Radiation treatment to turfgrass cultivar to improve drought tolerance cultivar breeding

Gamma irradiation is used in plant mutation breeding. This method was also applied to turfgrass breeding. Hallagreen 07, 09, and 10 are irradiated and confirmed by image analysis that objective basis on canopy cover. The canopy cover means it has drought tolerance than relatively smaller canopy cover turfgrass. Hallagreen 07 represents a low canopy cover, Hal- lagreen 09 is a medium, and Hallagreen 10 is excellent at the untreated control. However, after the radiation treatment of 30 entries per cultivar, the trait improved than the control appeared at Hallagreen 07, which showed low canopy cover. In contrast, Hallagreen 10, the excellent cultivar, has no entries at improving the trait by irradiation. So, by irradiation, we could find better drought tolerance entries than control. In this study, we found the potential of radiation treatment to turfgrass to improve drought tolerance, and the efficiency of finding trait improvement is effective at not an excellent cultivar. And despite the canopy cover analysis by the vertical image being low tech, it is an efficient solution for screening bottlenecks.
Response of storage insect species to ClO<sub>2</sub> fumigation conditions

<P><B>Abstract</B></P> <P>Insects damage grain during post-harvest storage. Grain loss during storage is a serious problem, especially in less-developed countries. In this study, a gaseous ClO<SUB>2</SUB> treatment was applied to four different insect species known to damage stored grains: <I>Sitophilus zeamais</I> (Motschulsky), <I>Trogoderma granarium</I> (Everts), <I>Callosobruchus chinensis</I> (Linnaeus), and <I>Tribolium castaneum</I> (Herbst). Different insect species showed different sensitivities to the same ClO<SUB>2</SUB> fumigation conditions. The 100 ppm treatment level with 24 h exposure is effective to controlling <I>S. zeamais</I>, <I>T. granarium</I>, and <I>C. chinensis.</I> However, to control <I>T. castaneum,</I> a higher concentration of the ClO<SUB>2</SUB> gas or increased exposure time is needed. <P><B>Abstract</B></P> <P> <I>Plodia interpunctella</I> and <I>Sitophilus zeamais</I> are among the most destructive insect pests of stored processed food products. The use of natural materials rather than conventional pesticides to control these pests is a promising alternative to conventional neurotoxic insecticides. In this study, we tested organic extracts from 13 species of plants; most showed different effects on <I>S. zeamais</I> and <I>P. interpunctella</I>. Eleven of the extracts had repellent effects on <I>S. zeamais</I> but either an attractant effect or no effect on <I>P. interpunctella</I>. <P>Polyunsaturated fatty acids usually undergo lipid peroxidation induced by reactive oxygen species (ROS). Calcium-independent cellular phospholipase A(2) (iPLA(2)) can maintain fatty acid compositions in phospholipids depending on physiological conditions. An insect iPLA(2) (Pi-iPLA(2)) was predicted from the transciptome of the Indianmeal moth, Plodia interpunctella. It encodes 835 amino acids. It possesses five ankyrin repeats in the N terminal and patatin lipase domain in the C terminal. Pi-iPLA(2) was expressed in all developmental stages of the Indianmeal moth. In the larval stage, it was expressed in all tissues tested. RNA interference (RNAi) specific to Pi-iPLA(2) was performed using specific double-stranded RNA (dsRNA). It resulted in almost 70% of reduction in gene expression. Under such RNAi condition, P. interpunctella exhibited significant accumulation of lipid per oxidation based on the amount of malondialdehyde. RNAi of Pi-PLA(2) expression also impaired cellular immune response of P. interpunctella. Chlorine dioxide (ClO2), an insecticidal agent by generating ROS, increased lipid peroxidation in a dose-dependent manner. However, the addition of vitamin E (an antioxidant) reduced the formation of lipid peroxidation. ClO2 treatment significantly reduced expression of Pi-iPLA(2) but increased lipid peroxidation in larval fat body of P. interpunctella. Furthermore, larvae treated with dsRNA specific to Pi-iPLA(2) were significantly susceptible to ClO2 treatment. These results suggest that Pi-iPLA(2) plays a crucial role in repairing damaged fatty acids from phospholipids. Our results also suggest that ClO2 can elevate lipid peroxidation through inhibiting Pi-iPLA(2) expression in addition to direct ROS production.</P>

