<P>Five new tirucallane saponins, paramignyosides A-E (1-5), were isolated from the water fraction of the Paramignya scandens stem and leaves. Their structures were elucidated on the basis of spectroscopic evidence including high-resolution electrospray ionization mass spectrometry (HR-ESI-MS) and one dimensional (1D)- and 2D-NMR. The effects of isolated compounds on pro-inflammatory cytokines were evaluated by measuring the production of interleukin (IL)-12 p40, IL-6, and tumor necrosis factor-α (TNF-α) in lipopolysaccharide (LPS)-stimulated bone marrow-derived dendritic cells (BMDCs). Paramignyoside C (3) exhibited selective and potent inhibitory effect (IC50=5.030.19??M) on the production of IL-12 p40 comparable to that of the positive control, SB203580 (IC50=5.000.16??M). Further studies are required to confirm efficacy in vivo and the mechanism of anti-inflammatory effects.</P>

In this study, graphene oxide (GO) was synthesized by the improved Hummers’ method. The degree of oxidation from graphite (Gi) to GO was determined through interlayer spacing calculated from X–ray diffraction. Besides, the effect of KMnO4:Gi ratios (X1), H2SO4 volume (X2), oxidation temperature (X3), oxidation time of stage 1 (X4), and oxidation time of stage 2 (X5) was screened by the Plackett–Burman model. The simultaneous impact of three factors that influenced the degree of oxidation (X1, X2, and X3) was studied by the Box–Behnken experimental model of response surface methodology to achieve suitable conditions for the GO synthesis process. The characterization of GO product was investigated via the modern analytical methods: X-ray diffraction, Raman spectroscopy, Fourier transform infrared spectroscopy, UV–Vis spectroscopy, field emission scanning electron microscopy, transmission electron microscopy, and atomic force microscopy. In addition, the study was also carried out on a pilot scale for orientation in industrial application with the yield of 14 g/batch.

Two new 2-C-β-D-glucopyranosyl benzoic acid derivatives named mallonanosides A (1) and B (2) were isolated from the methanolic extract of the leaves of Mallotus nanus along with five known flavonoids, kaempferin (3), juglanin (4), quercitrin (5), myricitrin (6), and rhoifolin (7). Their structures were established on the basis of spectral and chemical evidence. Their antioxidant activities were shown to depend on the number of hydroxyl groups, and the location and species of sugar moiety.
The Root Causes of Rural Tourism Development Intentions in Vietnam:A Local Government Perspective

Rural tourism has gained popularity in Vietnam, attracting more tourists, particularly foreigners and urban visitors. Despite the attention it receives from the central government, rural tourism development faces challenges, including the perceptions of local civil servants. This study combines the theories of planned behavior and organizational support theory to propose a model predicting the intention of local civil servants to develop rural tourism. A quantitative approach was used, with a questionnaire survey distributed to 405 local civil servants participating in training programs. The findings indicate that factors from both theories positively and significantly influence the intention of rural tourism development. Notably, perceived leader support directly impacts intention and moderates the relationship between perceived government support and intention. Despite its limited practical performance, the study emphasizes the importance of attitude toward rural tourism development. The research provides implications for academics, government, and policymakers to enhance local civil servants' intention for future rural tourism development.

In the eukaryote nucleus, DNA wraps around histone cores and each core complex contains eight proteins including four pairs of H2A, H2B, H3, and H4 protein. The histone tail can be modified to alter the interaction strength between the histone core and a given DNA region, resulting in a change in gene expression. Various types of histone modifications are known to be involved in a number of cellular responses. In this study, we identified and characterized a novel mutant based on a growth assay using histone deacetylase inhibitors in combination with auxin, and we have named the mutant sensitive to histone deacetylase inhibitors 1. Our results imply that histone acetylation plays an important role in shoot morphology and leaf production in Arabidopsis.

<P>In Arabidopsis, the MYB (myeloblastosis) gene family contains more than 190 members, which play a number of roles in plant growth and development. Based on their protein structure, this gene family was divided into several subclasses, including the MYB-related class. Currently, an MYB-related gene designated as MYB-like Domain (AtMYBD) has been shown to function as a positive regulator of anthocyanin biosynthesis in Arabidopsis. This gene was found to belong to the CCA1-like (circadian clock-associated 1) group, which represents several genes that are master regulators of the circadian clocks of plants. Here, we speculate that AtMYBD is able to regulate anthocyanin biosynthesis in Arabidopsis thaliana in a circadian clock-related manner.</P>

<P><B>Abstract</B></P> <P>Transcripts of the Arabidopsis transcription factor gene, <I>AtMYB44</I>, accumulate rapidly to mediate a tolerance mechanism in response to salt stress. The <I>AtMYB44</I> promoter is activated by salt stress, as illustrated in <I>AtMYB44pro::GUS</I> transgenic plants. Chromatin immunoprecipitation (ChIP) assays revealed that RNA polymerases were enriched on the <I>AtMYB44</I> gene, especially on TSS-proximal regions, and nucleosome density was markedly reduced in the <I>AtMYB44</I> gene-body region in response to salt stress. In addition, H2A.Z occupation was significantly decreased at the <I>AtMYB44</I> promoter, transcription start site (TSS), and gene-body regions. Histone modifications including histone H3 lysine 4 trimethylation (H3K4me3) and histone H3 and H4 acetylation (H3ac and H4ac) were not affected under the same stress conditions. We found a decrease in the number of AtMYB44 proteins bound to their own gene promoters in response to salt stress. These results suggest that salt stress induces the eviction of H2A.Z-containing nucleosomes from the <I>AtMYB44</I> promoter region, which may weaken its affinity for binding AtMYB44 protein that acts as a repressor for <I>AtMYB44</I> gene transcription under salt stress-free conditions.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Salt stress reduces the nucleosome density at <I>AtMYB44</I> gene-body regions. </LI> <LI> H2A.Z occupation at <I>AtMYB44</I> promoter is decreased under salt stress conditions. </LI> <LI> Salt stress diminishes the binding of AtMYB44 proteins on their own gene promoters. </LI> </UL> </P>

Peroxisome proliferator-activated receptors(PPARs) are ligand-activated transcription factors thatregulate the expression of multiple genes involved inmetabolic, anti-inflammatory, and developmental processes. This study evaluated the PPARs transactivationaleffects of thirteen cembranoid diterpenoids 1–13 from thesoft coral Lobophytum crassum, using PPAR-responsiveelements–luciferase reporter and GAL4–PPAR chimeraassays. All isolated compounds activated the transcriptionof PPARs in a dose-dependent manner, with EC50 valuesranging from 2.07 ± 1.73 to 130.20 ± 1.85 lM. Moreover,compounds 6–9 affected the transactivation of allthree PPAR types, PPARa, c, b(d), in a dose-dependentmanner, with EC50 values in a ranging from 11.92 ± 1.23to 122.50 ± 2.12 lM. These results provide a scientificrationale for further studies on the soft coral L. crassum andits diterpenoid constituents to develop medicinal productsagainst inflammatory and metabolic diseases.
