        miR-98 suppresses melanoma metastasis through a negative feedback loop with its target gene IL-6

        Dysregulated microRNA (miRNA) expression has a critical role in tumor development and metastasis. However, the mechanism by which miRNAs control melanoma metastasis is unknown. Here, we report reduced miR-98 expression in melanoma tissues with increasing tumor stage as well as metastasis; its expression is also negatively associated with melanoma patient survival. Furthermore, we demonstrate that miR-98 inhibits melanoma cell migration in vitro as well as metastatic tumor size in vivo. We also found that IL-6 is a target gene of miR-98, and IL-6 represses miR-98 levels via the Stat3-NF-κB-lin28B pathway. In an in vivo melanoma model, we demonstrate that miR-98 reduces melanoma metastasis and increases survival in part by reducing IL-6 levels; it also decreases Stat3 and p65 phosphorylation as well as lin28B mRNA levels. These results suggest that miR-98 inhibits melanoma metastasis in part through a novel miR-98-IL-6-negative feedback loop.

      • Pre-treatment Elevated Platelet Count Associates with HER2 Overexpression and Prognosis in Patients with Breast Cancer

        Purpose: To research the association between pre-treatment elevated platelet count and clinicopathologic characteristics in breast cancer (BC), as well as explore the relationship between pre-treatment elevated platelet count and HER2 status and prognosis of BC patients. Materials and Methods: A retrospective cohort of BC patients who were newly diagnosed or treated by surgery only and had pathological detection results and platelet values in the Department of Oncology, the First Affiliated Hospital of Liaoning Medical College were enrolled from 1/1/2008 until 31/12/2009, and followed up until 31/12/2014. Age, thrombocyte parameters before chemotherapy and/or radiotherapy, immunohistochemical (IHM) indexes, and regional lymph node (LN) involvement and progression-free survival (PFS) were recorded. Results: A total of 447 eligible subjects were included in this research. As we analyzed, for HER2, positive and negative, the incidence rates of elevated platelet count were 25.8% and 14.7% (P<0.05). In the Cox proportional hazards model both variables were independent risk factors for BC (for HER2, OR, 0.592, 95% confidence interval, CI, 0.355 to 0.985, P=0.044;f or PLT, OR, 0.998, 95% CI, 0.996 to 1.000, P=0.042). For ER, PR, Ki67 and LN involvement, the differences were not statistically significant (P>0.05). Conclusions: In this research, pre-treatment elevated level of platelet count demostrated a significantrelationship with HER2 amplification/overexpression, and both variables significantly influenced the prognosis of BC. However, elevated platelet count did not exhibit any association with ER, PR, Ki67 and LN involvement.

      • Making of Free View point 3D Images Using One Camera

        In recent years, it is necessary to show the sight of a remote place as a picture. In this case, to get an abundant amount of information. As the conventional technique, There were method which use two or more cameras to take photographs, and an ther method which use several photographs to generate a middle view point. However, the former, It tends to become an expensive and complicated system, and is the latter, there is a problem that information is missing with occlusion. In this paper, we propose a system which use done camera and a spin mirror to generate the free view point 3D image. This system used a method which called a light processing algorithm to make the free view point 3D image from the obtained photography picture. There liability of the proposed method is discussed with experimental result.

        ‘데페이즈망’ 표현기법을 통한 차별적 상업 공간 디자인 방법 연구

        The main purpose of this study is to conduct in-depth research on the performance characteristics of payment technology applied in the field of commercial space environment design. This study aims to understand the impact of these technologies on tourists. From a methodological perspective, this study provides a review of surrealist art works, with a particular emphasis on the visual expression techniques frequently used by surrealist artists. The article explores the interaction between these technologies and spatial design, thereby revealing innovative design methods in commercial space environment design practice. Initially, we needed to analyze the artistic practices of four famous surrealist painters, including Ren é Fran ç ois Ghislain Magritte, Salvador Dal í, Max Ernst, and others. Their works draw inspiration from the subconscious and deliberately blur the boundary between reality and imagination. In this analysis, the study depicted the inherent common characteristics of these technologies, including spatiotemporal properties, symbolic meanings, a preference for contradictions, and a tendency towards subtle dialectical differences. From these insights, the study continues to adopt design principles to synthesize these detailed findings. Subsequently, we designed a comprehensive evaluation framework, carefully customized it, and applied the evaluation method to four typical commercial space design cases based on screening. We hope to reveal the subtle implementation of payment technology in these cases and compare them with traditional thematic practices to determine innovative design methods.

