        Crystallization and preliminary crystallographic analysis of IlvC, a ketol-acid reductoisomerase, from Streptococcus pneumoniae

        IlvC, a ketol-acid reductoisomerase, plays a critical role in alkyl migration and catalyzes the second step in the biosynthesis of branched amino acids such as leucine, valine and isoleucine. As an initial step to investigate whether IlvC is involved in pneumococcal growth and virulence from the structural background, ilvC from Streptococcus pneumoniae D39 (SpIlvC) was cloned and overexpressed in Escherichia coli. Crystals of SpIlvC were obtained by hanging-drop vapour diffusion in 0.1 M HEPES pH 7.5, 0.1 M NaCl, 1.5 M ammonium sulfate and diffracted to 1.69 Å resolution. The SpIlvC crystal belonged to space group P2 1 2 1 2 1 with unit cell parameters a = 69.1°, b = 104.3°, c = 110.9° and contained two molecules in the asymmetric unit.

        A bellows can be used as an efficient energy absorbing clement of an automotive steering column. On the other hand, bending flexibility of the bellows has negative effects on the vibration characteristics. An effort is made to improve vibration characteristics of steering columns with bellows. To understand the effects of various design parameters on the collapse and vibration, sensitivity analyses are performed by finite element method using Taguchi's scheme. It is shown that the structure of upper mounting bracket is the most important parameter affecting the vibration characteristics. An optimal design is proposed for a lower tilt type steering columns.

      • Durability Improvement of Synthetic Dry Adhesives by Metal Coatings

        <P>Gecko-like synthetic dry adhesives (SDAs) have adhesion comparable to that of a real Gecko’s foot, but with very low durability. To address this problem, self-cleaning or stiff core embedding methods have been proposed. However, the proposed methods require special locomotion or complicated manufacturing. In this study, we suggested a metal coating on synthetic dry adhesives to improve durability. SDAs were fabricated via PDMS. Then, metals such as indium, zinc, and gold were coated on the SDAs. The adhesion tests show that the indium- and zinc-coated microstructures have a higher shear adhesion strength than the noncoated ones. Also, the shear adhesion strength of noncoated SDAs was only 14.5% of the initial strength while that of the zinc-coated ones was 35.6% after 200 times of attachment and detachment. We could find PDMS debris and fractures on noncoated SDAs, which results in weakening of the adhesion strength. On the other hand, a relatively high hardness, strength, and stiffness of the zinc coating layers reduced the wear and fractures of the micropatterns, which led to an improved durability in the SDAs. From these tests, we can conclude that the metal coating method could improve the durability of the SDAs.</P>

        Technological Characteristics by Industry and Innovation Strategy

        This aim of the study is to show the necessity of implementing an industrial innovation strategy with consideration given to characteristics of the industrial technology. The relationship between industrial technological characteristics and innovation performance is analyzed by using an innovation survey as well as a human capital corporate panel (HCCP). The time-lag effect is also examined. Findings of the analysis show that high-tech industries have entered the post catch-up technology innovation stage in the mid-2000s, but low-tech industries still seem to stay in the catch-up stage. In terms of technology policy, the additional technology innovation support policy should focus on enhancing the innovation capability of the middle and low technology industries, since high technology industries are already developing their own innovation capability. It is necessary to strengthen capacity building through technical cooperation with technology consulting, rather than providing technical support through suppliers.

      • 숙련변화와 교육훈련 수요

        본 연구는 직업의 변화, 숙련의 변화를 분석하여 교육훈련 수요에 대한 시사를 얻고자 한다. 미국 직업정보(O*Nnet) 2010년 854개 직업정보와 2019년 968개 직업정보의 숙련수준을 분석하였다. 숙련의 변화는 기존직업 내에서의 숙련변화보다 새로운 직업을 통해 발생하는 것으로 여겨진다. 사회 전반적으로는 새롭게 등장하는 직업을 중심으로 분석 스킬, 기초 스킬, 관리 스킬, 문제해결 스킬, 학습 스킬, 사회적 스킬의 순으로 고려되는 것이 바람직하다고 여겨진다. 정보분석직, 기술직 에서 새롭게 등장하는 직업의 숙련수준이 동일 직종내 기존 직업보다 기능직 지향으로 해석된다. 기능직에서는 신규직업의 숙련수준이 전반적으로 기존 직업의 숙련수준보다 높아지고 있으나, 이러한 신규직업의 등장이 많지는 않다. 생산직에서는 신규 직업의 등장에 따른 숙련수준 변화가 기능스킬 보다도 사회스킬, 기초스킬 등이 강조될 필요가 있는 것으로 해석된다. This study aims to analyze the change of occupation and skill and to get an implication of the demand for education and training. Based on US O*Net(Occupational Information), skills level of 854 job in 2010 and 968 job in 2019 were compared. Changes in skills were appeared through new jobs rather than changes in existing jobs. Analytical skills in emerging jobs across society. Overall focusing on newly emerging jobs. it is desirable to consider in the order of analysis skills, basic skills, management skills, problem solving skills, learning skills, and social skills. Change of Skills in information analyst and engineers are interpreted to be more functional skill-oriented than existing similar occupations. In technician level, the level of skill of new occupations is generally higher than that of existing occupations, but the appearance of such new occupations is not much. In manual worker level, changes in skills are interpreted as emphasis on social skills and basic skills rather than functional skills.

