The purpose of this study is to verify the effect of STEM education on affective characteristics for Science, Technology and Engineering of middle school students. The specific objectives of the study are as follows: 1) To develop STEM program in Technology education for middle school students. 2) To analyze the effect of STEM education on affective characteristics for science, technology and engineering. The results of this study were as follows: First, STEM program focusing on Technology education of middle school consists of worksheets, portfolios for students and a lesson plan for teacher, including evaluation questionnaire of affective domain. Second, Students performed Making a fan using solar cell module with a creative design. The students affective characteristics for science, technology and engineering had changed positively in the result of quantitative test. 70% of the students provided positive answers for their interest and understanding about the program after applied to STEM education program. Third, the result of students answering in the qualitative test is that STEM education program was instructive and interesting. The follow up experimental research should proceed to verify the educational efficiency in the area.
Antibiotic Resistance and Assessment of Food-borne Pathogenic Bacteria in Frozen Foods

One hundred ninety-three frozen food samples collected in Korea various public bazaars from October 2006 to September 2007. Staphylococci were detected in 21.8% of frozen food samples. Staphylococcus aureus was isolated from 17 (8.8%) samples. Other staphylococci isolates were identified as S. warneri (7.8%), S. epidermidis (2.1%), S. xylosus (1.6%), S. eguorum (1%), and S. vitulinus (0.5%). Additionally, the antimicrobial susceptibility of 42 staphylococcal isolates to ten different antimicrobial agents was determined. The staphylococcal isolates demonstrated antimicrobial resistance to mupirocin (31%) oxacillin (14.3%), gentamicin (9.5%), teicoplanin (7.1%) and ciprofloxacin (7.1%). Most of the staphylococcal isolates showed high-level resistance to mupirocin (MIC90, >128 μg/mL). Fortunately, most of the isolates were susceptible to vancomycin. The total bacteria and Escherichia coli count were tested to investigate the microbiological quality of frozen foods. From 193 frozen food samples, 43 (22.3%), 34 (17.6%) and 19 (9.8%) samples were shown to be of unacceptable quality due to total bacteria, coliform and E. coli counts, respectively.

A meningioma is the second most common primary intracranial tumor of the central nervous system. One critical obstacle in meningioma research and preclinical development of novel therapeutic agents is a relative lack of suitable preclinical in vitro and in vivo model systems. In the current study, we assessed the proliferative characteristics of patient derived five primary meningioma cancer cell lines (WHO grade I and II) from brain tumor lesions. All of the meningioma cell lines showed homogenous expression of meningioma marker, VIMENTIN. The GTG-banding analysis determined the existence of different patterns of chromosomal abnormalities in representative cancer cell lines. The almost of the meningioma cell lines showed homogeneously spindle shaped cells, except for M160425 which have two prominent cell morphologies, spindle and round. Population-doubling (PD) was measured for every passage. The M160425 cell line had significantly longer PD time (39.8 ± 0.9 h, P<0.05) than the other meningioma cell lines. Consistent with the PD time, we confirmed that mRNA expression of Ki67, the conventional proliferation marker, was significantly lower in M160425 cell line compared to the other cell lines. The correlation between the PD time and the abundance of Ki67 in the meningioma derived cell lines was negative, indicating higher Ki67 abundance and a shorter PD time. The patient derived meningioma cancer cell lines established in this study might contribute to understanding cancer biology and help the success in the clinic by explaining the slightly different clinical behavior among the patients.
A case of splenic extraskeletal mesenchymal chondrosarcoma in a Yorkshire Terrier dog

A 7-year-old male Yorkshire Terrier dog was present to an animal clinic and a large soft mass was found in the spleen by radiological examination, and total splenorectomy was performed. Grossly, a large protruded splenic mass was soft to moderately firm and multilobulated. On the cut surface, the mass was off-white to tan, dark red, and rust colored with many cavitation and had gelatinous areas. Histologically, the tumor cells were characterized by coexistence of the primitive mesenchymal tissues and mature or immature cartilage tissues. Primitive mesenchymal areas were composed of round/oval or spindle shaped immature cells with high mitosis. The tumor cells of the cartilage areas were located in basophilic cartilaginous matrix. Intercellular matrix in the cartilaginous areas was stained blue with Masson’s trichrome and deep blue with alcian blue, respectively. Immunohisto-chemically, the cartilaginous tumor cells demonstrated positive reactions for vimentin and S-100, and surrounding mesenchymal tumor cells are immunopositive for vimentin. This case was diagnosed as splenic extraskeletal mesenchymal chondrosarcoma of a Yorkshire Terrier dog, a toy breed.
Analysis and Sorting of CD133+ Cancer Stem Cells derived from Human Atypical Meningioma

