Boswellic acid suppresses cyclophosphamide induced chromosomal damage in mice

This study was performed to determine whether Boswellic acid suppresses genotoxicity induced by cyclophosphamide in mice. Genotoxicity was induced by the administration of cyclophosphamide (50 mg/kg) in swiss albino mice and was measured by counting the number of micronuclei and observing chromosomal aberration. In test group Boswellic acid was given in the dose of 200 mg/kg and various chromosomal aberration and formation of micronuclei were observed. Boswellic acid in dose of 200 mg/kg significantly ($P$ <0.001) inhibited the cyclophosphamide induced micronucleus formation and also significantly ($P$ <0.001) inhibited chromosomal aberrations in mice. Whereas Boswellic acid alone did not either induce formation of micronuclei or chromosomal aberration in mice. Boswellic acid itself doesn't have any genotoxic effect and suppresses genotoxicity induced by cyclophosphamide.

The wild-type repressor CI of temperate mycobacteriophage L1 and the temperature-sensitive (ts) repressor CIts391 of a mutant L1 phage, L1cIts391, have been separately overexpressed in E. coli. Both these repressors were observed to specifically bind with the same cognate operator DNA. The operator-binding activity of CIts391 was shown to differ significantly than that of the CI at 32 to $42^{\circ}C$. While 40-95% operator-binding activity was shown to be retained at 35 to $42^{\circ}C$ in CI, more than 75% operator-binding activity was lost in CIts391 at 35 to $38^{\circ}C$, although the latter showed only 10% less binding compared to that of the former at $32^{\circ}C$. The CIts391 showed almost no binding at $42^{\circ}C$. An in vivo study showed that the CI repressor inhibited the growth of a clear plaque former mutant of the L1 phage more strongly than that of the CIts391 repressor at both 32 and $42^{\circ}C$. The half-life of the CIts391-operator complex was found to be about 8 times less than that of the CI-operator complex at $32^{\circ}C$. Interestingly, the repressor-operator complexes preformed at $0^{\circ}C$ have shown varying degrees of resistance to dissociation at the temperatures which inhibit the formation of these complexes are inhibited. The CI repressor, but not that of CIts391, regains most of the DNA-binding activity on cooling to $32^{\circ}C$ after preincubation at 42 to $52^{\circ}C$. All these data suggest that the 131st proline residue at the C-terminal half of CI, which changed to leucine in the CIts391, plays a crucial role in binding the L1 repressor to the cognate operator DNA, although the helix-turn-helix DNA-binding motif of the L1 repressor is located at its N-terminal end.

Global trends towards market economies have signalled the ascendancy of neo-liberal paradigms. Their impact in the Asia-Pacific region has brought about significant social and cultural change. Background: Roots of Withania somnifera (WS) are a celebrated medicinal ingredient in Ayurvedic and many other indigenous systems of medicine. The present study investigates the effect of the phytochemical composition of the extracts on their antioxidant and reducing activities. Methods: WS roots were extracted with water, acetone, aqueous methanol (1:1), and methanol:-chloroform:water (1:1:1) to obtain aqueous, acetone, hydro-methanolic, and methanol-chloroform-water extracts. Thereafter, phytochemical constitution and antioxidant and reducing activities of the extracts were compared using different qualitative and quantitative tests. Results: Maximum extraction recovery was obtained with 50% aqueous methanol whereas extraction with acetone yielded the poorest recovery. Methanol-chloroform-water extract had the highest content of phytochemical constituents, except tannins, and also exhibited the highest antioxidant and reducing activities. Conclusion: Phytochemical composition and antioxidant and reducing activities of the extracts were positively associated with the use of organic solvents during the extraction process. Alkaloids and flavonoids were the most important contributors in the antioxidant and reducing activities of the extracts.

<P>The auxin efflux carrier PIN2 has a long hydrophilic loop (HL); when transplanted into PIN5 (a short-HL PIN), this HL confers the PIN5:PIN2-HL fusion protein with the capacity for phosphorylation and plasma membrane (PM) targeting but not for PIN2-like polarity. Moreover, depending on cell type, PIN5 can show internal, PM, or polar localization in the PM.</P>

<P>The subcellular polarity of PIN-FORMEDs (PINs) is critical for directional cell-to-cell transport of auxin. Phosphorylation of PIN proteins plays an important role in generating and maintaining specific PIN polarity. In a recent study, we have shown that phosphorylation in certain conserved residues of the PIN3 hydrophilic loop (HL) modulates its subcellular localization and polarity in a cell type-specific manner in different root tissues. Here, we additionally show that the phosphorylation code of PIN3-HL is operational for the determination of PIN3 polarity in the Arabidopsis guard cell and is deciphered in a differential way even in a single tobacco cell for the intracellular trafficking of PIN3. On the other hand, PIN3 localization often remained unaltered in certain cell types irrespective of its phosphorylation status. These findings, together with previous reports, indicate that the phosphorylation code of the PIN-HL along with cell type-specific factors, kinases, and developmental/environmental cues is instrumental for the PIN trafficking to different subcellular compartments as well as different plasma membrane domains.</P>

<P><B>Summary</B></P><P>Directional cell‐to‐cell movement of auxin is mediated by asymmetrically localized PIN‐FORMED (PIN) auxin efflux transporters. The polar localization of PINs has been reported to be modulated by phosphorylation. In this study, the function of the phosphorylation sites of the PIN3 central hydrophilic loop (HL) was characterized. The phosphorylation sites were located in two conserved neighboring motifs, RKSNASRRSF(/L) and TPRPSNL, where the former played a more decisive role than the latter. Mutations of these phosphorylatable residues disrupted <I>in planta</I> phosphorylation of PIN3 and its subcellular trafficking, and caused defects in PIN3‐mediated biological processes such as auxin efflux activity, auxin maxima formation, root growth, and root gravitropism. Because the defective intracellular trafficking behaviors of phospho‐mutated PIN3 varied according to cell type, phosphorylation codes in PIN3‐HL are likely to operate in a cell‐type‐specific manner.</P>

Purpose: The objective of this study was to investigate the effect of varying resolutions of cone-beam computed tomography images on the accuracy of linear measurements of edentulous areas in human cadaver heads. Intact cadaver heads were used to simulate a clinical situation. Materials and Methods: Fiduciary markers were placed in the edentulous areas of 4 intact embalmed cadaver heads. The heads were scanned with two different CBCT units using a large field of view ($13cm{\times}16cm$) and small field of view ($5cm{\times}8cm$) at varying voxel sizes (0.3 mm, 0.2 mm, and 0.16 mm). The ground truth was established with digital caliper measurements. The imaging measurements were then compared with caliper measurements to determine accuracy. Results: The Wilcoxon signed rank test revealed no statistically significant difference between the medians of the physical measurements obtained with calipers and the medians of the CBCT measurements. A comparison of accuracy among the different imaging protocols revealed no significant differences as determined by the Friedman test. The intraclass correlation coefficient was 0.961, indicating excellent reproducibility. Inter-observer variability was determined graphically with a Bland-Altman plot and by calculating the intraclass correlation coefficient. The Bland-Altman plot indicated very good reproducibility for smaller measurements but larger discrepancies with larger measurements. Conclusion: The CBCT-based linear measurements in the edentulous sites using different voxel sizes and FOVs are accurate compared with the direct caliper measurements of these sites. Higher resolution CBCT images with smaller voxel size did not result in greater accuracy of the linear measurements.
