중이온 빔 조사에 의한 담배의 돌연변이 유도와 내염성 식물의 선발

중이온 빔을 이용한 방사선 돌연변이의 기초자료를 얻고자 담배의 수분·수정 직후의 원배에 100 Gy의 중이온빔을 조사하여 M_1세대의 종자를 얻었다. NaCI 2.0과 2.5%를 포함된 MS배지에 M_1 세대의 종자를 파종하여 17개통의 NaCI 저항성 식물체를 선발하였다. 저항성 식물체는 BY-4품종에 ^14N빔을 조사한 처리구에서만 관찰되었다. M_1 세대의 저항성 식물체의 경우 영양생장기간 중에는 외형적 변이가 관찰되지 않았으나 생식생장기의 화기에 있어서는 다양한 변이가 관찰되었다. 특히 암술 수술의 길이의 변화, 꽃잎의 변화, 불완전 화분립, 줄기당 꽃의 착생수 감소 등의 변이가 발생하였다. M_1 세대에서 17개의 NaCI저항성 계통 중에서 8계통은 M_2 세대에도 저항성형질이 발현되었고, 또한 M_2 세대의 3계통은 Mannitol이 포함된 배지에서 저항성을 보여주었다. Tobacco proembryos were irradiated with 100 Gy of heavy-ion beams (^14N, ^20Ne : 135 Mev/u) after 24 to 96 hours of pollination as a mutagen and screened M_1 generation for morphological mutants and salt-tolerant plants. Morphological and physilolgical characteristics of the salt-tolerant plants derived from the irradiated proembryo are discussed in this report. Mutants irradiated porembryos with the beams after pollination produced various kinds of morphological variation. A total of 17 salt-tolerant plants were selected from tobacco cultivar (BY-4) by treatment with ^14N beam. Shapes of filament and pollen grain of most salt-tolerant plants were abnormal compared with non-irradiated wild type, and seeds weight and fertility obviously decreased. The germination rates of the several M_2 lines on the saline and the mannitol condition were higher than that of wild type.
중부지역 옥수수 파종기에 따른 생육단계 및 수량성 변화

Maize hybrids were planted on April 1, April 25, May 20 and June 15 in 2012, 2013 at the experimental paddy and upland fields in Suwon city , respectively to know changes of growth stage and yield according to different planting time. Maize planting on April 1 was not recommended because total growing days at planting on April 1 was not different with that at planting on April 25 due to many days from planting to emergence over 20 days at planting on April 1. Total growing days were not much different among three planting time after April 25 because days from silking to maturity was lengthened though days from planting to silking was shortened at later planting time. Stalk weight was decreased as planting time was delayed, particularly at planting times after May 20 at paddy field by excess water injury during early growth stage. But, grain yield was not low as in May and June. It is needed to introduce early maturity maize variety as Yanganok at late planting for higher grain yield and enough time for soil preparation for next winter crops

Dandelion has been widely utilized for medicinal and edible purposes. This research was conducted to evaluate the effect of supplemental LED (light-emitting diode) light on germination, growth characteristics and anthocyanin content of dandelion (Taraxacum officinale) seedling using LED blue (460 nm), red (660 nm, R), blue + red (B:R=6:4) and fluorescent lamp light treatment. By LED illumination to T. officinale seed germination speed was delayed, and germination rate was the highest in the fluorescent light. The growth characteristics (plant height, number of leaves, root length and fresh weight) were greatly influenced by supplemental LED light compared with control treatment, and the growth promotion was the most effective in the red LED illumination. After 60 days of red and mixed LED light treatments, anthocyanin content of dandelion plants was significantly changed. The anthocyanin content was increased by 12~19 mg/100 g under the red LED and the mixed light conditions compared with the control and the blue LED. Results indicate that illumination with red and mix LEDs, compared with other light treatments, is beneficial for promotion of growth and anthocyanin content in dandelion.

Herbicide tolerant rice (Oryza sativa L. cv. Ilpumbyeo) cell lines were selected from $\gamma$-ray-irradiated anther-derived cell cultures. The anther-derived cell clusters were small (300 to 400 ${\mu}{\textrm}{m}$ in diameter) and uniform ones that were screened by miracloth filtering. The cell suspensions were very efficient to plate one layer onto agar medium and to screen target cell lines. Herbicide tolerant cell lines were selected by 5 mg/L cyhalofop butyl (CHB) treatment by using the small cell suspensions on agar N6 medium containing 1 mg/L 2,4-D and 0.2 mg/L kinetin. Of the cell lines, one line (CHB-1) showed stable tolerance at 10 mg/L concentration after 6-month culture without herbicide suspension. Growth stability of CHB-1 was similar to that of control cell line on 10 mg/L CHB containing medium. In this experiment we established herbicide tolerant cell line selection system by using anther-derived uniform-cell suspensions with $\gamma$-ray-irradiation.