CD133, originally classified as a neural stem cell marker, could be used for enrichment of cancer stem cells (CSCs). In the current study, we analyzed the immunophenotypic characteristics of cancer cell lines derived from brain tumor lesions of 5 meningioma patients. Interestingly, tumor spheres only generated from the specimen of high-grade meningioma cell line (atypical meningioma cell line, WHO grade II), whereas there was no sphere formation in typical meningioma cell line (WHO grade I). The self-renewing capacity of these tumor sphere was assayed by dissociation of primary tumor spheres, and plating of single cells and maintained in the conditional medium. These single cells produced secondary sphere formation after another 7 days, indicating that these meningioma cell lines in spheres were capable of self-renewal and proliferation. Using flow cytometry, we observed the pattern of expression of dual markers CD44/CD133 in atypical meningioma cell line. The cytometry dot plot of CD133 expression in atypical meningioma cells with about 20.84% CD133− (gate Q4) and 79.15% CD133+ cells (gate Q2) before sorting. The CD44+CD133+ subpopulations showed stem cell properties, including extensive proliferation, instead of their CD44+CD- 133- counterparts. A primary culture of brain cancer cells and isolation of CD133+ CSCs might be used as a patient-specific, biology-driven tool to predict the outcome at diagnosis and to study the mechanism of the brain tumor.

Little is known to date about neural development of pig and directed differentiation of porcine pluripotent stem cells (PSCs) to neuronal cells remains elusive. To determine whether soluble factors from glioblastoma multiforme (GBM) promoted the neural differentiation from porcine induced PSCs (iPSCs), cells were treated cultured media of GBM cells. First of all, we isolated and established primary GBM cell line (WHO grade IV). The cellular morphology of GBM cancer cell line are dendritic-like with positive expression in NESTIN, SOX2, VIMENTIN and GFAP using immunofluorescence analysis. G-banded karyotype from primary GBM cell line revealed severe numerical chromosomal aberrations. GBM-cultured medium (CM) treated iPSC-NPCs survive well in vitro when supplemented with a combination of growth factors, including EGF and bFGF. The GBM-CM treated differentiated cells showed an increased mRNA expression level of astrocyte marker, GFAP and the dopaminergic neuron marker, tyrosine hydroxylase (TH). However, there was no significant difference in mRNA expression level of oligodendrocyte marker, MBP. The protocol developed in the present study for large animal models might provide an exciting tool to bridge the present gaps in neuroscience studies between rodents and humans.

Volatile components of crude fucoidan and its raw material, Undaria pinnatifida sporophylls were identified by GC/MSD analysis, and the effects of a combined method of solvent extraction and drying (hot-air drying or spray drying) on deordorization of fucoidans were examined. The major components of seaweed smell (odor) in crude fucoidan and Undaria pinnatifida sporophylls were identified as alkanes (octadecane, heptadecane, tetradecane) and acids (hexadecanoic acid, oleic acid). Solvent extraction and drying were combined for the deordorization of fucoidans: hot-air drying/deordorization process (HDDP) and spray drying/deordorization process (SDDP). The deordorization effects of solvent extraction using solvents including water, ethanol, and acetone were investigated. Among the tested solvents, ethanol was selected as a deordorization solvent, because the seaweed smell and taste were mostly lowered by ethanol extraction followed by drying. In addition, HDDP and SDDP were compared, showing that SDDP gave higher sensory characteristics than HDDP. When the crude fucoidan was treated by ethanol extraction for 30 min followed by SDDP, approximately 60% of off-flavor components of crude fucoidan was removed based on GC/MSD peak area, and the total sugar and sulfate contents and APTT-anticoagulant activity significantly increased compared to crude fucoidan. In conclusion, SDDP after ethanol treatment was proposed and established as a deordorization process for fucoidan preparation.
