      • Anti-Proliferative Effect of an Aqueous Extract of <i>Prunella vulgaris</i> in Vascular Smooth Muscle Cells

        <P>The abnormal proliferation of vascular smooth muscle cells (VSMCs) in arterial walls is an important pathogenic factor of vascular disorders such as diabetic atherosclerosis. We have reported the anti-inflammatory effect of an aqueous extract from <I>Prunella vulgaris</I> (APV) in vascular endothelial cell. In the present study, APV exhibited inhibitory effects on high glucose-stimulated VSMC proliferation, migration, and invasion activities, inducing G<SUB>1</SUB> cell cycle arrest with downregulation of cyclins and CDKs and upregulation of the CKIs, p21<SUP>waf1/cip1</SUP> and p27<SUP>kip1</SUP>. Furthermore, APV dose dependently suppressed the high glucose-induced matrix metalloproteinase activity. High glucose-induced phosphorylation of ERK, p38 MAPK, was decreased by the pretreatment of APV. NF-<I><I>κ</I></I>B activation by high glucose was attenuated by APV, as an antioxidant. APV attenuated the high glucose-induced decrease of nuclear factor E2-related factor-2 (Nrf2) translocation and heme oxygenase-1 (HO-1) expression. Intracellular cGMP level was also increased by APV treatment. These results demonstrate that APV may inhibit VSMC proliferation via downregulating ROS/NF-<I><I>κ</I></I>B /ERK/p38 MAPK pathways. In addition, APV has a beneficial effect by the interaction of Nrf2-mediated NO/cGMP with HO-1, suggesting that <I>Prunella vulgaris</I> may be useful in preventing diabetic atherosclerosis.</P>

        Purpose: The consequences of precipitously rising allergic skin inflammation rates worldwide have accelerated the risk of atopic dermatitis (AD). Natural product-based agents with good efficacy and low risk of side effects offer promising prevention and treatment strategies for inflammation-related diseases. We have already reported that Platycodon grandiflorum root-derived saponins (Changkil saponins, CKS) have many pharmacological effects, including anti-inflammatory and anti-allergic effects, but its influence on AD remains unclear. Therefore, we evaluated the inhibitory effect of CKS, mainly platycodin D, on AD-like skin symptoms in mice and the possible mechanisms in cells. Methods: Mice were sensitized and challenged with 2,4-dinitrochlorobenzene (DNCB). Four weeks after challenge, mice were treated with oral administration of CKS for 4 weeks. In addition, cells were used to evaluate the effect of CKS, mainly platycodin D, on the TARC expression regulated mechanism. Results: CKS attenuated DNCB-induced dermatitis severity, serum levels of IgE and TARC, and mRNA expression of TARC, TNF-α, IFN-γ, IL-4, IL-5, and IL-13 in mice. Histopathological examination showed reduced thickness of the epidermis/dermis and dermal infiltration of inflammatory cells and mast cells in the ears. Moreover, CKS and platycodin D inhibited TNF-α/IFN-γ-induced TARC expression through the suppression of NF-κB and STAT1 and induction of Nrf2/ARE-mediated hemeoxygenase-1 (HO-1) expression in cells. Conclusion: We suggest that CKS and platycodin D inhibited the development of AD-like skin symptoms by regulating cytokine mediators and may be an effective alternative therapy for AD-like skin symptoms.ⓒ2014 Elsevler Gmbh.         Sulforaphane protects against acetaminophen-induced hepatotoxicity

