        절수에 의한 mongolian gerbil 종뇌 및 간뇌에서 dopamine성 면역반응세포의 분포변화

        Nowadays, mongolian gerbil is notably utilized for the research of brain and water deprivation because of a congenital incomplete willis circle structure in the brain, audiogenic seizure in low noise, and special cholesterol metabolism without water absorption for a long time. In this study, we are intend to identify the morphological changes of the catecholaminergic neuron of brain according to the time lapse in the condition of long term water deprivation. 55 mongolian gerbil were divided 10 groups(control, 1, 2, 3, 4, 5, 10, 15, 20, 42th day water deprivation group), of which each group include 5 mongolian gerbils and 5 normal mongolian gerbils in control group were also used for brain atlas as a control. The brains were observed by the immunohistochemical stain using the TH, DBH and PMNT antibody. The results were as followings; 1. The nerve fibers of the TH-immunoreactive neuron were observed only in the and corpus striatum of the telencephalon. 2. Intensity of the immunostain of the nerve fiber in the cerebral cortex and corpus striatum was decreased gradually day by day after water deprivation. 3. The TH-immunoreactive nerve cells were observed in the paraventricular and periventricular nucleus of the 3rd ventricular in the hypothalamus of mongolian gerbil but the number of nerve cells were decreased from the first day of the water deprivation to the 10th day and increased until the 20th day, after than redecreased from the 20th day by the continuous water deprivation. The number of nerve fibers in this area were increased in the first day, but decreased from the 2nd day of water deprivation. The shape and density of the dopamine secreting cells in the brain of mongolian gerbil by the immunoreactive stain were changed in the continuous water deprivation. In this results, we can conclude that dopamine concerned in the water metabolism of mongolian gerbil, and mongolian gerbil could be used as an animal model for the research of water deprivation.

      • Interleukin 17 (IL-17) Increases the Expression of Toll-like Receptor-2, 4, and 9 by Increasing IL-1β and IL-6 Production in Autoimmune Arthritis

        <B>Objective.</B><P>To examine the effect of interleukin 17 (IL-17) on the expression of Toll-like receptor (TLR)-2, 4, and 9 in collagen-induced arthritis (CIA) in mice.</P><B>Methods.</B><P>On Days 28 and 32 after induction of CIA in mice, phosphate-buffered saline (PBS group) or IL-17 (IL-17 group) was injected into both knee joints. On Day 35, mice were sacrificed. The severity of knee joint arthritis, synovial inflammation, and bone destruction was measured by a scoring system using macrography and histological analysis. Synovial expression of TLR-2, 4, 9, IL-17, IL-1ß, tumor necrosis factor-α (TNF-α), and IL-6 was determined by real-time PCR and immunohistochemistry. Synoviocytes of CIA mice were cultured with IL-17 and with neutralizing antibodies to cytokine, and the expression of TLR-2, 4, 9, IL-1ß, TNF-α, and IL-6 was determined by real-time RT-PCR.</P><B>Results.</B><P>In CIA mice, knee arthritis scores, synovial inflammation, bone destruction scores, and expression of synovial TLR-2, 4, and 9, IL-17, IL-1ß, TNF-α and IL-6 were higher in the IL-17 and PBS groups than in normal DBA1 mice. These variables were also significantly higher in the IL-17 group than in the PBS group. In CIA synoviocytes, IL-17 increased the expression of TLR-2, 4, and 9, and this effect was significantly alleviated by neutralizing antibodies to IL-17, IL-1ß, and IL-6.</P><B>Conclusion.</B><P>IL-17 aggravates joint inflammation and destruction, and increases the synovial expression of TLR-2, 4, and 9 by increasing IL-1ß and IL-6. These results imply that the IL-17-induced increase in expression of TLR-2, 4, and 9, and IL-1ß and IL-6 production are involved in the IL-17-induced aggravation of arthritis.</P>

