New Insight for Fluoroquinophenoxazine Derivatives as Possibly New Potent Topoisomerase Ⅰ Inhibitor

Fluoroquinolones, represented by ciproxacin and norfloxacin, are well known clinical antimicrobial agents, and their phenyl ring expanded quinophenoxazines are reported as possible antitumor active compounds. These quinophenoxazines are known to inhibit DNA topoisomerase II essential for cell replication cycle. But there were no reports for topoisomerase I inhibition study for these compounds. In this report, we have prepared a few quinophenoxazine analogues and tested their topoisomerases I and II inhibitory activities and cytotoxicity. From the result, we found that qumophenoxazine analogues possessed strong topoisomerase I inhibitory capacity as well as topoisomerase II inhibition. Among the compounds prepared, A-62176 analogues showed strong topoisomerases I and II inhibitory activities. Interestingly, compound 8 missing the 3-aminopyrrolidine moiety at C2 position has similar potent inhibitory capacity against topoisomerases I & II at higher concentrations (20 and 10 uM, respectively). But compound 8 inhibited topoisomerase I function more selectively at lower concentration, 2 uM. Our observation mi&ht strongly implicate that fluoroquinophenoxazines can be developed as efficient topoisomerase I inhibitor with the elaborate modification.

The purpose of this study was to investigate the difference of nutritional and growth status for two preschool children with different socio-economic status. This study was conducted from June 10, 1984 to July 12, 1984. The information about general socio-economic status was obtained by survey questionnarre. Three day food recording method was utilized to assess food consumption and measured to estimate the difference of growth rate between the two groups. Hemoglobin and Hematocrit levels were also measured to assess the prevalence of anemia. According to these results. 1. Higher Educatonal status of mother and monthly income of each household were observed in Y, but higher Engel's coefficient was obtained in M. 2. Eventhough average nutrient intake was adequate compared to RDA for most of the preschool children in Y, inadquate intake of Ca, Fe, and Vit B2 were observed for 11.9%, 1.7%, and 2% of the preschool children, respectively. All the nutrients in take were observed below RDA in M expect Vit A, Vit B1 and Niacin. 42%, 18.7% and 10.4% of preschool children exhibited inadquate intake of Ca, Fe and Vit B2, respectively. 3. The proportion of preschool children with weight, height and skinfold thickness below standard in Y was significantly lower than those of preschool children in M(P<0.01). 4. Hemoglobin and hematocrit levels were positively correlated to the Calorie, protein, Fe and Vit. C intake in both area. It is suggestion nutritional education program in two areas.
Brightener breakdown at the insoluble anode by active chlorine species during Cu electrodeposition

It is highly important to design and develop appropriate insoluble anodes for industrial Cu electroplatingto lower the amount of organic additives that this process consumes. Conventionally, this rapid consumptionof additives (e.g., brighteners, suppressors, or levelers) is known to be due to the active radical species(e.g., OH) formed during oxygen evolution. In this study, we found that trace amounts of chlorideions present in the electroplating bath are the source of the active chlorine species that accelerate thebreakdown at the insoluble anode. A sacrificial, perfluorinated polymer coating effectively decreasedthe emission of active chlorine species from the electrode, thereby lowering the consumption of thebrightener. This study reveals that the suppression of chlorine evolution at the anode can be an effectiveapproach for decreasing brightener consumption during Cu electrodeposition.

In order for a novel test method to be applied for regulatory purposes, its reliability and relevance, I.e., reproducibility and predictive capacity, must be demonstrated, Here, we examine the predictive capacity of a novel non-radioisotopic local lymph node assay, LLNA:BrdU-FCM (5-bromo-2`-deoxyuridine-flow cytome-try), with a cutoff approach and inferential statistics as a prediction model. 22 reference substances in OECDTG429 were tested with a concurrent positive control. Hexylcinnamaldehyde 25%(PC), and the stimulation index(SI) representing the fold increase in lymph node cells over the vehicle control was obtained. The optimal cutoff SI(2.7≤cutoff<3.5), with respect to predictive capacity, was obtained by a receiver operating character-istic curve, which produced 90.9% accuracy for the 22 substances. To address the inter-test variability in respon-siveness, SI values standardized with PC were employed to obtain the optimal percentage cutoff(42.6≤Cutoff<57.3%of PC), which produced 86.4% accuracy. A test subastance may be diagnosed as a sensitizer if a statistically significant increase in Si is elicited. The parametric one-sided t-test and non-parametric Wilcoxon rank-sum test produced 77.3% accuracy. Similarly, a test substance could be defined as a sensitizer if the SI means of the vehicle control, and of the low, middle, and high concentrations were statistically significantly different, which was test-ed using ANOVA or Kruskal-Wallis, with post hoc analysis, Dunnett, or DSCF(Dwass-Steel-Critchlow-Fligner), respectively, depending on the equal variance test, producing 81.8% accuracy. The absolute SI-based cutoff approach produced the best predictive capacity, however the discordant decisions between prediction models need to be examined further.

Purpose: Postoperative hypocalcemia is one of the most common complications after thyroid surgery. However, the predictive efficacy of parathyroid hormone (PTH) level was still controversial. We investigated the cut-off value of perioperative PTH level for postoperative hypocalcemia after total thyroidectomy. Methods: Four hundred eighty-two patients performed total thyroidectomy from January 2015 to December 2016 were enrolled. Demographics, operative variables and the development of symptomatic hypocalcemia were collected and evaluated. Results: Transient symptomatic hypocalcemia occurred in 83 (17.2%) patients and 5 (1.0%) patients have progressed to the permanent hypocalcemia. Patients with transient symptomatic hypocalcemia showed significantly lower postoperative day 1 PTH level than those without hypocalcemia (7.7±6.5 vs. 13.1±10.4 pg/mL; P<0.001), while correlation between postoperative PTH level and development of symptomatic hypocalcemia was not found in a multivariate analysis (P=0.122). Percentage decline of the preoperative PTH only showed significant association with transient hypocalcemia and the recommended cut-off value was 75.0%. When percentage decline of 75.0% was used, the incidence of transient hypoparathyroidism was 3.4 times (95% confidence interval, 2.1-5.6) higher in high percentage decline group (P<0.001). In regard to the permanent hypoparathyroidism, there were no significant differences in all factors. Conclusion: Percentage decline of PTH can be used as a predictive factor for postoperative hypocalcemia. Cautions against preventing injury of the parathyroid glands with careful surgical technique should be recommended.

Objective The purpose of this study was to evaluate the group B streptococcus (GBS) colonization rate in pregnant Korean women using selective culture media for GBS and to identify obstetrical complications and GBS-induced early-onset neonatal sepsis. Methods We evaluated 1,014 pregnant women who delivered at Busan Paik Hospital between January 2015 and December 2016. GBS colonization was assessed using chromID Strepto B agar. We evaluated GBS colonization in pregnant women, as well as the obstetrical complication and GBS-induced neonatal sepsis rates. Results The total GBS colonization rate was 11.6% (117/1,014). No significant increase was observed in the rate of pregnancy-related complications between the GBS-positive and the GBS-negative groups. Among the 134 neonates born to colonized mothers, early neonatal sepsis was reported in 2 neonates (1.5%); however, these were cases of non-GBS-induced sepsis. Conclusion The GBS colonization rate (using selective culture media) in this study involving pregnant Korean women showed a higher colonization rate than that previously reported in Korea. Therefore, based on this study, we recommend GBS screening and the administration of intrapartum antibiotic prophylaxis in pregnant Korean women.
