        EphrinA1/EphA2 Promotes Epithelial Hyperpermeability Involving in Lipopolysaccharide-induced Intestinal Barrier Dysfunction

        Background/Aims Lipopolysaccharide (LPS) is the key factor inducing mucosal and systemic inflammation in various intestinal and parenteral diseases, which could initially disrupt the epithelial barrier function. EphrinA1/ephA2 is speculated to increase the epithelial permeability for its “repulsive interaction” between adjacent cells. This study aim to investigate the role of ephrinA1/ephA2 in LPS-induced epithelial hyperpermeability. Methods In vivo model challenged with oral LPS in C57BL/6 mice and in vitro model exposed to LPS in Caco2 monolayer were established. The barrier function was assessed including expression of tight junction proteins (occludin and claudin-1), transepithelial electrical resistance, and permeability to macromolecules (fluorescein isothiocyanate-labeled fluorescent dextran 4 kDa [FD4]). Moreover, the expression and phosphorylation of ephrinA1/ephA2 were quantified, and its roles in the process of epithelial barrier disruption were confirmed via stimulating ephA2 with ephrinA1-Fc chimera (ephrinA1-Fc) and inactivating ephA2 with ephA2-Fc chimera (ephA2-Fc), or ephA2 monoclonal antibody (ephA2-mab), as well as inhibiting extracellular signal-regulated kinase 1/2 (ERK1/2) with PD98059. Results LPS induced significant barrier dysfunction with dismissed occludin and claudin-1 expression, reduced transepithelial electrical resistance and increased FD4 permeability, accompanied by upregulated ephrinA1/ephA2 pathway and phosphorylation of ephA2 receptor. Furthermore, ephA2-Fc, and ephA2-mab ameliorated LPS-induced epithelial hyperpermeability, which was also inhibited by PD98059. Additionally, ephrinA1-Fc led to apparent epithelial leakage in Caco2 monolayer by promoting the phosphorylation of ERK1/2, which could be obviously blocked by ephA2-mab and PD98059. Conclusion EphrinA1/ephA2 promotes epithelial hyperpermeability with an ERK1/2-dependent pathway, which involves in LPS-induced intestinal barrier dysfunction.         Geometrically non-linear dynamic analysis of plates by an improved finite element-transfer matrix method on a microcomputer

        An improved finite element-transfer matrix method is applied to the transient analysis of plates with large displacement under various excitations. In the present method, the transfer of state vectors from left to right in a combined finite element-transfer matrix method is changed into the transfer of generally incremental stiffness equations of every section from left to right. Furthermore, in this method, the propagation of round-off errors occurring in recursive multiplications of transfer and point matrices is avoided. The Newmark-${\beta}$ method is employed for time integration and the modified Newton-Raphson method for equilibrium iteration in each time step. An ITNONDL-W program based on this method using the IBM-PC/AT microcomputer is developed. Finally numerical examples are presented to demonstrate the accuracy as well as the potential of the proposed method for dynamic large deflection analysis of plates with random boundaries under various excitations.

        Amino-Ended Hyperbranched Polyamide Modified SBA-15 as Support for Highly Efficient Cobalt Fischer-Tropsch Synthesis Catalyst

        Among the Fischer-Tropsch synthesis (FTS) catalysts, Cobalt catalysts are currently attracting a lot of research interests due to their high activity and high selectivity. But the dispersion and reducibility of cobalt catalysts with moderate interaction between cobalt and support are still a challenge. A novel amino-ended hyperbranched polyamide (AEHPA) was used to modify SBA-15 and then was applied as support for obtaining cobalt catalyst (15Co/SBA-15-N). The catalysts were characterized by XRD, TEM, XPS and H2-TPR techniques. The results showed that AEHPA doped mesoporous SBA-15 caused the generation of N species in the SBA-15 pore channels. The N-Co bonds resulted in the formation of highly-dispersed cobalt nanoparticles with uniform sizes inside the ordered mesopores of support. AEHPA doping was an effective way to modify the surface properties of the SBA-15 for immobilizing cobalt nanoparticles. Compared with the conventional 15Co/SBA-15 catalyst without doping AEHPA, the AEHPA doped 15Co/SBA-15-N catalyst showed improved cobalt dispersion and stabilized cobalt location, which led to much better reaction stability as well as C5+ selectivity.

        Inhibition of Tumoral VISTA to Overcome TKI Resistance via Downregulation of the AKT/mTOR and JAK2/STAT5 Pathways in Chronic Myeloid Leukemia

