Plasma zinc's alter ego is a low-molecular-weight humoral factor

<P>Mild dietary zinc deprivation in humans and rodents has little effect on blood plasma zinc levels, and yet cellular consequences of zinc depletion can be detected in vascular and other tissues. We proposed that a zinc-regulated humoral factor might mediate the effects of zinc deprivation. Using a novel approach, primary rat vascular smooth muscle cells (VSMCs) were treated with plasma from zinc-deficient (<1 mg Zn/kg) or zinc-adequate (35 mg Zn/kg, pair-fed) adult male rats, and zinc levels were manipulated to distinguish direct and indirect effects of plasma zinc. Gene expression changes were analyzed by microarray and qPCR, and incubation of VSMCs with blood plasma from zinc-deficient rats strongly changed the expression of >2500 genes, compared to incubation of cells with zinc-adequate rat plasma. We demonstrated that this effect was caused by a low-molecular-weight (∼2-kDa) zinc-regulated humoral factor but that changes in gene expression were mostly reversed by adding zinc back to zinc-deficient plasma. Strongly regulated genes were overrepresented in pathways associated with immune function and development. We conclude that zinc deficiency induces the production of a low-molecular-weight humoral factor whose influence on VSMC gene expression is blocked by plasma zinc. The competition of maintenance services in the offshore wind industry is continually increasing. The quality of the services acts as the distinguishing feature in the industry. Furthermore, there are public standards, which lead to the permanent necessity to offer further education and training programs for employees. To meet the requirements for further training in the specific field of application within the offshore wind industry, a gamified e-learning application has been developed and is introduced in this paper. It consists of a complete solution, which contains the automated analysis of service protocols to identify qualification needs, the involvement of service technicians in the generation of learning materials, the preparation, transmission as well as the further development of those materials in accordance with the principles of e-learning. Finally, the solution contains a gamified mobile application for qualification, which is designed to meet the individual learning needs of the service technicians. This concept paper follows a problem-centred approach. Based on the current state of technology and research, the problem and motivation are identified and the urgency is verified. Furthermore, a detailed specification of the solution and a first implementation approach is presented.
Steel-UHPC composite dowels’ pull-out performance studies using machine learning algorithms

Composite dowels are implemented as a powerful alternative to headed studs for the efficient combination of Ultra High-Performance Concrete (UHPC) with high-strength steel in novel composite structures. They are required to provide sufficient shear resistance and ensure the transmission of tensile forces in the composite connection in order to prevent lifting of the concrete slab. In this paper, the load bearing capacity of puzzle-shaped and clothoidal-shaped dowels encased in UHPC specimen were investigated based on validated experimental test data. Considering the influence of the embedment depth and the spacing width of shear dowels, the characteristics of UHPC square plate on the load bearing capacity of composite structure, 240 numeric models have been constructed and analyzed. Three artificial intelligence approaches have been implemented to learn the discipline from collected experimental data and then make prediction, which includes Artificial Neural NetworkParticle Swarm Optimization (ANN-PSO), Adaptive Neuro-Fuzzy Inference System (ANFIS) and an Extreme Learning Machine (ELM). Among the factors, the embedment depth of composite dowel is proved to be the most influential parameter on the load bearing capacity. Furthermore, the results of the prediction models reveal that ELM is capable to achieve more accurate prediction.

<P>CYP51 exists in all organisms that synthesize sterols de novo. Plant CYP51 encodes an obtusifoliol 14alpha-demethylase involved in the postsqualene sterol biosynthetic pathway. According to the current gene annotation, the Arabidopsis (Arabidopsis thaliana) genome contains two putative CYP51 genes, CYP51A1 and CYP51A2. Our studies revealed that CYP51A1 should be considered an expressed pseudogene. To study the functional importance of the CYP51A2 gene in plant growth and development, we isolated T-DNA knockout alleles for CYP51A2. Loss-of-function mutants for CYP51A2 showed multiple defects, such as stunted hypocotyls, short roots, reduced cell elongation, and seedling lethality. In contrast to other sterol mutants, such as fk/hydra2 and hydra1, the cyp51A2 mutant has only minor defects in early embryogenesis. Measurements of endogenous sterol levels in the cyp51A2 mutant revealed that it accumulates obtusifoliol, the substrate of CYP51, and a high proportion of 14alpha-methyl-delta8-sterols, at the expense of campesterol and sitosterol. The cyp51A2 mutants have defects in membrane integrity and hypocotyl elongation. The defect in hypocotyl elongation was not rescued by the exogenous application of brassinolide, although the brassinosteroid-signaling cascade is apparently not affected in the mutants. Developmental defects in the cyp51A2 mutant were completely rescued by the ectopic expression of CYP51A2. Taken together, our results demonstrate that the Arabidopsis CYP51A2 gene encodes a functional obtusifoliol 14alpha-demethylase enzyme and plays an essential role in controlling plant growth and development by a sterol-specific pathway.</P>

<P>In recent years, bismuth has been promoted as a 'green element' and is used as a substitute for the toxic lead in ammunition and other applications. However, the bioavailability and toxicity of bismuth is still not very well described. Following a hunting accident with bismuth-containing shots, a bioavailability study of bismuth from metal pellets inoculated into rat limb muscles was carried out. Bismuth could be found in urine and blood of the animals. Bio-imaging using laser ablation ICP-MS of thin sections of the tissue around the metal implant was carried out to find out more about the distribution of the metal diffusing into the tissue. Two laser ablation systems with different ablation cell designs were compared regarding their analytical performance. Low concentrations of bismuth showing a non-symmetrical pattern were detected in the tissue surrounding the metal implant. This was partly an artefact from cutting the thin sections but also bio-mobilisation of the metals of the implant could be seen. An accumulation of zinc around the implant was interpreted as a marker of inflammation. Challenges regarding sample preparation for laser ablation and bio-imaging of samples of diverse composition became apparent during the analysis.</P>
