        The effects of fatty acids related with elicitation mechanism were investigated in cell suspension cultures of Catharanthus roseus. The specific growth rate in control culture was 0,17 to 2.0 day-¹ and the cell doubling time(td) showed 3.5 to 4 days. The highest specific growth rate was obtained when the inoculum was added at 4g FCW, compared with 2 and 3 g FCW inoculum. The suspension cells were cultured at various concentrations of fatty acids and fatty acid was added at different culture time. The addition of methyl jasmonate among fatty acids only showed the slight promoting effect on cell growth but others did not show any increasing effect. The best injection period of fatty acid to promote the cell growth was the inoculation time.

      • 미세조류 Dunaliella bardawil을 이용한 β-carotene 생산조건에 관한 연구

        Low-growth rates favor β-carotene accumulation under conditions of stress, light intensity and wave length, oxygen free radicals, and high salinity. It was found from shake flask experiments that high density of algal cultures can be increased by the following environmental condition : white fluorescent light compared with blue and red light, addition five times concentrated medium, mixture of 104㎖/day CO_2 gas and 250mM sodium bicarbonate, and pH 8.0. The periodic feeding of 7mM nitrate ion under the nitrogen limitation condition enhanced the cell concentration up to 9 × 10 exp (6) cells/㎖ compared with 1.5 × 10 exp (6) cells/㎖ of the control culture. An photobioreactor system was successfully for algal growth and resulted in max cell density of 4.2 × 10 exp (6) cells/㎖ in comparison with that of batch flask culture(generally max cell density of batch flask culture is about 3.0 × 10 exp (6) cells/㎖). The following induction conditions such as change of salinity(NaCl), oxygen free radicals1 (H_2O_2), and blue light favored the massive accumulation of β -carotene. The change of culture conditions from a phase of favorable high cell density to that of optimum induction factors resulted in increased productivity.

      • Lignin Peroxidase 에 의한 Azo계 염료 분해

        Lignin peroxidase produced by white rot fungi Phanerochaete chrysosporium, can degrade aromatic compound such a lignin. In this study, the purified lignin peroxidase cultured from P. chrysosporium and commercial HrP as a control was used for determining the degradation ability a of azo dyes. In addition, the effect of immobilization matrix, pH, temperature, dye concentration and type have been studied. As a result, the order of azo dye degradation rate using HrP and LiP was, Congo red, Acid Yellow 9, Orange Ⅱ. LiP showed the highest degradation percentage at 35℃ if among tested reaction temperatures. LiP under the acidic condition was most activated and resulted in the highest degradation rate of azo dyes tested. However, LiP and HrP was unstable for pH 2.0 to 2.5. From the comparison of HrP and LiP for the degradation rate of azo dyes, LiP resulted in more degradation ability than HrP. The comparison of the former martial is more expensive than the latter best the degradation rate and immobilization percentage of both immobilization methods are the same.
