High-performance liquid chromatography method development for the quality control of Ginkgonis Semen

<P>Ginkgonis Semen (GS) is the seed of Ginkgo biloba Linne and a valuable material for herbal medicines and functional foods in China, Japan and Korea. The main bio-compounds of GS are GA, GB and GC like the leaves. There are many studies for the analysis of ginkgolides in the leaves or leaves extract of G. biloba because the leaves extract is a valuable material in pharmaceutical industries. However, there is no efficient analytical method for the quality control of GS based on the quantitation of ginkgolides because of matrix effect induced by different chemical composition. So, there are no content criteria of GS in Pharmacopoeia of Korea, Japan and China until now. This study aimed to develop HPLC method using ginkgolides based on the quantitation of GA, GB and GC for the quality control of GS with the optimization of sample preparation to enhance the analytical sensitivity and reproducibility. At first, defatting process using petroleum ether and liquid-liquid extraction were applied for sample preparation to remove matrix effect. The HPLC-ELSD method was developed with the mobile phase of a 0.5% aqueous acetic acid and methanol-acetonitrile solution (1: 1 ratio) under gradient conditions. GA, GB and GC contents in GS were different between Korea and China. The mean quantity of Korean samples was 4.85 +/- 2.33 lg/g GA, 48.38 +/- 5.10 lg/g GB, and 37.83 +/- 7.64 lg/g GC. Those contents of Chinese samples were higher than Korean samples as 9.39 +/- 2.51 lg/g GA, 123.59 +/- 26.24 lg/g GB and 53.39 +/- 4.97 lg/g GC. It indicated that the discrimination of GS between Korea and China could be achieved by marker compound contents. Furthermore, the geographical discrimination of GS between Korea and China was confirmed by PCA using the quantitative data of marker compounds. By statistical analysis, the calculated content criteria of GS by regression method were 2.35 lg/g of GA, 29.20 lg/g of GB, and 27.75 lg/g of GC, based on dry weight. Thus, our HPLC method shows potential toward the development of a universal quality control methodology to quantify GS quality and origin. (C) 2016 The Authors. Insulinomas are tumors that, in more than 90% or cases, are both single and benign, so the potential for surgical cure is very high. But, their localization is somewhat difficult, as they are small in size and located deep in pancreatic parenchyma. Because success of operation is depentdent on the localization rather than the surgical techniques, the availability of good licalization technique is mandatory. Recently, endoscopic ultrasonography(EUS) has been reported to be a very sensitive and specific test for the localization of insulinomas. We report a case of insulinoma that was localized successfully by EUS, after failure in localization by abdominal ultrasonography, abdominal CT, selective angiography, and even by transhepatic portal vein sampling. Preparation of 2,2,2-trifluoroethanol from 1,1,1-trifluoroethylchloride with alkali-metals in aprotic polar solvents has been studied. Effects of alkali-metals and solvents on the 2,2,2-trifluoroethanol yield as well as the reaction mechanism were investigated. 2,2,2-trifluoroethanol was produced at high temperatures between 150℃ and 250℃. CH_3COOK produced better yields than KOH, and the co-solvent system of v -butyrolactone and ionic liquid [BMIM]^+BF_4^- produced remarkably improved results.