<P><I>Celtis choseniana</I> is the traditional plant used at Korea as a herbal medicine to ameliorate inflammatory responses. Although<I> Celtis choseniana</I> has been traditionally used as a herbal medicine at Korea, no systemic research has been conducted on its anti-inflammatory activity. Therefore, the present study explored an anti-inflammatory effect and its underlying molecular mechanism using<I> Celtis choseniana </I>methanol extract (Cc-ME) in macrophage-mediated inflammatory responses.<I> In vitro</I> anti-inflammatory activity of Cc-ME was evaluated using RAW264.7 cells and peritoneal macrophages stimulated by lipopolysaccharide (LPS), pam3CSK4 (Pam3), or poly(I:C).<I> In vivo</I> anti-inflammatory activity of Cc-ME was investigated using acute inflammatory disease mouse models, such as LPS-induced peritonitis and HCl/EtOH-induced gastritis. The molecular mechanism of Cc-ME-mediated anti-inflammatory activity was examined by Western blot analysis and immunoprecipitation using whole cell and nuclear fraction prepared from the LPS-stimulated RAW264.7 cells and HEK293 cells. Cc-ME inhibited NO production and mRNA expression of inducible nitric oxide synthase (iNOS), cyclooxygenase (COX-2), and tumor necrosis factor-alpha (TNF-<I>α</I>) in the RAW264.7 cells and peritoneal macrophages induced by LPS, pam3, or poly(I:C) without cytotoxicity. High-performance liquid chromatography (HPLC) analysis showed that Cc-ME contained anti-inflammatory flavonoids quercetin, luteolin, and kaempferol. Among those, the content of luteolin, which showed an inhibitory effect on NO production, was highest. Cc-ME suppressed the NF-<I>κ</I>B signaling pathway by targeting Src and interrupting molecular interactions between Src and p85, its downstream kinase. Moreover, Cc-ME ameliorated the morphological finding of peritonitis and gastritis in the mouse disease models. Therefore, these results suggest that Cc-ME exerted<I> in vitro</I> and<I> in vivo</I> anti-inflammatory activity in LPS-stimulated macrophages and mouse models of acute inflammatory diseases. This anti-inflammatory activity of Cc-ME was dominantly mediated by targeting Src in NF-<I>κ</I>B signaling pathway during macrophage-mediated inflammatory responses.</P>
Sliding Wear Behaviors of Steam Generator Tubes in Various Environments

<P>Wear damage of steam generator tubes for nuclear power plants can cause the leakage of radioactive substances. Therefore, the evaluation of integrity and safety for tubes is very important from the viewpoint of nuclear ecocide. In the present study, to investigate the wear properties of Inconel 600 and 690 steam generator tube materials mated with 409 stainless steel commonly used as support plate, sliding wear tests were performed with increasing sliding distance in air and in elevated temperature water environment, respectively. The wear volume of tube materials was less than those of supports under all conditions. There were no significant differences in the wear behavior for the Inconel 600 and 690 tubes, independently of the testing environment.</P>

<P>Wear damage of steam generator tubes of nuclear power plants can cause leakage of radioactive substances. So the evaluation of tubes’ integrity is very important from the viewpoint of nuclear ecocide. In the present study, sliding wear behaviors of Inconel 600 and 690 steam generator tube materials mated with 409 stainless steel commonly used as the support plate were investigated at room temperature in an air environment. For more precise prediction of wear behaviors of steam generator tubes, Archard equation was modified, and the modified wear coefficients were estimated as a function of sliding distance. When using the modified Archard equation, the reliabilities for prediction of wear behavior of Inconel 600 and 690 mated with 409 stainless steel increased from 71.8% to 83.8% and from 60.2% to 85.2%, respectively.</P>
