      • Inhibition of tau aggregation by a rosamine derivative that blocks tau intermolecular disulfide cross-linking

        <P>Abnormal tau aggregates are presumed to be neurotoxic and are an important therapeutic target for multiple neurodegenerative disorders including Alzheimer’s disease. Growing evidence has shown that tau intermolecular disulfide cross-linking is critical in generating tau oligomers that serve as a building block for higher-order aggregates. Here we report that a small molecule inhibitor prevents tau aggregation by blocking the generation of disulfide cross-linked tau oligomers. Among the compounds tested, a rosamine derivative bearing mild thiol reactivity selectively labeled tau and effectively inhibited oligomerization and fibrillization processes <I>in vitro</I>. Our data suggest that controlling tau oxidation status could be a new therapeutic strategy for prevention of abnormal tau aggregation.</P>

        Nutrient Uptake and Productivity as Affected by Nitrogen and Potassium Application Levels in Maize/Sweet Potato Intercropping System

        Field experiment was conducted during 1993-94 season to determine the pattern of nutrient uptake and productivity of maize/sweet potato intercropping system. Four levels of nitrogen (0, 50, 100 and 150kg N ${ha}_{-1}$) and four levels of potassium (0, 40, 80 and 120kg $K_2$O ${ha}_{-1}$) formed treatment variables. Plants were sampled periodically to determine dry matter and tissue concentrations of N and K in the individual plant components of intercropped maize and sweet potato. Nitrogen and potassium fertilizer did not interact significantly to nutrient uptake by any plant parts of intercropped maize and sweet potato. But application of N fertilizer independently enhanced N uptake in all the plant parts of maize and sweet potato. The uptake of N in leaf, leaf sheath, stem, husk, and cob of maize increased upto 90 days after planting (DAP) but grain continued to accumulate N till its maturity. Sweet potato exhibited a wide variation in N uptake pattern. Sweet potato leaf shared the maximum uptake of N at 50 DAP which rapidly increased at 70 DAP and then declined. Declination of N uptake by petiole and stem were observed after 120 DAP whereas N uptake by tuber increased slowly upto 90 DAP and then rapidly till harvest. Rate of applied K had very little effect on the uptake patterns in different components of intercropped maize. Pattern of K uptake by leaf, petiole and stem of sweet potato showed almost similar trend to N uptake. But uptake of K by tuber increased almost linearly with the K application. Pattern of N and K uptake by grain and tuber paralleled the grain yield of maize and sweet potato respectively. Intercropped productivity of maize and sweet potato found to be better by the application of 100kg N and 120 kg $K_2$O ${ha}_{-1}$

      • Stigmasterol activates Cdc42-Arp2 and Erk1/2-Creb pathways to enrich glutamatergic synapses in cultures of brain neurons

        <P>The naturally occurring phytosterol stigmasterol (ST) ameliorates scopolamine-induced memory impairment and promotes neuritogenesis and synaptogenesis. Therefore, in this study, we investigated the hypothesis that ST promotes spinogenesis and the formation of glutamatergic synapses and thus improves memory in cultured rat hippocampal neurons. Immunoblots showed that ST activated extracellular signal-regulated kinase 1/2 (Erk1/2) and cAMP response element binding protein (Creb), and upregulated actin-related protein 2 (Arp2) and cell division cycle 42 (Cdc42), central components in actin organization and spine head development. Consistently, DiO staining of live neurons showed that ST increased the densities of dendritic filopodia and mushroom-type mature spines, and immunocytochemistry showed an increase in the expressions of the GluN2A and GluN2B subunits of N-methyl-D-aspartate receptors (NMDARs) at synapses. Finally, staining the recycling membranes with FM1-43 showed that ST increased the sizes of synaptic vesicle pools at presynaptic terminals. Together, our data indicate that ST upregulates Cdc42-Arp2/3 and Erk1/2-Creb signaling and thus induces the formations of mushroom-type spines and glutamatergic excitatory synapses.       • The experimental study of the effect of microwave on the physical properties of multi-walled carbon nanotubes

        This paper reports the effect of microwave on the physical properties of multi-walled carbon nanotubes (MWCNTs) where different power levels of microwave were applied on MWCNTs in order to apprehend the effect of microwave on MWCNTs distinctly. A low energy ball milling in aqueous circumstance was also applied on both MWCNTs and microwave treated MWCNTs. Temperature profile, morphological analysis by field emission scanning electron microscopy (FESEM), defect analysis by Raman spectroscopy, thermal conductivity, thermal diffusivity as well as heat transfer coefficient enhancement ratio were studied which expose some strong witnesses of the effect of microwave on the both purification and dispersion properties of MWCNTs in base fluid distilled water. The highest thermal conductivity enhancement (6.06% at 40<SUP>o</SUP>C) of MWCNTs based nanofluid is achieved by five minutes microwave treatment as well as wet grinding at 500rpm for two hours.