        Cloning and Expression of a Rice cDNA Encoding a Lls1 Homologue of Maize

        A cDNA (Oslls1) encoding Lls1-homologue of maize was isolated from cDNA library of rice (Oryza sativa cv. Ilpum). The 2,138 bp of full length Oslls1 clone contains an open reading frame of 1,623 nucleotides encoding 575 amino acid residues. The deduced amino acid sequence of Oslls1 has a high level of homology with chlorophyll a oxygenases of Arabidopsis thaliana (67%) and Marchantia polymorpha (65%). Southern blot analysis of genomic DNA indicates the existence of a small gene family for Oslls1 in the rice genome. The expression of Oslls1 mRNA was induced in leaves and germinating seeds. Treatment of $H_2O$$_2$significantly down-regulated Oslls1 expression. The expression of Oslls1 mRNA was consititutively down-regulated in the blm, a rice mutant exhibiting spontaneous necrotic lesions. These results suggest that this Oslls1 gene may be involved incell death mechanisms in the blm mutant of rice.

        Expression of TLR2, TLR4, and TLR9 in dermatomyositis and polymyositis

        <P>The aim of this study was to investigate the expressions of Toll-like receptor (TLR) 2, TLR4, TLR9, and their correlations with the expression of cytokines that are associated with activation of CD4<SUP>+</SUP> T cells and inflammation including interferon γ (IFNγ), interleukin 4 (IL4), interleukin 17 (IL17), and tumor necrosis factor α (TNFα) in muscle tissues of patients with dermatomyositis (DM) and polymyositis (PM). The expressions of TLR2, TLR4, TLR9, IFNγ, IL4, IL17, and TNFα were measured by real-time reverse transcription–polymerase chain reaction in muscle tissues from 14 patients with DM and PM (nine patients with DM, five patients with PM) and three controls. The expressions of TLR2, TLR4, and TLR9 were also localized with immunohistochemistry. The expression levels of TLR2, TLR4, TLR9, IFNγ, IL4, IL17, and TNFα were significantly high in patients with DM and PM compared with those in the controls, and the expression levels of TLR4 and TLR9 had significant positive correlations with the expressions of IFNγ, IL4, IL17, and TNFα. Immunohistochemistry showed that TLR2, TLR4, and TLR9 were expressed by infiltrating cells of perimysium in DM, whereas they were expressed by infiltrating cells of endomysium in PM. These results suggest that the involvement of TLR4 and TLR9 in immunopathogenesis of DM and PM might be connected with activation of CD4<SUP>+</SUP> T cells.</P>

      • 저수지 특성에 따른 EAP 붕괴홍수량과의 상관관계 분석

        최근 기상이변에 따른 대규모 호우 및 지진피해가 빈번히 발생하고 이로 인해 많은 인명 및 재산피해가 발생하고 있다. 강우의 형태 또한 수공구조물의 설계빈도를 무색하게 만들 정도로 단기간의 집중호우 양상을 띠고 있어 댐 붕괴와 같은 비상상황에 대한 실질적 대처방안이 요구되고 있으며 이와 더불어 댐과 저수지와 같은 수공구조물에 대한 안정성을 종합적으로 검토할 필요성이 있다. 따라서 본 연구에서는 저수지의 가상붕괴로부터 유출량을 산정하여 저수지 특성에 따른 붕괴홍수량과의 상관관계를 분석, 추후 저수지 주변 및 하류지역의 안정성 평가를 위한 EAP수립의 신뢰도 향상에 기여하고자 한다. 본 연구에서는 비상대처계획(Emergency Action Plan ; EAP)이 수립된 한국농어촌공사 관할 173개소의 저수지를 대상으로 하였으며, 총저수량이 100만㎥ 이상의 필댐을 선정하였다. 붕괴의 형태는 월류에 의한 붕괴를 모의하였고 이를 수행하기 위해 만수위 조건에서 PMF가 유입되는 상황을 채택하였으며 DAMBRK모형을 적용하였다. 붕괴부 형성시간은 대상저수지가 필댐인 점을 고려하여 0.1시간부터 0.5시간까지로 구성하고 붕괴부 평균폭은 댐 높이를 기준으로 가정하여 각각의 경우에 대한 붕괴모의를 실시하였으며, 붕괴부 측면경사는 기존의 여러 연구에서 적용한 바 있는 1:1로 가정하였다. 그 결과 첨두붕괴홍수량은 붕괴부 폭의 크기에 비례하고, 붕괴부 형성시간에 반비례하는 것으로 나타났다. 이를 바탕으로 제체높이, 총저수량, 제체높이×총저수량, 유역면적, 만수면적 PMF 등과 같은 저수지 특성별 붕괴에 따른 붕괴홍수량을 산정하여 이들 관계곡선의 상관관계를 회귀방정식으로 유도하였으며 그 결과를 비교·검토하기 위해 2002년 태풍 “루사” 내습 시 강릉 남대천 유역의 장현 및 동막저수지의 실제 붕괴사례를 적용해 보았다. 금회 연구 대상저수지는 총저수량 100만㎥ 이상의 저수지를 대상으로 분석하여 총저수량 100만㎥ 이하의 소규모 저수지에 대해서는 그 범위가 다소 벗어날 것이라 사료되지만, 저수지특성에 따른 붕괴홍수량의 상관관계 분석을 통해 저수지의 적정 붕괴홍수량에 대한 판단의 척도로 사용할 수 있으며, 이에 따른 홍수범람피해 예측과 최상의 EAP를 수립하는 데 도움이 될 것으로 판단된다.

