      • Expression of Factors Involved Embryo Quality is Correlated Genes in Endometrial Epithelial Cells with Different Types of Bovine Embryos

        In mammals, the uterus is the organ of pregnancy. In most mammals, conceptus implantation to the uterine endometrium consists of blastocyst hatching, migration, apposition/ attachment, invasion and subsequent placental formation. It is known that close to 50% of fertilized pre-implantation embryos in mammals, including humans, fail to implant. We hypothesized that there was the embryo quality-dependent expression of embryo development associated genes on Endometrial Epithelial Cells (EECs) of superovulated Hanwoo. Therefore examined to study the mode of EECs expression during pre-implantation. Additionally, we investigated how EECs respond to the presence of embryos by FSH and LH in bovine EECs in vitro(The EECs were divided into three different groups as follows good (n=4), fair (n=4), bad (n=2)). Our results found that the different expression of EST mRNA to each the group. In addition, it was found that the expression of genes related to MMP-9 and development showed different appearance in each group, and the result of embryo development was different. In particular, the expression of genes associated with the growth of cells from the Good group was very high, and the expression of genes affecting cell activity was also very high. However, Bad group had a high expression of Casp-3 protein and expression of metabolism-related genes of other cells was very low. Also, it is interesting that the expression of FSH and LH in the EEC of Good group is very high, and even in the evaluation of the development rate of in vitro fertilized egg using co-curture, the expression of genes related to cellular metabolic processes increases It was confirmed. These results suggest that FSH and LH and cellular action increase the metabolism of EEC and also increase the development of Embryo.

        Effect of Embryo Transfer Seven Days after Artificial Insemination with Sexed and Conventional Semen from Superovulated Cattle

        Sexed sperm can contribute to increase the profitability of the cow industry through the production of offspring of the craved sex, such as males for meat or females for dairy production. Therefore, the utilization of sexed sperms plays a very important role in the production of offspring of superior cattle. In this study, we examined the pregnancy rates and calves sexing proportion of male and female calves produced using AI, both performed using sexed and conventional sperm. In the result, the conception rates after ET were 73.3% (33/45) sexed semen and 52% (55/104) conventional semen. Thus, the sex ratio for sexed-semen inseminations was 70% (21/30) females for singleton births within a 272 to 292 day gestation interval. The sex ratio for conventional semen was 61% (34/56) females for births. As a result, it is suggested that the use of sex classification sperm will play a very important role in the offspring production of Korean bovine.

      • Expression Pattern of Matrix Metalloproteinase and Their Inhibitors Genes of Bovine Endometrial Epithelial Cells in Hanwoo

        Matrix metallopeptidases (MMP) contribute to spatial and temporal matrix remodeling in the bovine endometrium. To test the this study that MMP expression in bovine Endometrial Epithelial Cells (bEECs) is regulated by implantation and embryo development in in vivo. Uterine bEECs impacts in structure and function in preparation for blastocyst implantation. We investigated the analysis of pre-implantation embryo development and quality on day 7 of gestation on the mRNA expression of MMPs and TIMPs in bEECs. bEECs and embryos were recovered 7 days after the first insemination by flushing the uterus with bEECs and Embryo Collection Medium. Next, we divided the bEECs sample into four groups: good (n=4), poor (n=5), low (n=3) and bad (n=2) samples and compared the group differences in MMP-2, and TIMP-2, TIMP-3 and their correlations within each group. The good group of samples: a total of 111 embryos were collected at the zona (n=5), non-fertilization (n=13), 2-cell (n=1), morula (n=7), compact morula (74) and early blastocyst (n=11). The poor group of samples: a total of 84 embryos were collected at the zona (n=3), non-fertilization (n=47), 4-cell (n=2), 8-cell (n=6), 16-cell (n=3), morula (n=2), compact morula (20) and early blastocyst (n=1). This low group of samples: a total of 10 embryos were collected at the zona (n=1), non-fertilization (n=1), 4-cell (n=2), 8-cell (n=1), 16-cell (n=2) and compact morula (n=3). The embryos collected not occur at bad group samples. To showed the average number of bEECs for the four groups, 5.6×10⁴ in the good group, 4.2×10⁴ in the poor group, 1×105 in the low group, and 8.0×10⁴ in the bad group of bEECs. The research was to examine MMP-2, TIMP-2, and TIMP-3 expression in the bovine EECs throughout gestation. The results imply that TIMP-2 is involved in very early local maternal recognition of pregnancy. In collected samples, the number of bEECs different behaviors of MMP-2, TIMP-2 and TIMP-3. Interestingly, active MMP2 in the endometrium may determine the allocation of growth factors supporting conceptus development. The presence of a day 7 conceptus in vivo and good samples mostly high expression than in another group samples endometrial TIMP2 mRNA expression. MMP are likely to participate in remodeling processes preparing a receptive endometrium for a timely and precise regulation of embryo development. These results suggest the bEECs regulates MMPs in the endometrium under progesterone-rich conditions and may thereby modulate bovine endometrial cell functions during gestation.

      • Effect of Sex-sorted Semen in Superovulated Hanwoo

        Sex-sorted sperm is now widely used in cattle breeding to enhance industrial competitiveness and achieve high female fertility. We bought a "Whole-mom" from the Nuri Science Co., Ltd company. The objective of this study was to evaluate the pregnancy rates and calves sexing proportion of male and female calves produced using Artificial insemination (AI), both performed using sexed and conventional sperm. The study was carried out spring to autumn season on 150 Hanwoo bred on a farm. In all superovulated donors, inseminations were initiated 12 hours after the onset of standing estrus. In a experiment, to take a sexed semen was mixed with "Whole mom" and conventional semen in a water bath at 37℃ for 20 minutes. The sexed and nonsexed semen was deposited in the body of the uterus. After collection, embryo morphology was assessed under a stereomicroscope. Embryos were transferred to the uterine horn ipsilateral to the CL. The conception rates were 73.3% (45/33) sexed semen and 48.8% (90/44) conventional semen. The sex ratio for sexed-semen inseminations was 70% (21/30) females for singleton births within a 279.2 to 282.1 day gestation interval. The sex ratio for unsorted semen was 56.8% (25/44) females for births. The use of X-sorted sperm, already widely employed for AI in dairy cattle has the potential to alter the structure of the industry by increasing the replacement heifer supply, creating opportunities for using a proportion of the dairy herd for producing beef animals, and improving the rate of genetic selection.

        Embryo transfer of dorper breed to Mongolian sheep

        The sheep can be reproduced by natural mating as well as applied reproductive biotechnology, embryo transfer (ET). However, this method in sheep is influenced by several factors such as season, photoperiod, latitude, temperature, nutrition, and breed. In addition, there is still less research on assisted reproductive technologies in small ruminants, compared to other livestock species such as cattle and pigs. Because there has been a need for an optimization and a continuous improvement of ET techniques in small ruminants. the main objective of this study was to evaluate the conception rate obtained after ET in Mongolian sheep (Dorper breed). After embryo recover, code 1 and 2 embryos (morula or blastocyst stage) for ET in the present study were 63% (63/100) and 24% (24/100), respectively. Then Each single embryo was transferred to a synchronized recipient who prepared by estrous synchronization protocol with fluorogestone acetate-cloprostenol sodium. The results demonstrated that an average conception rate and lambing rate was 35.6% (31/87) and 33.3% (29/87), respectively. Further study is still necessary, but these results indicated that single embryo of Mongolian sheep with the present protocol was enough to conducting ET when the genetically superior sheep were necessary to be expanded.