        <P><B>Abstract</B></P> <P>Oxidative stress is closely associated with acetaminophen (APAP)-induced toxicity. Heme oxygenase-1 (HO-1), an antioxidant defense enzyme, has been shown to protect against oxidant-induced tissue injury. This study investigated whether sulforaphane (SFN), as a HO-1 inducer, plays a protective role against APAP hepatotoxicity <I>in vitro</I> and <I>in vivo</I>. Pretreatment of primary hepatocyte with SFN induced nuclear factor E2-factor related factor (Nrf2) target gene expression, especially HO-1 mRNA and protein expression, and suppressed APAP-induced glutathione (GSH) depletion and lipid peroxidation, which eventually leads to hepatocyte cell death. A comparable effect was observed in mice treated with APAP. Mice were treated with 300 mg/kg APAP 30 min after SFN (5 mg/kg) administration and were then sacrificed after 6 h. APAP alone caused severe liver injuries as characterized by increased plasma AST and ALT levels, GSH depletion, apoptosis, and 4-hydroxynonenal (4-HNE) formations. This APAP-induced liver damage was significantly attenuated by pretreatment with SFN. Furthermore, while hepatic reactive oxygen species (ROS) levels were increased by APAP exposure, pretreatment with SFN completely blocked ROS formation. These results suggest that SFN plays a protective role against APAP-mediated hepatotoxicity through antioxidant effects mediated by HO-1 induction. SFN has preventive action in oxidative stress-mediated liver injury.</P> <P><B>Highlights</B></P> <P> <UL> <LI> SFN pretreatment increases the cell viability against APAP-induced toxicity. </LI> <LI> SFN pretreatment protects depletion of cellular GSH after APAP treatment. </LI> <LI> SFN pretreatment enhances Nrf2 target gene expression, especially HO-1 after APAP treatment. </LI> <LI> SFN has protective effect against APAP overdose-induced liver injury <I>in vivo</I> model as well. </LI> </UL> </P>

        Ethephon 및 Aminoethoxyvinylglycine 처리가 ‘원황’ 배 모의 수출 환경에서의 품질 및 생리장해 발생에 미치는 영향

        We investigate the effect of preharvest spray of aminoethoxyvinylglycine (AVG), an inhibitor of ACC synthase activity, on fruit quality of ‘Wonhwang’ pear (Pyrus pyrifolia Nakai). And the effects were compared with ethephon which releasing ethylene to elucidate treatment efficacy of ethylene biosynthesis inhibitor on Asian pears. Chemicals were sprayed on 30 days before harvest with AVG (75 and 150 mg/L) and ethephon (100 mg/L), respectively. Highest flesh firmness during 21 days of shelf-life at 25℃ after 30 days of cold storage was attained in the fruit treated with 150 mg/L AVG (26.5N) when compared with untreated control (16.7N), while the fruits treated with 100 mg/L ethephon completely lost their marketability within 14 days of shelf-life by mass drop of firmness down to 11.6N. Low incidence of physiological disorders including pithiness and core browning were attained at the fruits treated with 150 mg/L AVG when we compared with untreated control. No occurrence of mealiness found in the fruits treated with AVG but ethephon treated fruits showed 36.4% during 14 days of shelf-life. Consequently, AVG spray at the concentration of 150mg/L on Asian pear ‘Wonhwang’ fruit regarded as useful preharvest management approach for keeping quality during marketing period after simulated exportation.

        Polyozellus multiplex가 생산하는 지질과산화 저해물질

        담자균류로부터 지질과산화 저해물질을 탐색하던 중 강원도 오대산 지역에 자생하는 버섯의 한 종류로 부터, Sparague-Dawley계 rat의 간 mirosome 분획의 지질과산화를 강하게 억제하는 물질이 있음을 발견하였다. 이 버섯의 미생물학적 특성을 관찰한 결과 민주름버섯목, 굴뚝버섯과에 속하는 까치버섯(Polyzellus multiplex)으로 동정하였으며 본 버섯의 자실체 methanol 추출물을 benzene, ethylacetate로 순차적으로 추출하고 각종 chromatography를 이용하여 활성물질을 정제한 결과, ethylacetate층으로 부터 황산화활성을 나타내는 PM1을 benzene층으로 부터 지방산 계열의 PM2와 PM3 화합물을 순수하게 분리 정제하였다. PM1은 정색반응 및 각종 기기분석 결과 thelephoric acid계의 화합물로 동정되었으며 PM2 및 PM3는 각각 linoleic acid와 oleic acid로 동정되었다. 지질과산화 활성물질로 분리정제한 PM1 화합물의 IC_50 값은 1.96 ppm이었으며 LD_50 값은 500 mg/Kg 이상이었다. In the course of screening lipid peroxidation inhibitor from basidiomycetes, a mushroom, which was collected at O-Dae mountain in Kangweon-Do, was found to have active compound. The mushroom was identified as Polyzellus multiplex, which belongs to Aphylloporalles Thelephoraceae, on the basis of macroscopic and microscopic characteristics of the fruiting body. The methanol extract of fruiting body was extracted with benzene and ethylacetate, sequentially. By using various kinds of chromatographies, PM1, and PM2 and PM3, were purified from the ethylacetate extract and the benzene extract, respectively. Color reaction and analyses of IR, UV, and NMR spectra indicated that PM1 was a derivative of thelephoric acid, and PM2 and PM3 were linoleic acid and oleic acid, respectively. IC_50 of PM1 for inhibition of lipid peroxidation was 1.96 ppm and LD_50 was 500 mg/kg.