        Tyrosine kinase inhibitors (TKIs) have revolutionized the treatment landscape for chronic myeloid leukemia (CML). However, TKI resistance poses a significant challenge, leading to treatment failure and disease progression. Resistance mechanisms include both BCR::ABL1-dependent and BCR::ABL1-independent pathways. The mechanisms underlying BCR::ABL1 independence remain incompletely understood, with CML cells potentially activating alternative signaling pathways, including the AKT/mTOR and JAK2/STAT5 pathways, to compensate for the loss of BCR::ABL1 kinase activity. This study explored tumoral VISTA (encoded by VSIR) as a contributing factor to TKI resistance in CML patients and identified elevated tumoral VISTA levels as a marker of resistance and poor survival. Through in vitro and in vivo analyses, we demonstrated that VSIR knockdown and the application of NSC-622608, a novel VISTA inhibitor, significantly impeded CML cell proliferation and induced apoptosis by attenuating the AKT/ mTOR and JAK2/STAT5 pathways, which are crucial for CML cell survival independent of BCR::ABL1 kinase activity. Moreover, VSIR overexpression promoted TKI resistance in CML cells. Importantly, the synergistic effect of NSC-622608 with TKIs offers a potent therapeutic avenue against both imatinib-sensitive and imatinib-resistant CML cells, including those harboring the challenging T315I mutation. Our findings highlight the role of tumoral VISTA in mediating TKI resistance in CML, suggesting that inhibition of VISTA, particularly in combination with TKIs, is an innovative approach to enhancing treatment outcomes in CML patients, irrespective of BCR::ABL1 mutation status. This study not only identified a new pathway contributing to TKI resistance but also revealed the possibility of targeting tumoral VISTA as a means of overcoming this significant clinical challenge.

        It is a dream since the ancient time that people can communicate with each other in anytime and anyplace. With the development of networks, new technology and services spring out to meet every kind of demands of customers. Communication over Internet has changed from text data mode to multimedia mode. Internet will become an Integrated Service Internet that provides means to transmit real-time multimedia data over networks. The key issue now is to enhance the quality of the multimedia communication (real-time multimedia constructs and network database). At present, the multimedia technologies are progressing swiftly and violently. An upsurge for studying and developing multimedia techniques and products has formed. In this paper we will present a new system named Multimedia Information Service Center (MISC), which integrates real-time multimedia transmission system and web database system to provide single and simple access to multimedia information for customers. MISC can be used in on-line education, network radio, network TV and other multimedia communication over Internet.

        회원제 스포츠센터의 서비스 품질이 고객행동의도에 미치는 영향

        The purpose of the research is to investigate the relationship between service quality and customer behavior intention in membership sports center of Henan province, China. To achieve the goal, a total of 671 questionnaires used for analysis. Based on the collected data, Win SAS Ver.9.2 was applied for analysis with frequency analysis, confirmatory analysis, construct validity, correlation analysis and multiple linear regression analysis. The results of this study are as follows: The sub-scales of service quality such as communicability, environmental and social quality factors had a significant effect on the recommendation intention of behavioral intention, but no effect was found in product quality. The sub-scales of service quality such as environmental and social quality factors had a significant effect on word-of-mouth diffusion of behavioral intention, but no effect was found in product quality and communicability factors.

        Opportunistic Spectrum Access with Discrete Feedback in Unknown and Dynamic Environment:A Multi-agent Learning Approach

        This article investigates the problem of opportunistic spectrum access in dynamic environment, in which the signal-to-noise ratio (SNR) is time-varying. Different from existing work on continuous feedback, we consider more practical scenarios in which the transmitter receives an Acknowledgment (ACK) if the received SNR is larger than the required threshold, and otherwise a Non-Acknowledgment (NACK). That is, the feedback is discrete. Several applications with different threshold values are also considered in this work. The channel selection problem is formulated as a non-cooperative game, and subsequently it is proved to be a potential game, which has at least one pure strategy Nash equilibrium. Following this, a multi-agent Q-learning algorithm is proposed to converge to Nash equilibria of the game. Furthermore, opportunistic spectrum access with multiple discrete feedbacks is also investigated. Finally, the simulation results verify that the proposed multi-agent Q-learning algorithm is applicable to both situations with binary feedback and multiple discrete feedbacks.

      • DNA methylation involved in environmental adaptation of birch

        The boreal forest, with large tracts of unmanaged forests and low human impacts, is critical for the stability of global ecosystems. However, the boreal regions are subject to extensive ecosystem shifting, thinning and even loss under the climate changes and human activities, especially with the increasing of global warming. How to cope with the trend is extremely urgent. To illustrate responds of future populations of forest trees to climate change, it is essential to uncover past and present signatures of molecular adaptation in their genomes. Ecological epigenetics, as an interdisciplinary mainly answering how species, especially perennial, adapt to environmental changes epigenetically, is an important approach to detect the epigenetic mechanisms of boreal forest plants adapting to climate changes. Here, we will report the recent results about DNA methylation involved in environmental adaptation of birch, the pioneered trees of boreal forest.

        Characteristics of lipoxygenase-based and lipoxygenase-deficient soy yogurt with modified okara

        Lipoxygenase-based and lipoxygenase-deficientokara were modified by Kluyveromyces marxianus fermentation,then adding modified okara back to the correspondingsoymilk to prepare soy yogurt. Thephysicochemical properties, texture, and volatile componentsof soy yogurt were characterized. The results showedthat okara modified by Kluyveromyces marxianus fermentationwas rich in soluable dietary fiber and was impartedbetter water-holding capacity, swelling capacity, and oilholdingcapacity. The soy yogurt with the modified okarawas greatly enhanced in its appearance, texture and wasrelatively stable during storage. Moreover, lipoxygenasebasedsoy yogurt had a unique soybean flavor whilelipoxygenase-deficient soy yogurt had a slight beany flavorand soybean flavor. This article guides a bio-modifiedmethod for okara and provides a theoretical basis for thefurther development and application of soy yogurt withhigh dietary fiber as well as lipoxygenase-deficient soyyogurt.
