In seeking aryl acylamidase (EC 3.5.1.13) acting on an amide bond in p-acetaminophenol (Tylenol$^{TM}$), we identified a novel gene encoding 496 residues of a protein. The gene revealed a conserved amidase signature region with a canonical catalytic triad. The gene was expressed in E. coli and characterized for its biochemical properties. The optimum pH and temperature for the activity on p-acetaminophenol were 10 and 37$^{\circ}C$, respectively. The half-life of enzyme activity at 37$^{\circ}C$ was 192 h and 90% of its activity remained after 3 h incubation at 40$^{\circ}C$. Divalent metals was found to inhibit the activity of enzyme. The $K_m$ values for various aryl acylamides such as 4-nitroacetanilide, p-acetaminophenol, phenacetin, 4-chloroacetanilide and acetanilide were 0.10, 0.32, 0.83, 1.9 and 19 mM, respectively. The reverse reaction activity (amide synthesis) was also examined using various chain lengths ($C_1{\sim}C_4$ and $C_{10}$) of carboxylic donors and aniline as substrates. These kinetic parameters and substrate specificity in forward and reverse reaction indicated that the aryl acylamidase in this study has a preference for aryl substrate having polar functional groups and hydrophobic carboxylic donors.
Therapeutic response of cryotherapy on actinic keratosis (AK): a retrospective analysis

Background: Actinic keratosis (AK) is a premalignant lesion of the skin caused by long-term ultraviolet radiation exposure. Cryotherapy is one of the most common treatment modalities for AK. But, there have been quite a few studies about the therapeutic response of cryotherapy on AK including cure rate, recurrence rate, side effects and etc.. Objectives: To investigate the therapeutic response of cryotherapy on AK in Korean patients. Methods: We enrolled and assessed 87 AK patients with 104 AK lesions who had pre-and post- standardized cryotherapy (freezing AK lesion with 3-5mm margin and 5-10s freezing time) at 4 months photos at the Pusan National University Hospitals (Busan and Yangsan) from 2010 through 2019. Results: The final cure rate and recurrence rate at 4 months after cryotherapy were 89.4% and 8.4% respectively. And, the cure rate at 2 months after cryotherapy were 65.3%. In the comparison of demographics, clinical findings and pathologic findings between cure group and non-cure group, AK clinical grade and histopathologic subtype showed a statistical significance. The side effect were various with postinflammatory hypopigmentation (30/104), persistent erythema (18/104), postinflammatory hyperpigmentation on margin (13/104) and scar (15/104). Conclusion: The result of this study could be useful for the dermatologists who are the main manager of AK treatment.

Purpose: To investigate the rater reliability of a Pediatric Balance Scale (PBS) for children with cerebral palsy, and to investigate possible differences among raters according to their clinical work experience and testing experience. Methods: Study participants included 18 children with spastic cerebral palsy who could walk. They were instructed by pediatric physical therapists, two of whom had ten years of clinical work experience and two who had less than one year of experience. The children`s ability to achieve physical balance was videotaped for PBS items. The raters watched the tapes and evaluated each child twice. Rater reliability was analyzed using the intraclass correlation coefficient (ICC). Differences between experienced and novice raters were analyzed using a paired t-test. The statistical significance level was set to 0.05. Results: The total PBS scores averaged 45.78~48.00 and 45.72~47.67 for first and second tests. Intra-rater reliability was very high (ICC=0.89~0.99), and the repeated measurement coincidence was high (p>0.05). Inter-rater reliability was high (ICC=0.83~0.84), but there was a bit of a difference in the coincidence (p<0.05). The experienced raters` reliability and coincidence were higher than those of the novices, and there were differences in reliance and coincidence between experienced and novice raters (p<0.05). Conclusion: Inter-rater and intra-rater reliability is very high. However, rater reliability showed defferences depending on clinical work experience and testing experience. When testing pediatric patients with the PBS, the rater`s clinical experience and test experience may affect the test results.
Active Use of DFIG-Based Variable-Speed Wind-Turbine for Voltage Control in Power System Operation

This paper presents an active use of doubly-fed induction-generator(DFIG)-based variable-speed wind-turbine for voltage control in power system operation. For reasonable simulation studies, a detail dynamic model of a DFIG-based wind-turbine grid-connected system is presented. For the research objective, an innovative reactive power control scheme is proposed that manipulates dynamically the reactive power from the voltage source converter(VSC) with taking into account its operating state and limits.

<P>Aryl acylamidase (EC 3.5.1.13, AAA) acts on the amide bond between aryl and acyl groups. Whole cells of Escherichia coli overexpressing a novel bacterial AAA synthesized p-acetaminophenol (p-AAP) from p-aminophenol (p-AP, aryl compound) and acetate (acyl donor). Optimum conditions were pH 5.5 and 35C with 100 mM p-AP and 600 mM sodium acetate in 100 mM sodium phosphate buffer including 1% (v/v) Triton X-100 for 60 h. 13.1 g p-AAP l(-1) was produced with a conversion yield of 87%.</P>
