A gene that encodes a thermostable protease, coined thermicin, has been isolated from Thermoanaerobacter yonseiensis that is expressed and characterized in E. coli.. In order to elucidate the molecular characteristics on thermostability of the enzyme, molecular modeling and mutagenesis technology were applied. In the modeling structure, the structural core, including the active site, was well conserved; whereas, the two loop regions were unique when compared to thermitase. The mutant enzyme with the small loop deleted (D190-I196), based on modeling structural information, showed identical enzyme activity. However, when the large loop was deleted (P233-P244), a little lower $K_m$ and even a lower kcat was found. This indicates that the large loop could influence catalytic activity. However, the unfolding temperature ($T_m$), which was determined by a differential-scanning calorimetry for the mutant enzyme deleted the small loop, was $96^{\circ}C$. This is $14^{\circ}C$ lower than that for the parent thermicin. These results suggest that the small loop may play a role in maintaining the proper folding of the enzyme at high temperatures, whereas the large loop might be related to catalysis.

This study explains an agricultural mobile commerce case using Oauth Based Real-Time Information Sharing Technique according to the growth of the agricultural e-commerce market. Considering the characteristics of the agricultural market, it can confirm differential values with the existing agricultural mobile commerce market according to the functions and characteristics of its technique. We compare variables for introducing agricultural mobile commerce with the existing mobile commerce. Trust through information sharing with real-timefarming diaries, suitability and perceived ease of use variables are positively influenced, but influence on completeness and authority of information variables are relatively insufficient. Through this, we explained differential values of agricultural mobile commerce and suggested an applicable business model.

The present study investigated the effect of Cortex mori radicis (CMR) and Patrinia saniculaefolia (PS) on the osteogenic differentiation of the human mesenchymal stem cell. CMR and PS have been used as herbal medicine in traditional Korean medicine for a long time. Mesenchymal stem cells that can differentiate into adipocyte, chondrocyte and osteocyte recently issued as a therapeutic agent for degenerative disease. Here, mesenchymal stem cells isolated from synovial fluid of osteoarthritis patient, were cultured in specific media to differentiate into osteogenesis. Osteogenic differentiated mesenchymal stem cells were confirmed using Von Kossa and Arizarion Red S staining. And, the cells were divided into 4 groups: control group, CMR treated group, PS treated group and mixture treated group. To determine the effect of the herbal samples on the osteogenic mesenchymal stem cell, OCT4, SOX2 and NANOG mRNA expressions, known as the key maintenance factors of mesenchymal stem cell was measured using real-time Polymerase Chain Reaction (PCR). As a result, the maintenance factors, OCT4, SOX2 and NANOG, were more increased in cells treated by CMR, PS and the mixture of two herbs than control group. Therefore, we confirmed the enhancing effect of CMR, PS and their mixture on the osteogenic differentiation of mesenchymal stem cells which derived from the synovial fluid of osteoarthritis patient. This study demonstrates that the effect of the herbal samples on the osteogenic differentiation of human mesenchymal stem cell may have a possibility to be a therapeutic agent for the osteoarthritis patients.

Backgrounds: Inflammatory lung disease can arise due to the immoderate expression of pro-inflammatory genes and the disharmony of complex cytokines. Chunkoongkeigi-tang (CKT) has been prescribed to patients with asthmatic symptoms in herbal medicine clinics. However, the effects of CKT on the inflamed human lung cell line A549 have yet to be revealed. Thus we investigate whether CKT can suppress inflammatory response on interleukin 1β (IL-1β)-induced inflamed A549. Methods: In an effort to understand the inhibitory activity of CKT on inflamed A549, we use IL-1β to induce inflammatory gene COX-2 expressions on A549. Inhibition of COX-2 expression and inflammatory cytokine secretion through blocking mitogen activated protein kinase (MAPK) pathway and transcriptional nuclear factor kappa B (NF-κB) activity was confimed by western blot and real-time PCR. Compounds in CKT were identified by liquid chromatography-mass spectrometry (LC-MS). Results: CKT inhibited increased COX-2 in an IL-1β- induced inflammatory state of A549. We confirmed that CKT regulated the mitogen activated protein kinase (MAPK) pathway and transcriptional nuclear factor kappa B (NF-κB). Additionally, a suppressive effect of CKT on the mRNA expression levels of cytokine- related asthma ((IL-5, IL-6, IL-17 and TNF-α) was noted in IL-1β-induced inflamed A549. According to LC-MS analysis, we identified several components in CKT which can be expected to be the active compounds of the anti-inflammatory effect of CKT. Conclusion: We verify the anti-inflammatory effect of CKT via the reduction of the IL-1β-induced increased COX-2 expression through the MAPK and NF-κB pathways, suggesting useful anti-asthmatic potential by inhibiting inflammatory cytokines, especially in patients with asthma.

In 2018, great progress in display performances was achieved in the field of virtual reality (VR), augmented reality (AR), quantum dot light-emitting diode (QLED), and organic light-emitting diode (OLED) displays, in addition to the thin-film transistor (TFT) for displays. In this work, the recent progress of the device performances in each display field was summarized, and the future direction of each technology was proposed.

A gene, coined tay, for a thermostable DNA polymerase from the novel, extremely thermophilic bacterium Thermoanaerobacter yonseiensis was cloned and expressed in E. coli. Using a DNA polymerase homologous PCR product as a hybridization probe, tay was isolated and sequenced to consist of 2616 nucleotides that encode 872 amino acids. A database analysis showed that DNA polymerase, coined Tay, from T. yonseiensis shared a 39% to 47% identity in the amino acid sequence with those from other DNA polymerases. Tay was overexpressed in E. coli as a fusion protein with a poly-histidine tag at the Cterminus. It was purified by heat treatment, followed by a Ni2+-chelate column. The molecular weight of purified Tay was approximately 97 kDa, as shown by SDS PAGE, and it showed high DNA polymerase activity and thermostability. However, it had no 3’→5’ exonuclease activity.