        EGFR-TKI Combined with Pemetrexed versus EGFR-TKI Monotherapy in Advanced EGFR-Mutated NSCLC: A Prospective, Randomized, Exploratory Study

        Purpose We aimed to evaluate whether the addition of pemetrexed is effective in improving progression-free survival (PFS) in epidermal growth factor receptor (EGFR)–mutated patients with or without concomitant alterations. Materials and Methods This multicenter clinical trial was conducted in China from June 15, 2018, to May 31, 2019. A total of 92 non–small cell lung cancer (NSCLC) patients harboring EGFR-sensitive mutations were included and divided into concomitant and non-concomitant groups. Patients in each group were randomly treated with EGFR–tyrosine kinase inhibitor (TKI) monotherapy or EGFR-TKI combined with pemetrexed in a ratio of 1:1. PFS was recorded as the primary endpoint. Results The overall median PFS of this cohort was 10.1 months. There were no significant differences in PFS between patients with and without concomitant and between patients received TKI monotherapy and TKI combined with pemetrexed (p=0.210 and p=0.085, respectively). Stratification analysis indicated that patients received TKI monotherapy had a significantly longer PFS in non-concomitant group than that in concomitant group (p=0.002). In concomitant group, patients received TKI combined with pemetrexed had a significantly longer PFS than patients received TKI monotherapy (p=0.013). Molecular dynamic analysis showed rapidly emerging EGFR T790M in patients received TKI monotherapy. EGFR mutation abundance decreased in patients received TKI combined chemotherapy, which supports better efficacy for a TKI combined chemotherapy as compared to TKI monotherapy. A good correlation between therapeutic efficacy and a change in circulating tumor DNA (ctDNA) status was found in 66% of patients, supporting the guiding role of ctDNA minimal residual disease (MRD) in NSCLC treatment. Conclusion EGFR-TKI monotherapy is applicable to EGFR-sensitive patients without concomitant alterations, while a TKI combined chemotherapy is applicable to EGFR-sensitive patients with concomitant alterations. CtDNA MRD may be a potential biomarker for predicting therapeutic efficacy.

        Localization of α-, γ-, and δ-tubulin in the hypotrich ciliate Stylonychia pustulata (Hyportrichida, Ciliophora)

        Ciliature microtubular organelles, which organize into ciliary units, are highly complex microtubular systems. Toinvestigate the formation of ciliature microtubular organelles and the role that different tubulins play in this process,we examined the distribution and function of α-, γ-, and δ-tubulins in the hypotrich ciliate Stylonychia pustulata. Immunofluorescence analysis demonstrated that α-tubulin localized to all microtubular organelles, while γ- and δ-tubulinscolocalized at basal bodies. Immunogold labeling revealed that γ- and δ-tubulins colocalize to the proximalpart of basal bodies, suggesting that γ- and δ-tubulins probably interact and may function together. In addition, thepermanent localization of γ-tubulin to the adoral zone of membranelles suggests that γ-tubulin play a role in themaintenance of oral ciliature base-associated microtubules, but does not play a role in the biogenesis of theseciliature. We observe that δ-tubulin is restricted to the basal body and the connective fibers of the basal body,suggesting this protein may be required for the extension and stability of the microtubules in these structures. Dramatically different concentrations of these three tubulins were observed in the cytoskeleton and the insoluble andsoluble fractions of the cell, suggesting that these highly related proteins perform different functions in S. pustulata.