        Kainic Acid가 미성숙 오리 망막에 미치는 영향에 관한 전자현미경적 연구

        In order to identify the effects of the kainic acid in the retina kainic acid (120 nmol/$5{\mu}l$) were injected through the pars plana of the ciliary body into the duck eye ball at 7th day after hatching. The animals were sacrificed by decapitation on the 1st, 4th and 10th day after injection of the kainic acid. The specimen was processed for the light and electron microscopes respectively. The results obtained were as followings 1. The inner nuclear, inner plexiform and ganglion cell layers of the retina showed marked changes in all of the groups. 2. The cells in the inner nuclear layer showed marked degenerative changes of the organelles including the pyknoses and destruction of the nucleus on the 1st day after injection. These destructive changes of the cells were progressively disappeared until the 10th day after injection. 3. After injection of the kainic acid the neurites of the inner plexiform layer were swollen at the first, and thereafter those changes were gradually disappeared until the 10th day. 4. In the ganglion cell defected by the kinic acid, the enlargement and segmentation of the endoplasmic reticulum and pyknosis of the nucleus were-observed in all the groups, although the lipid droplets were only shown in the 4th day. From the above results, the evidence suggests that kainic acid gives to the toxic effect on the nuurons of the retina with exception of the photoreceptors.

        Glutamate and GABA concentrations in the cerebellum of novel ataxic mutant Pogo mice

        The Pogo mouse is an autosomal recessive ataxic mutant that arose spontaneously in the inbred KJR/MsKist strain derived originally from Korean wild mice. The ataxic phenotype is characterized by difficulty in maintaining posture and side to side stability, faulty coordination between limbs and trunk, and the consequent inability to walk straight. In the present study, the cerebellar concentrations of glutamate and GABA were analyzed, since glutamate is a most prevalent excitatory neurotransmitter whereas γ-aminobutyric acid (GABA) is one of the most abundant inhibitory neurotransmitters, which may be the main neurotransmitters related with the ataxia and epilepsy. The concentration of glutamate of cerebellum decreased significantly in ataxic mutant Pogo mouse compared to those of control mouse. However, GABA concentration was not decrease. These results suggested that the decrease in glutamate concentration may contribute to ataxia in mutant Pogo mouse.

        Objective: We`ve research a tendency of how a study of Kimchi using herbal medicine and herbal food was developed. Methods: We investigated a recent tendency and functionality of herbal medicinal kimchi through researching studies and thesis of acadernic societies. Conclusion: Kimchi has demonstrated anticancer and weight reduction, cleaning of intestines, preventing blood clotting. It will be more conducted to produce more newly-developed various Kimchi and study of various functions and verification. A study of correct mechanism in process of fermentation and mature of Kimchi is required.
