      • Pathway and Network Analysis in Glioma with the Partial Least Squares Method

        Gene expression profiling facilitates the understanding of biological characteristics of gliomas. Previous studies mainly used regression/variance analysis without considering various background biological and environmental factors. The aim of this study was to investigate gene expression differences between grade III and IV gliomas through partial least squares (PLS) based analysis. The expression data set was from the Gene Expression Omnibus database. PLS based analysis was performed with the R statistical software. A total of 1,378 differentially expressed genes were identified. Survival analysis identified four pathways, including Prion diseases, colorectal cancer, CAMs, and PI3K-Akt signaling, which may be related with the prognosis of the patients. Network analysis identified two hub genes, ELAVL1 and FN1, which have been reported to be related with glioma previously. Our results provide new understanding of glioma pathogenesis and prognosis with the hope to offer theoretical support for future therapeutic studies.

      • ZD1839 and Cisplatin Alone or in Combination for Treatment of a Nasopharyngeal Carcinoma Cell Line and Xenografts

        This study evaluated the effects of ZD1839, an orally active, selective epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor, on nasopharyngeal carcinoma (NPC) both in vitro and in vivo. Influence of ZD1839 alone or combined with cisplatin on the NPC cell line CNE2 was detected by MTT assay with flow cytometry assessment of cell cycle distribution and apoptosis rates. Nude mice NPC xenografts were also used to evaluate the effects of ZD1839 alone or combined with cisplatin. The Student's t test evaluated statistical significance. ZD1839 alone or combined with cisplatin inhibited CNE2 cell line proliferation. ZD1839 induced CNE2 cell cycle arrest in the G1 phase, and higher concentrations induced apoptosis. Xenograft tumors were significantly smaller when treated with 200 mg/kg ZD1839, cisplatin, or cisplatin combined with 100 mg/kg ZD1839 than untreated controls. ZD1839 (200 mg/kg) alone showed good tumor inhibition effects, reduction of tumor weights, and smaller tumor volume without loss of body weight. ZD1839 (200 mg/kg) might provide a good and effective therapeutic reagent for NPC.

        Terahertz Computed Tomorgraphy with a Quantum Cascade Laser and a Quantum-well Photodetector

        In this letter, a terahertz (THz) tomography system based on a quantum cascade laser (QCL)and a quantum-well photodetector (QWP) is demonstrated. The QCL provides a relatively high power (a few microwatts) in a continuous-wave mode at 3.9 THz as a light source, and the QWP provides a narrow response range from 3 to 7 THz with a calibrated peak response of 0.5 A/W under background-limited performance. We employ the widely-used filtered back projection (FBP)and algebraic reconstruction algorithms for terahertz tomography technology. The reconstructed cross-sectional image of a sample has successfully revealed the internal and the external structures with a resolution of around 1.5 mm. This experiment demonstrates that the terahertz QCL and the photodetector are potentially useful for terahertz tomography applications.

        Genome-wide identification and characterization of the Dof gene family in moso bamboo (Phyllostachys heterocycla var. pubescens)

        The Dof (DNA binding with One Finger) family of single zinc finger proteins is a family of plant-specific transcription factors. These transcription factors have a variety of important functions in different biological processes in plants. In the current study, we identified 26 Dof family genes in moso bamboo (Phyllostachys heterocycla var. pubescens). A complete overview of PhDof genes in moso bamboo is presented, including the gene structures, phylogeny, protein motifs and expression patterns. Phylogenetic analysis of the 26 PhDof proteins identified four classes constituting seven clusters (A, B1, C1, C2, D1, D2 and D3). In addition, a comparative analysis between the Dof genes in moso bamboo, Arabidopsis (Arabidopsis thaliana L.) and rice (Oryza sativa L.) was also performed, and several putative paralogous and orthologous genes were identified. The exon numbers in Dof genes ranged from one to three in many plants; however, the exon number in PhDofs ranged from one to four. The PhDof genes displayed differential expression in different parts of the shoot and at different flower development stages. This study represents the first step towards a genome-wide analysis of the Dof genes in moso bamboo. Our study provides a useful reference for cloning and functional analysis of members of the Dof gene family in moso bamboo and other species.