        Exploring the spatio-temporal characteristics and driving factors of urban expansion in Xi'an during 1930–2014

The researchfocusesonindicatorsincludingexpansionintensity, expansion direction,citycentreofgravity,compactness,and elasticity, andidentifyanddiscussesthedrivingfactorsofurban expansion. TheresultsshowthattheurbanexpansionofXi’an has undergonetwocyclicalchangesduring1930-2014.Theurban form presentsa ’horizontal rectangle-invertedtriangle-concave’ evolution feature.Thecitycentreofgravityshiftstothe northwest. Thecompactnesscontinuestodecline,andthe elasticity coefficient rise.Ithasbeenidentified thateconomicand demographic factorstogetherwiththeindustrialstructurehave driven urbanexpansion,andurbanplanningpolicieshas influenced thedirectionsoftheexpansion.Theconservationof great ruinsinXi’an hasaffected theevolutionofitsurban morphology. Basedontheresults,wefurtherdiscussthefuture expansion directionsofthecity,andprovidespecific suggestions for futureurbanplanninginordertobettercoordinateurban expansion, andatthesametimeprovideadequateprotectionto large heritagesitesandsupporturbansustainability.These findings providenotonlyvaluableinsightsabouturbanplanning of Xi’an, themethodologicalapproachdevelopedinthisresearch is alsoapplicabletootherhistoriccitieswithsimilar characteristics inChinaandbeyond.

        Molecular and Cellular Microbiology / Biomedical Sciences : A Homogeneous Immunoassay Method for Detecting Interferon-Gamma in Patients with Latent Tuberculosis Infection

        IFN-γ release assays (IGRAs) have been developed as viable alternative diagnostic tools for detecting latent tuberculosis infection (LTBI). A customized homogeneous sandwich luminescent oxygen channeling immunoassay (LOCI) was used to quantify IFN-γ levels in IGRAs. Samples were collected from healthy volunteers (n = 40) who were T-Spot-negative and T-Spot-positive patients (n = 32) at rest. Then the amount of IFN-γ in the supernatant of IGRAs was measured by LOCI. The results demonstrated a low background, and high sensitivity, specificity, accuracy, and reproducibility, and a short assay time (only 30 min) with LOCI for IFN-γ. The recovery range was 81.63-102.06%, the coefficients of variation were below 5%, and the limit of detection was 19.0 mIU/ml. Excellent agreement between LOCI IFN-γ and the T-SPOT.TB test was obtained (97.2% agreement, κ = 0.94). The LOCI IFN-γ concentrations were significantly higher in T-Spot-positive patients than in the healthy group (p < 0.001). Moreover, as observed for the comparative LOCI IFN-γ assay, IFN-γ concentrations were related to the numbers of T-SPOT.TB spots. We have established an in vitro blood test for LTBI diagnosis, defined as LOCI IFN-γ. A high level of agreement between the LOCI IFN-γ method and T-SPOT.TB assay was observed in clinical studies that showed the LOCI IFN-γ method could determine LTBI. This study shows acceptable performance characteristics of the LOCI IFN-γ assay to diagnose LTBI.

      • Diffusion Mechanism of Lithium Ion through Basal Plane of Layered Graphene

        <P>Coexistence of both edge plane and basal plane in graphite often hinders the understanding of lithium ion diffusion mechanism. In this report, two types of graphene samples were prepared by chemical vapor deposition (CVD): (i) well-defined basal plane graphene grown on Cu foil and (ii) edge plane-enriched graphene layers grown on Ni film. Electrochemical performance of the graphene electrode can be split into two regimes depending on the number of graphene layers: (i) the corrosion-dominant regime and (ii) the lithiation-dominant regime. Li ion diffusion perpendicular to the basal plane of graphene is facilitated by defects, whereas diffusion parallel to the plane is limited by the steric hindrance that originates from aggregated Li ions adsorbed on the abundant defect sites. The critical layer thickness (<I>l</I><SUB>c</SUB>) to effectively prohibit substrate reaction using CVD-grown graphene layers was predicted to be ∼6 layers, independent of defect population. Our density functional theory calculations demonstrate that divacancies and higher order defects have reasonable diffusion barrier heights allowing lithium diffusion through the basal plane but neither monovacancies nor Stone-Wales defect.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jacsat/2012/jacsat.2012.134.issue-20/ja301586m/production/images/medium/ja-2012-01586m_0003.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/ja301586m'>ACS Electronic Supporting Info</A></P>