        One of the most ridiculous issue of the moving target tracking system is real-time processing of the input scene so as to extract the target features and estirmate the target states but there are no way to solve this problem sensationally in a digital algorithm. Recently an optical approach for this problem is presented; named BJTC tracker. In this paper, we propose a hardware that can the control the JTC signal for the BJTC system and show the timing chart of the hardware system to prove the design method. We use the FPGAs, low-accesstime RAM, and high performance ADC and DAC. And we use NTSC type CCDs and monitors.

        The New Village (Sae-ma-eul) movement is Korean version of Community Development that has under-gone widespread exploration, experimentation and application in developing countries throughout the world in recent years. In most countries the national governments have undertaken the iniative in Community Development through appropriate ministries or special agencies. The New Village Movement was initiated in 1970 by Korean government. But it became a people's movement for self-development as more and more people participated in the developmental programs. The major objectives of the New Village movement are to redress the economic disparities between the rural and urban areas by improving the economic, social and cultural conditions of rural Communities, to reform the way of life in rural villages by encouraging the attitude of self-help, and to increase the income of the farmers and fisher man. Since 1971, An enormous amount of public and priate money has been invested into the New Village Movement. It seems that the time has arrived when some framework for evaluating the whole program is to be developed. Thus, The objective of this study is to develop an appropriate to evaluate the new village project at the village level.         Snakes is an active contour model for representing image contours. To detect boundary on MRI sequential images, we proposed a method based on snakes. Because there is spatial correlation between two adjacent MRI images, an energy term related with spatial correlation was added to Snakes energy formulation. A measure of shape similarity called the BMD was used to evaluate the accuracy of the algorithm. The average BMD value for the modified alogrithm's result is higher than existing Greedy algorithm's. And 3D image was reconstructed by applying triangle interpolation to the detected boundary on each slice.

To solve these problems, small retailers should try to systematize organization by central buying system, point of sale, electronic ordering system and so on. Furthermore, they should cooperate with regional retailers by establishing central institution. Finally, they focus their business on specialization rather than generalization.

        Small and Medium sized Enterprises(SMEs) have not yet explored various business strategies to cope with the epochal turns in business environment characterized by the so-called paradigm shift, that is, the recent trend toward open market and deregulation accompanied by changing of political and economic orders, One of the most remarkable strategies in these movements is the power on securing competitive edge in information infrastructure where a relatively low level of information exists. Since information is valuable resources and contributed to enhance competitive edge for enterpises, most of enterprises are investing a large amount of money to develop information technology. However, SMEs are having difficulty in enhancing information infrastructure due to a shortage of fund, technical experts as well as low level of information mind of a chief executive officer and employees. In order to improve information infrastructure of SMEs, we need a methodology for evaluating current information infrastructure and providing the direction of information infrastructure and providing the direction of information planning based on the evaluation. Especially business environment of Korean SMEs are so much different from other leading countries, therefore we need as evaluation methodology suited to Korean SMEs. Even though there are some evaluation methodologies, they are not applicable to Korean SMEs ; most of methodology are developed by U.S.A or Japan ; no systematic steps are provided; current information techonology are not incorporated; and just conceptual model and empirical studies are conducted. The objective of this study is to develop a methodology for evaluating information infrastructure in Korean SMEs. The methodology is providing systematic evaluation procedures incorporating the business environment of Korean SMEs and current information technology. In order to develop and verify the methodology, several research methodologies such as literature review and case study are conducted. The results show that the evaluation factors are valid and useful in Korean SMEs environment. Future research direction would be focusing on investigation of potential evaluation factors, conducting more surveys to various SMEs, and developing software package for automation of evaluation procedures.
