Random amplified polymorphic DNA (RAPD) markers were used to survey genetic variability among 34 Botrytis cinerea isolates from nine different host plants in Korea. For RAPD analysis, 115 arbitrary decamer primers were initially screened for polymorphic major DNA bands with 11 representative B. cinerea isolates. Eleven primers that initially detected polymorphisms were tested a second time with additional 23 isolates of B. cinerea as well as one isolate of Botrytis squamosa as an outgroup. The RAPD analyses revealed that all isolates except one showed different molecular phenotypes. Dendrograms obtained from dissimilarity matrices using the unweighted paired group method of arithmetic means (UPGMA) showed the 36.4% to 90.0% similarity among all B. cinerea isolates. The B. squamosa isolate showed the least similarity to all B. cinerea isolates. The cluster analyses indicated no correlation among all the characteristics examined including molecular phenotypes, host and geographic origins, year of isolation, or pathogenicity. The RAPD data suggest that a high level of genetic variation exists among Korean populations of B. cinerea and it seems to be caused by heterokaryosis among preexisting molecular phenotypes.
Radon Exposure-induced Genetic Variations in Lung Cancers among Never Smokers

<P><B>Background</B></P><P>Lung cancer in never smokers (LCINS) differs etiologically and clinically from lung cancer attributed to smoking. After smoking, radon exposure is the second leading cause and the primary risk factor of lung cancer among never smokers. Exposure to radon can lead to genetic and epigenetic alterations in tumor genomes affecting genes and pathways involved in lung cancer development. The present study sought to explore genetic alterations associated with LCINS exposed to radon gas indoors.</P><P><B>Methods</B></P><P>Genetic associations were assessed via a case-control study of LCINS (39 cases and 30 controls) using next generation sequencing. Associations between genetic mutations and high exposure to radon were investigated by OncoPrint and heatmap graphs. Bioinformatic analysis was conducted using various tools. According radon exposure levels, we divided subjects in two groups of cases and controls.</P><P><B>Results</B></P><P>We found that ABL2 rs117218074, SMARCA4 rs2288845, PIK3R2 rs142933317, MAPK1 rs1803545, and androgen receptor (AR) rs66766400 were associated with LCINS exposed to high radon levels. Among these, Chromodomain helicase DNA-binding protein 4 (CHD4) rs74790047, TSC2 rs2121870, and AR rs66766408 were identified as common exonic mutations in both lung cancer patients and normal individuals exposed to high levels of radon indoor.</P><P><B>Conclusion</B></P><P>We identified that CHD4 rs74790047, TSC2 rs2121870, and AR rs66766408 are found to be common exonic mutations in both lung cancer patients and normal individuals exposed to radon indoors. Further analysis is needed to determine whether these genes are completely responsible for LCINS exposed to residential radon.</P>

<P>The herb Liriope platyphylla (LP) has been considered to have curative properties for diabetes, asthma and neurodegenerative disorders. To examine the effects of steaming time and frequency of manufactured red LP (RLP) on the nerve growth factor (NGF) secretion ability and NGF receptor signaling pathway, the NGF concentration, cell differentiation, NGF signaling pathway and calcium concentration were analyzed in neuronal cells treated with several types of LPs manufactured under different conditions. The maximum NGF secretion was observed in B35 cells treated with 50 ?g/ml LP extract steamed for 9 h (9-SLP) and with two repeated steps (3 h steaming and 24 h air-dried) carried out 7 times (7-SALP). No significant changes in viability were detected in any of the cells treated with the various LPs, with the exception of 0-SLP and 0-SALP. In addition, PC12 cell differentiation was induced by treatment with the NGF-containing conditional medium (CM) collected from the RLP-treated cells. The levels of TrkA and extracellular signal-regulated kinase (ERK) phosphorylation in the high affinity NGF receptor signaling pathway were significantly higher in the cells treated with 3-SLP or 1-SALP/3-SALP CM compared with those treated with the vehicle CM. In the low affinity NGF receptor pathway, the expression levels of most components were higher in the 9-, 15- and 24-SALP CM-treated cells compared with the vehicle CM-treated cells. However, this level was significantly altered in cells treated with 3-SALP CM. Furthermore, an examination of the RLP function on calcium regulation revealed that only the LP- or RLP-treated cells exhibited changes in intracellular and extracellular calcium levels. RLP induced a significant decrease in the intracellular calcium levels and an increase in the extracellular calcium levels. These results suggest the possibility that steaming-processed LP may aid in the relief of neurodegenerative diseases through the NGF secretion ability and NGF signaling pathway.</P>

The paper manufactories in Sangpyeong Industrial Estates, Chinju, produce more than 80 tons of paper-mill sludge cakes every day, which costs about 840 million won for dump per year. Since the paper-mill sludge is biodegradable, the sludge can be utilized as an organic fertilizer if it is properly decomposed. This may lead to not only dramatic cut of the treatment cost but prevention from secondary environmental contamination due to dump. The primary objective of the study was to explore the quantitative range of major environmental parameters influencing early composting of paper-mill cakes, naturally pretreated in warn and cold weather. The automatically-controlled reactor was designed, manufactured, and operated for nine days to decompose about 220l of raw sludge cakes. Four tests(Test1~Test4) were implemented for the study of Phase I. Treatments of two levels of initial temperature(40℃ and 28℃) and two levels of water content+C/N ratio(35%+40 and 63%+80) were made to test the significance of their parameters for decomposition of raw sludge cakes pretreated in warm weather. Another four tests(Test5~Test8) were implemented for the Study of Phase II. Treatments of initial temperature and water content(W/C)+C/N ratio of raw sludge pretreated in cold weather were made to 16℃ and 13℃+58for Test5, 6℃ and 53%+55 for Test6,7℃ and 36%+81 for Test7, 31℃ and 30% +81 for Test8. Natural weater condition(pretreatment condition) revealed the importance in composting of the paper-mill sludge cakes. Combination of water content adjustment to about 30% with C/N ratio amendment of about 20 and initial temperature of 30~40℃ was concluded to be the best for early composting of paper-mill sludge cakes with aeration rate and pH fixed. Temperature and C/N ratio were adapted as judging variables for composting degree. In addition tests for microbial activity were performed to validate the experimental results. Since the tempera-ture and C/N ratio did not coincide in some tests as judging variables for the maturity of the composting sludges, taking one of these parameters could mislead the concept of the maturity (composting conceptually new criterion to provide more reliable information for early composting of paper-mill sludge cakes.
