performance, blood components and histology of growing olive flounder. The experiment was carried out to determine the optimum feeding rate of the fish (initial fish mean weight of 97±3.0 g) at the optimum water temperature. Two replicated groups of fish were fed commercial diet at the feeding rates of 0, 1.0, 1.65 and 2.3% body weight (BW)/day and satiation. Feeding trial was conducted under a flow-through system with ten 1.2 metric ton aquaria receiving filtered seawater at 21±2℃ for three weeks. Weight gain and specific growth rate (SGR) for fish fed to satiation were significantly higher than those for the unfed fish and for fish fed at 1.0 and 1.65% BW/day. There were no significant differences in values of such parameters between fish fed at 1.65% and those fed at 2.3% BW/day and between fish fed at 2.3% and those fed to satiation. Weight gain and SGR for the unfed fish were significantly lower than those for fish in the other treatments. All the tissues (hepatopancreas, kidney and anterior intestine) were in good condition in fish fed the experimental diet at different feeding rates. This result indicates that the optimum feeding rates of olive flounder (97-160 g) was approximately 2.52% BW/day at the optimum water temperature.

This study was conducted to investigate the effects of dietary inclusion of various additives Solid sulfur, fucoidan, and glucan on growth performance and immune responses in Juvenile olive flounder paralichthys olivaceus. Six experimental diets supplemented with : no additives (CON); 0.5 and 1.0% solid sulfur (S); 0.2 and 0.4% fucoidan (F); 0.1% glucan (G) of these additives diet on a dry-matter basis. Fish averaging 8.2±0.17g were fed one of six experimental diets in triplicate groups for 6 weeks. By the end of the feeding trial, weight gain, feed efficiency, specific growth rate, protein efficiency ratio, hepatosomatic index and condition factor of fish fed diet CON were significantly lower then those of fish fed the all additives diets (P < 0.05). In challenge test, fish were infected by intraperitonel injection of 0.1 ml bacterial suspension with Edwardsiella tarda per fish after the feeding trial. As a result fish fed All additive diet showed a lower cumulative mortality then did fish fed CON diet throughout the challenge test. In conclusion, these results indicated that solid sulfur, fucoidan and glucan enhanced the growth, feed efficiency and non-specific immune activity of juvenile flounder and protect the fish against microbial infections.
제주도 양식현장 내 배합사료와 생사료의 급여가 넙치의 성장, 사육수질 및 조직성상에 미치는 영향 비교

Two types of diets, extruded pellet (EP) and raw-fish-based moist pellet (MP) were fed to compare growth, water quality and histology of olive flounder, Paralichthys olivaceus in Jeju fish farm. Two groups of 7000 fish per each tank (initial mean weight 51 g) were fed one of two diets for 7 months. At the end of the feeding trial, significant differences were found in weight gain and feed utilization. Dissolved oxygen (DO) level of the MP tank was decreased from 7.98 to 6.89 after feeding. There were no significant differences in hematocrit, hemoglobin, alanine aminotransferase and aspartate aminotransferase of fish fed the two diets. Whole body protein content of fish fed the EP was significantly higher than that of fish fed the MP. While the feeding rate by month indicated numerically decreased, fish fed the MP had higher feed intake than that of fish fed the EP. Histological examination of hepatopancreas, kidney, and anterior intestine of fish fed the EP did not show any negative effects compared to those of fish fed the MP. This study revealed that EP could substitute for MP in terms of growth performance, blood parameters, water quality and histology of olive flounder.
