        A Synthetic Naringenin Derivative, 5-Hydroxy-7,4′-diacetyloxyflavanone-<i>N</i>-phenyl Hydrazone (N101-43), Induces Apoptosis through Up-regulation of Fas/FasL Expression and Inhibition of PI3K/Akt Signaling Pathways in Non-Small-Cell Lung Cancer Cells

        <P>Naringenin, a well-known naturally occurring flavonone, demonstrates cytotoxicity in a variety of human cancer cell lines; its inhibitory effects on tumor growth have spurred interest in its therapeutic application. In this study, naringenin was derivatized to produce more effective small-molecule inhibitors of cancer cell proliferation, and the anticancer effects of its derivative, 5-hydroxy-7,4′-diacetyloxyflavanone-<I>N</I>-phenyl hydrazone (N101-43), in non-small-cell lung cancer (NSCLC) cell lines NCI-H460, A549, and NCI-H1299 were investigated. Naringenin itself possesses no cytotoxicity against lung cancer cells. In contrast, N101-43 inhibits proliferation of both NCI-H460 and A549 cell lines; this capacity is lost in p53-lacking NCI-H1299 cells. N101-43 induces apoptosis via sub-G<SUB>1</SUB> cell-cycle arrest in NCI-H460 and via G<SUB>0</SUB>/G<SUB>1</SUB> arrest in A549 cells. Expression of apoptosis and cell-cycle regulatory factors is altered: Cyclins A and D1 and phospho-pRb are down-regulated, but expression of CDK inhibitors such as p21, p27, and p53 is enhanced by N101-43 treatment; N101-43 also increases expression levels of the extrinsic death receptor Fas and its binding partner FasL. Furthermore, N101-43 treatment diminishes levels of cell survival factors such as PI3K and p-Akt dose-dependently, and N101-43 additionally induces cleavage of the pro-apoptotic factors caspase-3, caspase-8, and poly ADP-ribose polymerase (PARP). Cumulatively, these investigations show that the naringenin derivative N101-43 induces apoptosis via up-regulation of Fas/FasL expression, activation of caspase cascades, and inhibition of PI3K/Akt survival signaling pathways in NCI-H460 and A549 cells. In conclusion, these data indicate that N101-43 may have potential as an anticancer agent in NSCLC.</P>

        Sodium butyrate has context-dependent actions on dipeptidyl peptidase-4 and other metabolic parameters

        Sodium butyrate (SB) has various metabolic actions. However, its effect on dipeptidyl peptidase 4 (DPP-4) needs to be studied further. We aimed to evaluate the metabolic actions of SB, considering its physiologically relevant concentration. We evaluated the effect of SB on regulation of DPP-4 and its other metabolic actions, both in vitro (HepG2 cells and mouse mesangial cells) and in vivo (high fat diet [HFD]-induced obese mice). Ten-week HFD-induced obese C57BL/6J mice were subjected to SB treatment by adding SB to HFD which was maintained for an additional 16 weeks. In HepG2 cells, SB suppressed DPP-4 activity and expression at sub-molar concentrations, whereas it increased DPP-4 activity at a concentration of $1,000{\mu}M$. In HFD-induced obese mice, SB decreased blood glucose, serum levels of insulin and $IL-1{\beta}$, and DPP-4 activity, and suppressed the increase in body weight. On the contrary, various tissues including liver, kidney, and peripheral blood cells showed variable responses of DPP-4 to SB. Especially in the kidney, although DPP-4 activity was decreased by SB in HFD-induced obese mice, it caused an increase in mRNA expression of $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$. The pro-inflammatory actions of SB in the kidney of HFD-induced obese mice were recapitulated by cultured mesangial cell experiments, in which SB stimulated the secretion of several cytokines from cells. Our results showed that SB has differential actions according to its treatment dose and the type of cells and tissues. Thus, further studies are required to evaluate its therapeutic relevance in metabolic diseases including diabetes and obesity.

        2,4-bis (p-hydroxyphenyl)-2-butenal (HPB242) induces apoptosis via modulating E7 expression and inhibition of PI3K/Akt pathway in SiHa human cervical cancer cells.

        <P>The Maillard reaction is a chemical reaction occurring between an amino acid and a reducing sugar, usually requiring thermal processing. Maillard reaction products (MRPs) have antioxidant, antimutagenic, and antibacterial effects, and although 2,4-bis (p-hydroxyphenyl)-2-butenal (HPB242), a fructose-tyrosine MRP, appears to inhibit proliferation of cancer cells, its mechanism of action has not been studied in detail. We found that HPB242 treatment modulated expression of cyclins and tumor suppressor genes in SiHa human cervical cancer cell lines: cyclins and phospho-pRB were downregulated, whereas the expression of CDK inhibitors and p53 was enhanced. HPB242 induced apoptosis dose-dependently by suppressing E7 expression and leading to sub-G1 cell-cycle arrest in SiHa cell lines; treatment also led to the proteolytic cleavage of caspase-3, -9, and poly (ADP-ribose) polymerase. Moreover, HPB242 upregulated Fas expression, altered expressions of pro- and antiapoptotic factors, and also inhibited nuclear translocation of nuclear factor κB and phosphorylation of IκB. HPB242 treatment decreased phosphatidyl inositol-3 kinase and p-Akt expression levels, demonstrating that this survival pathway may also be inhibited by HPB242. Cumulatively, HPB242 promotes apoptosis by influencing E7 expression, inducing cell-cycle arrest at sub-G1 phase, and promoting both intrinsic (mitochondrial) and extrinsic (Fas-dependent) apoptosis in SiHa human cervical cancer cells.</P>

      • High-throughput screening of protein variations in soybean seeds.