        Culture-independent analysis of yeast diversity in Korean traditional fermented soybean foods (doenjang and kanjang) based on 26S rRNA sequence

        The yeast-26S rRNA libraries were constructed from two different fermented soybean foods, doenjang and kanjang. A total of 42 clones, containing the partial 26S rRNA sequences, 0.6 kb in length, were sequenced and subjected to an online similarity search. All doenjang yeast (DY) clones only appeared in the Saccharomycotina class. The 21 clones from the doenjang library were classified into five groups: Debaryomyces hansenii (DY I, 76.0 %), Zygosaccharomyces pseudorouxii (DY II, 9.6 %), Candida versatilis (DY III, 4.8 %), Candida etchellsii (DY IV, 4.8 %), and Debaryomyces castellii (DY V, 4.8 %). The 21 kanjang yeast (KY) clones were affiliated with the Saccharomycotina (52.4 %), Urediniomycetes (19.0 %), Ustilaginomycetes (23.8 %), and Hymenomycetes (4.8 %) classes and divided into six groups: D. hansenii (KY I, 38.0 %), Sterigmatomyces halophilus (KY II, 19.0 %), Malassezia restricta (KY III, 23.8 %), Cryptococcus magnus (KY V, 4.8 %), and Pichia triangularis (KY VI, 9.6 %). Yeast belonging to the Saccharomycotina class was predominant (76.2 %) in fermented soybean foods, doenjang and kanjang. These findings are of fundamental value for understanding the complexity of two different fermented soybean foods.

      • Crosstalk between Oxidative Stress and Tauopathy

        <P>Tauopathy is a collective term for neurodegenerative diseases associated with pathological modifications of tau protein. Tau modifications are mediated by many factors. Recently, reactive oxygen species (ROS) have attracted attention due to their upstream and downstream effects on tauopathy. In physiological conditions, healthy cells generate a moderate level of ROS for self-defense against foreign invaders. Imbalances between ROS and the anti-oxidation pathway cause an accumulation of excessive ROS. There is clear evidence that ROS directly promotes tau modifications in tauopathy. ROS is also highly upregulated in the patients’ brain of tauopathies, and anti-oxidants are currently prescribed as potential therapeutic agents for tauopathy. Thus, there is a clear connection between oxidative stress (OS) and tauopathies that needs to be studied in more detail. In this review, we will describe the chemical nature of ROS and their roles in tauopathy.</P>

        Homeopathy - A Safe, Much Less Expensive, Non-Invasive, Viable Alternative for the Treatment of Patients Suffering from Loss of Lumbar Lordosis

        Objectives: Loss of lumbar lordosis causing pain and curvature of the vertebral skeleton to one side is a relatively uncommon disease. To our knowledge, successful treatment of loss of lumbar lordosis with any potentized homeopathic drug diluted above Avogadro's limit (that is, above a potency of 12C) has not been documented so far. In this communication, we intend to document a relatively rare case of loss of lumbar lordosis with osteophytic lippings, disc desiccation, and protrusion, causing a narrowing of secondary spinal canal and a bilateral neural foramina, leading to vertebral column curvature with acute pain in an adolescent boy. Methods: The patient had undergone treatment with orthodox Western medicines, but did not get any relief from, or cure of, the ailment; finally, surgery was recommended. The patient's family brought the patient to the Khuda-Bukhsh Homeopathic Benevolent Foundation where a charitable clinic is run every Friday with the active participation of four qualified homeopathic doctors. A holistic method of homeopathic treatment was adopted by taking into consideration all symptoms and selecting the proper remedy by consulting the homeopathic repertory, mainly of Kent. Results: The symptoms were effectively treated with different potencies of a single homeopathic drug, Calcarea phos. X-ray and magnetic resonance imaging (MRI) supported recovery and a change in the skeletal curvature that was accompanied by removal of pain and other acute symptoms of the ailment. Conclusion: Homeopathy can be a safe, much less expensive, non-invasive, and viable alternative for the treatment of such cases.

      • Syntheses of Metal–Organic Frameworks and Aluminophosphates under Microwave Heating: Quantitative Analysis of Accelerations

        <P>Several porous materials such as metal–organic frameworks (MOFs) and aluminophosphates have been synthesized with microwave and conventional electric heating in various temperatures and times to investigate the quantitative acceleration in the synthesis of porous materials by microwaves. From the analysis of the acceleration under microwave heating with the Eyring equation, it can be understood that, irrespective of the type of porous materials, the acceleration by microwaves is mainly due to decreased activation free energy (Δ<I>G</I><SUP>⧧</SUP>) even though the activation energy (<I>E</I><SUB>a</SUB>) and activation enthalpy (Δ<I>H</I><SUP>⧧</SUP>) are increased. The decreased activation free energy is mainly due to the high activation entropy (Δ<I>S</I><SUP>⧧</SUP>) of microwave synthesis compared with the entropy of conventional electric synthesis.       • Global warming as affected by incorporation of variably aged biomass of hairy vetch for rice cultivation

        <P> Hairy vetch (Vicia villosa Roth) is cultivated during the cold fallow season in paddy soils of temperate countries such as South Korea and Japan, mostly as animal feed and green manure. Information on the effect of ageing of hairy vetch incorporation in relation to greenhouse gas (GHG) emissions and global warming potential (GWP) is not available. Therefore, hairy vetch biomass of ages 183, 190, 197, and 204 days was incorporated in paddy soil to estimate GWP during rice cultivation. The emission rates of methane (CH4), carbon dioxide (CO2), and nitrous oxide (N2O) gases were monitored once a week by using the closed-chamber method. The net ecosystem carbon budget was used to estimate pure CO2 emission fluxes. Biomass production of hairy vetch was 6.5 Mg ha-1 at 204 days, which was similar to other treatments. The GWP was lower with the 204-day-old vetch biomass incorporation than with other treatments. High content of cellulose and lignin in 204-day-old hairy vetch might have affected decomposition rate and subsequently reduced GHGs emissions during rice cultivation. Our results suggest that hairy vetch can be allowed to grow for 204 days before incorporation at 3 Mg ha-1 without sacrificing rice yield, while maximising biomass production and minimising GWP during rice cultivation. </P>