The effect of carbenicillin on the dedifferentiation and the regeneration efficiency of plant tissues of horseradish(Armoracia rusticana) was evaluated. Inhibition effect for callus initiation was observed when leaf blade, root and petiole segments were grown on MS medium containing 500 mg/L to 2000 mg/L carbenicillin and 0.5 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D). The regeneration of horseradish shoots from leaf blade, root and petiole explants were decreased as the addition of carbenicillin increased from 1000 mg/L to 2000 mg/L in MS medium containing 0.5 mg/L of 6-benzylaminopurine (BAP) or kinetin. Especially, 500 mg/L carbenicillin treatment significantly inhibited shoot induction when leaf blade explants were grown on hormone-free MS medium. It was suggested that the toxic effects of combinations of carbenicillin and 2,4-D may be due to high auxin activity levels.

In this study, ISSR markers were applied to assess the genetic diversity and structure analysis of tea (Camellia sinesis L. O. Kuntze) among the 3 cultivated populations, a wild population and single variety population (cv. Yabukita) as a control in Korea. Out of 129 loci detected overall, 120 were identified as being polymorphic with a rate of 93.02% in the 50 individuals from the populations. The polymorphic rate of the within population was over (more than) 80% in the four populations, but 16% in the single variety population. The mean of Nei's gene diversity (h) and Shannon‘s information index (S.I.) of the cultivated populations were lower than that of the wild populations. The high values for the total gene diversity, effective and Nei’s genetic diversity indicated substantial variations among the cultivated population. In addition, the insights into the relative gene diversity among and within population of Jeonggeum-ri would be useful in breeding and for the development of strategies for tea genetic population. Based on the cluster analysis of UPGMA, the genetic relationship among the all populations was coincided with the pattern of cultural environment.
담배와 벼의 발아와 생장에 대한 Proton 빔조사의 영향

Effects of proton beam irradiation on seed germination and growth pattern of tobacco (Nicotiana tabacum L. cv. BY-4; N. plumbaginifolia) and rice (Oryasativa L.) plants were estimated to develop the efficient conditions of irradiation. Seed germination rate was decreased by increasing the proton beam the current and the beam irradiation time in both tobacco and rice seeds. The beam irradiation conditions showing 50% germination were over 60 sec at 10 nA, approximately 5 sec at 100 nA and at 500 nA beam current in tobacco seeds. And the conditions of 50% germination were 60 sec at 10 nA, and 100 nA and 30 sec at 500nA in rice (cv. Dongjin 1) seeds. The growth of irradiated plants was decreased, but significant difference in morphological changes was not observed by the proton beam treatment. The proton beam is able to use as a mutagen, but some of the factors including beam size and beam detector-system must be established for efficient usage of the beam.

재배종담배(Nicotiana tabacum L. cv.), 야생종담배 (N. plumbaginifolia)와 벼종자(Orya sativa L, cv.)에 neutron빔을 각각 조사(irradiation)하여 발아, 식물체의 생장과 DNA의 변이에 미치는 영향을 검토하였다. 담배종자와 벼종자에 90, 180, 270, 360, 450, 540 Gy까지 조사로 발아율은 크게 감소하지 않았고, 생장에 있어 유의적인 차이는 없었다. 그러나 담배식물체에서 총 200개체 중 71개체(약 36%)가 잎에서 형태 이상을 나타내었다. 또한 줄기색 변이체, 엽색변이체, 화형변이체를 유도하였다. 이 결과는 neutron빔이 유용한 돌연변이원으로서 가능성이 있음을 시사해 준다. Neutron빔을 조사한 후 생장한 담배식물체의 잎에 대하여 총 34개의 primer를 이용하여 RAPD 분석한 결과 20개의 primer에서 총 104개의 DNA 단편이 증폭되었고, 중성자 빔조사처리구에서만 출현하는 DNA 단편은 나타나지 않았다. Effects of neutron beam irradiation on seed germination, growth and RAPD pattern of tobacco (Nicotiana tabacum L. cv.; N. plumbaginifolia) and rice (Orya sativa L. cv.) plants were estimated. Seed germination rate was not significantly changed by the neutron beam treatment in both tobacco and rice seeds. And there was no significant differance in growth of the plants by the neutron beam treatment. Interestingly, however, some of morphological changes, including leaf shape (about 36%), stem color and leaf color were observed in neutron beam treated tobacco plants. In addition, abnormal flower in petal was observed in the neutron beam treated plant. This results indicate that neutron beam is able to use as an effective mutagen in plant mutations. Scorable products from 20 primers were obtained by RAPD analysis in the leaves of the beam irradiated tobacco plants and most of the plants showed the similar band patterns.