        Bacillus subtilis에 의해 발효된 다양한 콩의 품질 특성

        Purpose: For dissecting the possible benefits and usages of Cheonggukjang, five different beans, soybean, black soybean, red bean, chickpea, and lentil bean, underwent the fermentation process along with Bacillus subtilis. Methods: Resultant Cheonggukjang physicochemical and sensory properties such as pH, amino type nitrogen, slime contents, rheological properties, and sensory strength and consumer acceptance were analyzed. Results: Values of pH were found to be highest in Cheonggukjang made with soybean (SC), followed by Cheonggukjang made with black bean (BSC) and Cheonggukjang made with red bean and soybean mixture (RBSC), and lastly Cheonggukjang made with chickpea (CBC) and lentil bean (LBC). Amino type nitrogen values were shown to be highest in SC with a value of $394.74{\pm}28.80mg%$, subsequently were values taken from RBSC, followed by BSC, CBC, and lastly LBC. Slime contents were the highest in SC with a value of $3.44{\pm}0.36%$, with RBSC, BSC, CBC, and LBC without statistical difference. Total colony count was found to show higher value in BSC, SC, and RBSC, in contrast to the lower values found in CBC and LBC. Color was shown to be lightest in CBC and with darker shades shown that of SC, LBC, RBSC, and BSC in descending order. In rheological properties, hardness, adhesiveness, and viscosity were found to be highest in SC. Sensory strength analysis and sticky appearance were highest in SC, with fermented odor being lowest in CBC and BSC. The strength of sweetness, sourness, and umami taste did not show statistical difference, however, bitterness was shown to be highest in LBC and lowest in CBC, SC, and BSC. In the consumer acceptance test, overall acceptance was highest in SC and CBC. Conclusion: In conclusion, SC was found to be the best bean variety for making Cheonggukjang fermented with Bacillus subtilis, however, CBC may act as an alternative for manufacture of Cheonggukjang resulting in sound consumer acceptance.

        The optimal standard protocols for whole-genome sequencing of antibiotic-resistant pathogenic bacteria using third-generation sequencing platforms

        Background The emergence of antibiotic-resistant bacterial pathogens in the environment has been increasing, posing a threat to public health. Next-generation sequencing technology, which is both low cost and large scale, was used to identify antibiotic-resistance genes or toxin genes in these pathogens. Short-read sequencing cannot fully reconstruct bacterial chromosomes and plasmids carrying toxin genes and antibiotic-resistance genes because of their location on the insertion sequences or repeat regions. Third-generation sequencing generated long reads that could cover the repetitive and/or insertion sequences, allowing for complete chromosome and plasmid reconstruction. However, the optimal protocols for whole-genome sequencing of antibiotic-resistance pathogenic bacteria, from DNA extraction to genome assembly, are still being researched. Objective To develop a pipeline of optimal methods for whole-genome sequencing of bacteria, we compared three commercial DNA extraction kits (column extraction, magnetic bead extraction, and precipitation extraction), two third-generation sequencing platforms (MinION and PacBio), and three assembly methods (flye, unicycler, and unicycler_hybrid). The assembly results were evaluated based on the number of contigs and the detection of anti-microbial-resistance genes. Results Magnetic bead extraction method generated longer N50 read lengths and greater read length distribution than the other two extraction methods. The Flye assembler in combination with magnetic bead extraction and MinION sequencing provided consistent successful plasmid assembly results and detected all antimicrobial-resistance gene in all datasets. Conclusion DNA extraction, sequencing platform, and assembly methods can all have an impact on the results of bacterial whole-genome sequencing. Our findings could be a practical protocol for researchers who use third-generation sequencing to perform bacterial whole-genome sequencing by consistently resolving small plasmids carrying antibiotic-resistance genes.