본 설문조사는 국내 육가공제품의 시장에 대한 소비자 의식과 구매형태를 파악하고 시장확대를 위한 기본방안 구축을 위하여 10월부터 12개월까지 3개월 조사한 결과이다. 육가공제품의 구입횟수는 월 1∼2회로 구입하는 소비자가 40.73%로 가장 많았고 그 다음이 주 1∼2회 구입하는 것(28.07%)으로 나타났다. 햄과 소시지를 구분할 수 있는지에 대한 질문에 구분한다가 86.75%로 나타난 반면 구분하지 못한다가 13.25%로 나타났다. 햄과 소시지를 구분하지 못하는 소비자들은 그 이유로서 두 제품의 고기 함량 및 첨가내용물들이 비슷하여서라는 응답이 47.19%로 가장 많았다. 햄과 소시지를 구입할 때 가장 중요하게 고려하는 사항은 회사명(brand name)인 것으로 나타났으며(32.76%) 원료육의 종류(16.53%), 유통기한(16.38%) 및 원료육 생산지(16.23%)인 것으로 나타났다. 햄 또는 소시지 구입시 제품설명 표시내용을 읽느냐는 질문에 응답자들 중 72.25%가 그렇다고 응답한 반면 27.25%는 읽지 않는다고 응답하였다. 소비자들은 햄과 소시지 구입시 불만사항으로 소비자들이 제품내에 고기보다는 결착제를 비롯한 다른 첨가제들이 많아서 고기맛을 거의 느낄 수 없다고 응답하였으며(47.65%), 외관 및 내용물이 비슷한 제품이 너무 많다고 응답한 소비자 및 맛이 없다고 응답한 소비자가 각각 27.70%, 11.62%인 것으로 나타났다. 햄 및 소시지 제품에 대한 요구사항으로 고기 함량이 높은 제품을 생산하기를 원한다고 응답한 소비자가 64.04%였다. 햄과 소시지 제품 판매시 품질에 따른 가격차등제 실시에 대하여 찬성한다고 응답한 소비자가 83.54%인 것으로 나타나 현재 많은 소비자들이 햄과 소시지 제품의 품질에 따른 가격차등제 실시를 원하는 것으로 나타났다. 우리나라도 품질에 따른 등급 기준 설정과 품질별 가격차등제 실시를 통하여 육가공 제품시장의 확대가 가능할 것으로 판단되며 이는 국내돈육의 비선호부위의 소비촉진도 가능할 것이다. Consumer survey(n=1,343) was carried out to investigate the consumer's perception, purchase behavior and demand on the processed meat products. Most consumers purchased the ham and sausage products once a month (40.73%). Consumers(47.19%) didn't distinguish between ham and sausage products due to the similarity of meat contents and additives in products. Consumers(72.75%) considered the product label when they purchased whereas 27.25% of consumers were not interested in the product label. Consumers(47.65%) were not satisfied with ham and sausage products because there were too much additives other than meat, there were many similar processed products in kinds and appearance(27.70%) and they could hardly detect meat taste(11.62%). Consumers demanded to produce the ham and sausage products which contain high meat contents(64.04%). Consumers were favored with establishment of the price differential system depending on the quality based on meat contents in the product(83.54%). In conclusion, the establishment of the price differential system depending on quality as well as the quality grading system for processed meat products is needed not only to increase the processed meat market but also increase the pork consumption especially for non-preferred pork portions.

<P><B>Abstract</B></P> <P>The dynamic strain aging (DSA) of metallic materials due to solute atom diffusion to mobile dislocations induce deformation instability with load fluctuations and deformation localizations, hence reducing their sheet formability. In this paper, DSA behaviors of twinning induced plasticity (TWIP) steel with and without Al during tensile testing and deep drawing are investigated in terms of strain localization and the Portevin-Le Chatelier (PLC) band. A theoretical DSA model with internal variables of dislocation density and twin volume fraction is presented for an estimation of strain localization and strain hardening behavior of TWIP steels. The simulation results of the load history and PLC bands during tensile testing and deep drawing are in good agreement with the experimental values. A serration behavior is observed in high-Mn TWIP steels and its tensile residual stress is higher than that in the Al-added TWIP steels, which results in a deformation crack or delayed fracture of deep drawn specimens.</P>
Mammalian Systems Biotechnology Reveals Global Cellular Adaptations in a Recombinant CHO Cell Line

Effective development of host cells for therapeutic protein production is hampered by the poor characterization of cellular transfection. Here, we employed a multi-omics-based systems biotechnology approach to elucidate the genotypic and phenotypic differences between a wild-type and recombinant antibody-producing Chinese hamster ovary (CHO) cell line. At the genomic level, we observed extensive rearrangements in specific targeted loci linked to transgene integration sites. Transcriptional re-wiring of DNA damage repair and cellular metabolism in the antibody producer, via changes in gene copy numbers, was also detected. Subsequent integration of transcriptomic data with a genome-scale metabolic model showed a substantial increase in energy metabolism in the antibody producer. Metabolomics, lipidomics, and glycomics analyses revealed an elevation in long-chain lipid species, potentially associated with protein transport and secretion requirements, and a surprising stability of N-glycosylation profiles between both cell lines. Overall, the proposed knowledge-based systems biotechnology framework can further accelerate mammalian cell-line engineering in a targeted manner.