      • Experimental assessment on flexural behavior of demountable steel-UHPC composite slabs with a novel NPR steel plate

        This study experimentally investigates the flexural behavior of steel-UHPC composite slabs composed of an innovative negative Poisson’s ratio (NPR) steel plate and Ultra High Performance Concrete (UHPC) slab connected via demountable high-strength bolt shear connectors. Eight demountable composite slab specimens were fabricated and tested under traditional four-point bending method. The effects of loading histories (positive and negative bending moment), types of steel plate (NPR steel plate and Q355 steel plate) and spacings of high-strength bolts (150 mm, 200 mm and 250 mm) on the flexural behavior of demountable composite slab, including failure mode, load-deflection curve, interface relative slip, crack width and sectional strain distribution, were evaluated. The results revealed that under positive bending moment, the failure mode of composite slabs employing NPR steel plate was distinct from that with Q355 steel plate, which exhibited that part of highstrength bolts was cut off, part of pre-embedded padded extension nuts was pulled out, and UHPC collapsed due to instantaneous instability and etc. Besides, under the same spacing of high-strength bolts, NPR steel plate availably delayed and restrained the relative slip between steel plate and UHPC plate, thus significantly enhanced the cooperative deformation capacity, flexural stiffness and load capacity for composite slabs further. While under negative bending moment, NPR steel plate effectively improved the flexural capacity and deformation characteristics of composite slabs, but it has no obvious effect on the initial flexural stiffness of composite slabs. Meanwhile, the excellent crack-width control ability for UHPC endowed composite members with better durability. Furthermore, according to the sectional strain distribution analysis, due to the negative Poisson's ratio effect and high yield strength of NPR steel plate, the tensile strain between NPR steel plate and UHPC layer held strain compatibility during the whole loading process, and the magnitude of upward movement for sectional plastic neutral axis could be ignored with the increase of positive bending moment.

        The Gender-Sensitive Social Risk Factors for Internet Addiction in College Undergraduate Students

        Objective The current study aims to explore precipitating and social risk factors for internet addiction (IA) in university undergraduate students, and to provide evidence for interventions and the early prevention of IA in different genders. Methods Four thousand eight hundred and fifty-eight college sophomores completed an online survey on their internet use-related behaviours and social risk factors. Results We found that more male (8.3%) than female students (5.4%) had moderate and severe IA. The main online activity in the moderate and severe IA groups was online gaming in males and online streaming in females. Roommates engaging in similar internetbased entertainment was a risk factor of IA only for males, while not being in a romantic relationship was a risk factor of IA for females only. Infatuation with the internet before college and adjustment problems for college life were shared risk factors for both genders in the mild and moderate IA groups. Conclusion IA was a common phenomenon in college students with shared and unique precipitating and social risk factors in males and females. The gender-sensitive risk factors for IA warranted earlier and individualized intervention and prevention strategies for IA in this population.

      • MiR-421 Regulates Apoptosis of BGC-823 Gastric Cancer Cells by Targeting Caspase-3

        MicroRNAs might act as oncogenes or tumor suppressors in cancer. Recent studies have shown that miR-421 is up-regulated in human gastric cancer. Here, we found that miR-421 was over-expressed in gastric cancer tissues and cell lines. Bioinformatics analysis predicted that the caspase-3 gene was a target of miR-421. Caspase-3 was negatively regulated by miR-421 at the post-transcriptional level. Bax and Bcl-2 were also regulated by miR-421. Moreover, tumor necrosis factor receptor-I and -II, death receptors in the apoptosis pathway, were up-regulated by miR-421. The over-expression of miR-421 promoted gastric cancer cell growth and inhibited apoptosis of the BGC-823 gastric cancer cell line. These observations indicate that miR-421 acts as a tumor promoter by targeting the caspase-3 gene and preventing apoptosis of gastric cancer cells through inhibition of caspase-3 expression. These findings contribute to our understanding of the functions of miR-421 in gastric cancer.

        A Study on Cutting and Tribology Performances of TiN and TiAlN Coated Tools

        TiN and TiAlN coatings were deposited on the surface of AISI 5140 steel and cemented carbide YT15 by magnetron sputteringtechnique (MSP). The reciprocating sliding tests of TiN and TiAIN coatings on the surface of AISI 5140 steel were performed toinvestigate the friction coefficients of coatings affected by various normal loads with friction pair of 304 stainless steel balls. Drymachining tests on AISI 5140 hardened steel were carried out with the TiN and TiAlN coated tools on a CA6140A lathe. The effectsof cutting speed on cutting forces and surface roughness of TiN and TiAlN coated tools were obtained and analyzed to assess thecutting performance of coated tools. The microcosmic micrographs of wear areas of coated tools were observed and investigated byscanning electron microscope and energy dispersive spectrum. The results show that the friction coefficients of TiN coatings are lowerthan that of TiAlN coatings. The cutting force of TiAlN coated tool decreases and flank wear resistance enhances in comparison withTiN coated tool. The wear form and mechanisms of TiN coated tool are mainly crater wear on the rake face and adhesive wear andabrasive wear on the flank face. The wear form and mechanisms of TiAlN coated tool are mainly adhesive wear, the breakage ofcutting edge and the damage of tip, accompanied with diffusion and oxidation wear.
