Isonitrosoethylacetoacetate Imine 리간드의 니켈(II) 착물 합성 및 구조 특성

Nickel(lI) Complexes of the type Ni(IEAA-NH)(IEAA-NR) (R = H₁, CH₃, C₂H_(5), n-C₃H_(7), n-C₄H_(9), or CH₂C_(6)H_(5)), where IEAA-NH and IEAA-NR represents isonitrosoethylacetoacetate imine and its N-alkyl derivative, respectively, have been prepared. And the structure of the complexes have been studied by elemental analyses, electronic, infrared,1H and 13C-NMR spectra. It has been determined that IEAA-NH ligand coordinates to nickel through the nitrogen of imine group and the oxygen of isonitroso group to form 6-membered chelate ring and IEAA-NR coordinates to nickel through the nitrogens of imino group and isonitroso group to form 5-membered chelate ring in the square-planar Ni(IEAA-NH)(IEAA-NR) complexes.

Methylation of CpG islands in the promoter region of genes acts as a significant mechanism of epigenetic gene silencing in head and neck squamous cell carcinoma (HNSCC). DNA methylation markers are particularly advantageous because DNA methylation is an early event in tumorigenesis, and the epigenetic modification, 5-methylcytosine, is a stable mark. In the present study, we assessed the genome-wide preliminary screening and were to identify novel methylation biomarker candidate in HNSCC. Genome-wide methylation analysis was performed on 10 HNSCC tumors using the Methylated DNA Isolation Assay (MeDIA) CpG island microarray. Validation was done using immunohistochemistry using tissue microarray of 135 independent HNSCC tumors. In addition, in vitro proliferation, migration/invasion assays, RT-PCR and immunoblotting were performed to elucidate molecular regulating mechanisms. Our preliminary validation using CpG microarray data set, immunohisto-chemistry for HNSCC tumor tissues and in vitro functional assays revealed that methylation of the Homeobox B5 (HOXB5) and H6 Family Homeobox 2 (HMX2) could be possible novel methylation biomarkers in HNSCC.

isonitrosomethylacetoacetate(H-IMAA)의 이민유도체를 리간드로하는 새로운 Ni(II) 및 Pd(II)착물, Ni(IMAA-NH)(IMAA-NH'), Ni(IMAA-NH)(IMAA-NR),$ Pd(IMAA-NH)_2$, 및 $Pd(IMAA-NR)_2(R=CH_3,\;n-C_3H_7,\;n-C_4H_9$, 또는 $CH_2C_6H_5)$을 합성하고, 이들 착물의 구조를 원소분석, 전자흡수분광법, 적외선흡수분광법, $^1H$ 및 $^{13}C$ 핵자기공명분광법으로 조사하였다. 여기서 H-IMAA-MH 및 H-IMAA-NR은 각각 isonitrosomethylacetoacetate imine 및 N-alkylisonitrosomethylacetoacetate imine 유도체를 나타낸다. Effects of Araliae Radix on the Central Nervous System and the Cardiovascular System

These studies were conducted as an attempt to investigate the effects of Araliae Radix on analgesic, sedative, the relaxing action of isolated ileums and cardiovascular system. The results of these studies were summarized as follows: 1. Analgesic and sedative actions were recognized. 2. The relaxing action was shown on the extracted ileum in mice, antagonistic actions were seen on acetylcholine and BaCl₂-induced contraction of the ileum of mice and histamine-induced contraction of the ileum of guinea-pigs that the relaxing effect of the intestinal smooth muscle was recognized. 3. Significant inhibition of cardiac systolic action was recognized. 4. Hypotensive effect was observed.

<P><B>Background:</B></P><P>Head and neck squamous cell carcinomas (HNSCCs) are highly lethal epithelial tumours containing self-renewal cancer stem cells (CSCs). CSCs in HNSCCs are strongly associated with tumour initiation, invasion, and chemoradiation resistance. However, the important factors regulating stemness in HNSCCs remain unclear. Here, we investigated the molecular roles and clinical significance of inhibitor of DNA binding 2 (Id2) protein to determine if it constitutes a novel therapeutic target for ablating HNSCC cells with stemness.</P><P><B>Methods:</B></P><P>We performed <I>in vitro</I> and <I>in vivo</I> studies of Id2 function and its effects on stemness using HNSCC cells. We also examined whether Id2 expression could be used as a prognostic indicator through immunohistochemical staining of 119 human HNSCC tumours.</P><P><B>Results:</B></P><P>Expression of Id2 was higher in HNSCC cells with stemness compared with differentiated HNSCC cells. Overexpression of Id2 increased proliferation, self-renewal, and expression of the putative stemness marker CD44 in HNSCC cells <I>in vitro</I> and <I>in vivo</I>. In contrast, silencing of <I>Id2</I> using short hairpin RNA attenuated the stemness phenotype of HNSCC cells by reducing self-renewal, CD44 expression, cisplatin chemoresistance, and xenograft tumourigenicity. Most importantly, increased expression of Id2 was closely associated with poorer post-treatment survival rates in HNSCC patients.</P><P><B>Conclusions:</B></P><P>Inhibitor of DNA binding2 represents a novel and promising therapeutic target for treating and improving the clinical outcomes for patients with HNSCC.</P>