        The protein in soybean seeds accounts for approximately 40% of the dry seed weight. Two major storage proteins, 7S and 11S, constitute 70-80% of the total storage proteins in the seeds. In this study, the variation of total soluble protein extracts from 1152 soybean landraces that have been collected from South Korea were studied using high-throughput screening method with HT Protein Express Labchip (Caliper Life Sciences, Inc.). Seven distinct protein band patterns - four protein sub-units of 11S and three sub-units of 7S, were taken into account and their presence or absence were analyzed. Among the 1152 landraces, 525 genotypes were identified as lacking lipoxygenase, 255 lacking α1 subunit, 680 lacking α subunit, 169 lacking β subunit, 140 lacking acidic, 114 lacking Kunitz Trypsin Inhibitor (KTi) and 199 lacking basic protein patterns. The high-throughput protein analysis is helpful in screening a large number of populations with less time and minimum labor. The selected genotypes with low amounts or lacking of anti-nutritional factors such as trypsin inhibitor, lipoxygenase and α subunit would be used for future breeding purpose of quality improvement in soybean protein.

      • 탄화규소 세라믹 필터를 이용한 여과기술에 관한 연구

        Candle type SiC filters applicable to hot gas filtration of IGCC and PFBC were manufactured and tested using two different dusts collected from cokes and water glass manufacturing processes. Particle sizes, porosities, and mean pore sizes of manufactured SiC filters range 150∼300㎛, ∼40%, and 43∼83㎛, respectively. Collection efficiencies of two different types of dusts rage 99.09∼99.95% at 600℃ and latm test condition. At the same condition, Darcy's resistance coefficients rage 1.149×10^10∼3.514×10^11m^-2 and differential pressures range 187∼391㎜H₂O. Also it was found that the smaller the mean pore size, the higher the differential pressure at the same operating conditions. SiC filters have high potential as a fine dust removal medium from HTHP processes like PFBC and IGCC. Furthermore, long-term performance tests of the filters are necessary for the commercialization of SiC filters.

        Many European institutions and companies are developing ceramic filters which can substitute particulates and gas removal devices currently in use and which are believed to be the control devices of next generation. Because of their resistance to both chemical and thermal shocks, Ceramic filter system alone can remove particulates and gaseous air pollutants simultaneously without compulsory cooling systems, thus reducing installation and operating cost significantly. In this research, there is developed and tested, which is catalyst impregnated ceramic filters applicable to simultaneous removal of particulates and gaseous air pollutants. Emission standards of air pollutants are getting stringent and the market share of air pollutants control devices in Korea is projected up to 800 billion won by the year 2002 and low density ceramic filter system can substitute electrostatic precipitators and conventional bag filter systems significantly. catalyst doped ceramic filter were believed to be the most promising technology which can substitute particulates and gaseous pollutants removal devices in the near future.

      • Alumina 첨가에 따르는 Hydroxyapatite의 기계적 성질

        For the purpose of improving mechanical strength of hydroxyaptite, various amount of alumina powder were added to hydroxyaptite which were prepared by precipitation method using Ca(NO₃)₂4H₂O and (NH₄)₂HPO₄under the Ca/P mole ratio 1.67 and pH 11. Hydroxyaptite-alumina powder composite were sintered in H₂O gas atmosphere at 1150-1400℃. Adding alumina, hydroxyaptite was decomposed to β-tricalcium phosphate and co-existed with hydroxyaptite. In case of added alumina, bending strength was higher than that of hydroxyaptite. Maximum bending strength was 161MPa at 1300℃, 5.0wt% alumina. Added alumina restrained grain growth of the hydroxyaptite.

        In this paper, in order to obtain an excellent transparent conducting thin film with low resistivity and high optical transmittance for Plasma Display Panel (PDP), aluminum doped zinc oxide films were deposited on Corning glass substrate by rf magnetron sputtering method. The volume resistivity of the film was strongly influenced by the Ar pressure, substrate temperature, rf discharge power and Al doping contents. The lowest volume resistivity of 7×10^(-4) Ω-cm, associated with a transmittance of 89 % were obtained under the conditions of lmtorr and 400℃ in argon pressure and substrate temperature, respectively. The diffraction peaks of the film also became more intense and narrower with decreasing Ar pressure and increasing substrate temperature up to 400 ℃.

      • 저밀도 세라믹 필터의 특성 및 내구성에 관한 연구

        This study was Designed to examine the applicability of low density ceramic filters to actual processes through long term experiment to reveal the characteristics ceramic filter, durability against thermal and mechanical shocks. The experiment was also conducted at 600℃ and 300℃. 1m long low density ceramic filter with 60㎜ outer diameter and 10㎜ thickness was tested with dust collected from cokes process of P-steel. Collection efficiency ranged 99.98∼99.99% and conditioning was completed after 150∼200hrs. Resistance coefficients of filters range 5.9×10^10 at 600℃, 1.2×10^10 at 300℃. Long term experiments show that conditioning was completed after 155 hours of test at 300℃ and 165 hours at 600℃ with high collections efficiencies. Also tested filters show mechanical stabilities at different particulate feed rates ranging 0.5g/min to 1.0g/min. Results show that tested filters can be applied to commercial processes with one year guarantee.
