<P>Protein transduction domains (PTDs) have been shown to cross the biological cell membranes efficiently through a receptor and energy independent mechanism. Because of its ease in membrane transducing ability, PTDs could be used as a gene delivery vector. Since we already have shown that purified Hoxc8 homeoprotein has the ability to cross the cellular membrane, we analyzed the possibility of the third helix of the Hoxc8 homeodomain as a useful gene delivery vector. For that purpose, a 16-aa long synthetic oligopeptide Hoxc8 Protein Transduction Domain (HPTD) was chemically synthesized and then tested to see whether the HPTD could form a complex with DNA or not. Gel retardation analysis revealed that the HPTD interacts with plasmid DNA efficiently but failed to transfer the DNA into the cells. However, HPTD can enhance the efficiency of gene transfer in combination with Lipofectamine which doubled the gene transfer rate into COS-7 cells compared with the DNA/Lipofectamine control. An MTT assay indicated that the amount of HPTD used in the complex for the transfection did not show any cytotoxicty in COS-7 cells. The TEM studies showed compact particle formation in the presence of HPTD. These results indicate that the HPTD could be a good candidate adjuvant molecule to enhance the gene transfer efficiency of Lipofectamine in eukaryotic cells.</P>

β-glucosylceramide (GC) is a naturally occurring glycosphingolipid that was shown to improve hepatic steatosis, steatohepatitis, and insulin resistance in animal models of nonalcoholic fatty liver disease. In this study, we evaluated the safety and efficacy of oral administration of GC in subjects with nonalcoholic steatohepatitis (NASH). Twenty-three patients with biopsy proven NASH were enrolled in a double-blind, placebo-controlled trial. Patients were orally administered daily with 7.5 mg of GC. Patients were followed for safety, liver enzymes, HbA1c, insulin sensitivity, lipid profile, hepatic fat content as measured by magnetic resonance imaging (MRI), and NASH score on liver biopsy. No treatment-related adverse events were observed during treatment. In a per protocol analysis of data, oral administration of GC decreased the hepatic fat content as measured by MRI in GC-treated compared with placebo. HbA1C decreased in patients treated with GC. GC treatment was associated with a milder decrease in the high-density lipoprotein serum levels. The beneficial effects were associated with a decrease in CD4 and NKT cell subsets of lymphocytes. Due to the small number of subjects enrolled, differences did reach statistical significance. Oral administration of GC is safe and biologically active in patients with NASH and insulin resistance.

<P><I>Sox11</I> deletion mice are known to exhibit developmental defects of craniofacial skeletal malformations, asplenia, and hypoplasia of the lung, stomach, and pancreas. Despite the importance of <I>Sox11</I> in the developing skeleton, the role of <I>Sox11</I> in osteogenesis has not been studied yet. In this study, we identified that <I>Sox11</I> is an important transcription factor for regulating the proliferation and survival of osteoblast precursor cells as well as the self-renewal potency of mesenchymal progenitor cells via up-regulation of <I>Tead2.</I> Furthermore, <I>Sox11</I> also plays an important role in the segregation of functional osteoblast lineage progenitors from osteochondrogenic progenitors. Facilitation of osteoblast differentiation from mesenchymal cells was achieved by enhanced expression of the osteoblast lineage specific transcription factors <I>Runx2</I> and <I>Osterix</I>. Morpholino-targeted disruption of <I>Sox11</I> in zebrafish impaired organogenesis, including the bones, which were under mineralized. These results indicated that <I>Sox11</I> plays a crucial role in the proliferation and survival of mesenchymal and osteoblast precursors by <I>Tead2</I>, and osteogenic differentiation by regulating <I>Runx2</I> and <I>Osterix</I>.</P>
Fashion Granfluencers as a Cultural Intermediary in Fashion and Style -A Case Study on Iris Apfel-

This study investigated the role of fashion granfluencers as cultural intermediaries in the present digital era. The concept of fashion granfluencer as cultural intermediaries and their fashion style characteristics were defined and examined. The literature on cultural intermediaries, social media, and seniors in the context of fashion was reviewed. Primarily, three characteristics of cultural intermediaries‒interactive communication, nonconformist attitude, and aesthetic disposition‒were identified and used as analytical variables in this study. Through netnography, 1,452 Instagram posts from the renowned fashion granfluencer Iris Apfel (@iris.apfel) were examined. This study presented Iris as a fashion granfluencer and a senior professional, who uses fashion to express herself and define her identity. As a cultural mediator, she contributes to revitalizing the cultural economy and creating cultural products. Iris Apfel' fashion and style embody her digital persona. Her engagement in fashion activities‒producing fashion content or attending fashion events‒mediates the relationship between fashion producers and fashion consumers, thereby promoting the aesthetic value of fashion.

The purpose of this study is to determine the concepts of heterotopic image and fashion space, and the characteristics of fashion space and images from the perspective of fashion brands and users. This study examines the evolution of fashion space and consumers with it, based on Foucault’s theory of heterotopia, which refers to spaces that blend contradictory features not typically found within a single physical structure. This is accomplished by employing a single case study of Ader Error’s Ader Space, a Seoul-based brand known for its unique approach to presenting and communicating fashion. Based on an analysis of Instagram posts of Ader Error along with the hashtag searches “aderspace” and “adererror”, this study categorizes heterotopia from the perspective of fashion brands into three properties: fashion space as a medium for selling fashion products; fashion space as getaway to hybrid fashion practices; and fashion space as an illusionary place to experience fashion. From the user perspective, the heterotopic image of Ader Space portrayed on Instagram is characterized by the image of fashion products in an extraordinary fashion space, the image of a fashion space beyond space and time, and the image of exposing the hidden and the illusioncompensation of fashion space. This study contributes to a heightened understanding of the evolutionary concept of the fashion space.

Volcanic eruptions have serious impacts on ecosystems fromindividual behavior to community structure. Volcanic ash has an especially widespread effect on organisms, as it can be dispersed over a greater distance by the wind than are other volcanic products.We examined the effect of volcanic ash on calling songs and mating behavior of themale field cricket Gryllus bimaculatus De Geer (Orthoptera: Gryllidae). Adult males were randomly assigned to one of four treatments comprising different floormaterial (volcanic ash, filter paper, vermiculite, and Akadama soil), and parameters of calling songs andmating behaviorweremeasured. The dominant frequency of calling song lowered by the volcanic ash. Males reared on volcanic ash and soil exhibited lower calling rates per night. Furthermore, males reared on ash and vermiculite exhibited a longer latency to courtship period than those reared on the other floor materials. Our results showed that floor material does affect the mating efforts of male field crickets.
