        This study aims to develop a performance evaluation system for the Construction Factory(CF). The CF is a kind of full automation system for building construction which consists of the lifting system for building materials, the built-up unit for steel structural components, the bolting robots, the control center for the site management, and the site covering system. The CF is developing now as a project of the construction automation and robotics. In this study we firstly reviewed the state-of-the-art of the construction automation and robotics in the foreign and the domestic, and investigated the precedent case of the CF such as the SMART System of Shimizu Co., Japan. We believe that without an objective evaluation of the results there won``t be growth in technological R&D. Therefore, this study sees the developing CF as an new technology and method in building construction, and proposes the direction and frame of the appropriate evaluation which can be applied into the CF.

        하수처리장 적용을 위한Semi-카플란 수차가 장착된 마이크로수력발전 시스템

        소수력발전은 하수처리장 에너지 자립을 위한 효과적인 대안이다. 본 연구는 유량변동이 크고 유효낙차가 낮은 중소형하수처리장(기흥레스피아) 적용을 위해 피치조절형 세미카플란(semi-kaplan) 마이크로수력발전의 적용 타당성을 평가하였다. 가변피치 semi-kaplan 수차는 유량조절을 위한 가이드베인은 생략하고 피치조절형 런너를 장착하여 기계적 결함은 줄이면서 유량변동이 큰 처리장에 특화된 기술이다. 마이크로수력발전 시스템은 설계조건(유량 0.35 m3/s, 유효낙차 4.7 m)에서 90.2%의 수차효율 달성이 가능하였고 발전용량은 13.4 kW로 산정되었다. 설비가동률 74%로 가동 시 연간 약 86.8 MWh 에너지 생산을 통해 2.1%의 에너지 자립이 가능하고 이는 연간 49톤의 CO₂ 감축효과와 맞먹는다. 경제성 평가결과 초기 건설공사비가 200,000,000원 이하인 경우에는 내부수익률은 6.1%, 순현가는 15,539,000원, 편익-비용률은 1.08, 투자회수년은 15.5년으로 경제성이 충분한 것으로 나타났다. Small scale hydropower is one of most attractive and cost-effective energy technologies for installation within sewage treatment plants. This study was conducted to evaluate the potential of a semi-kaplan micro-hydropower (MHP) system for application to sewage treatment plants with high flow fluctuations and a low head. The semi-kaplan MHP is equipped with an adjustable runner blade, and is without a guide vane, so as to reduce the incidence of mechanical problems. A MHP rating 13.4 kWp with a semi-kaplan turbine has been considered for Kiheung Respia sewage treatment plant, and this installation is estimated to generate 86.8 MWh of electricity annually, which is enough to supply electricity to over 25 households, and equivalent to an annual reduction of 49 ton CO₂. The semi-kaplan turbine showed a 90.2% energy conversion efficiency at the design flow rate of 0.35 m3/s and net head of 4.7 m, and was adaptable to a wide range of flow fluctuations. Through the MHP operation, approximately 2.1% of total electricity demand of Kiheung Respia sewage treatment plant will be achievable. Based on financial analysis, an exploiting MHP is considered economically acceptable with an internal rate of return of 6.1%, net present value of 15,539,000 Korean Won, benefit- cost ratio of 1.08, and payback year of 15.5, respectively, if initial investment cost is 200,000,000 Korean Won.

        Development of a One-Step Multiplex PCR Assay for Differential Detection of Major <i>Mycobacterium</i> Species

        <P>The prevalence of tuberculosis continues to be high, and nontuberculous mycobacterial (NTM) infection has also emerged worldwide. Moreover, differential and accurate identification of mycobacteria to the species or subspecies level is an unmet clinical need. Here, we developed a one-step multiplex PCR assay using whole-genome analysis and bioinformatics to identify novel molecular targets. The aims of this assay were to (i) discriminate between the Mycobacterium tuberculosis complex (MTBC) and NTM using rv0577 or RD750, (ii) differentiate M. tuberculosis (M. tuberculosis) from MTBC using RD9, (iii) selectively identify the widespread M. tuberculosis Beijing genotype by targeting mtbk_20680, and (iv) simultaneously detect five clinically important NTM (M. avium, M. intracellulare, M. abscessus, M. massiliense, and M. kansasii) by targeting IS1311, DT1, mass_3210, and mkan_rs12360. An initial evaluation of the multiplex PCR assay using reference strains demonstrated 100% specificity for the targeted Mycobacterium species. Analytical sensitivity ranged from 1 to 10 pg for extracted DNA and was 10(3) and 10(4) CFU for pure cultures and nonhomogenized artificial sputum cultures, respectively, of the targeted species. The accuracy of the multiplex PCR assay was further evaluated using 55 reference strains and 94 mycobacterial clinical isolates. Spoligotyping, multilocus sequence analysis, and a commercial real-time PCR assay were employed as standard assays to evaluate the multiplex PCR assay with clinical M. tuberculosis and NTM isolates. The PCR assay displayed 100% identification agreement with the standard assays. Our multiplex PCR assay is a simple, convenient, and reliable technique for differential identification of MTBC, M. tuberculosis, M. tuberculosis Beijing genotype, and major NTM species.</P>

      • Cytoskeleton-associated proteins are enriched in human embryonic-stem cell-derived neuroectodermal spheres

        <P>The ability to generate neural lineages from human embryonic stem cells (hESCs) in a controlled manner would further investigation of human neurogenesis and development of potential cell therapeutic applications to treat neurological diseases; however, generating such neural stem cells (NSCs) remains a challenge. In an attempt to characterize the cellular mechanisms involved in hESC differentiation into neuroprogenitor cells, we performed 2-DE using protein extracts from hESC-derived embryoid bodies (EBs) and neuroectodermal spheres (NESs) bearing neuroprogenitors. Of 47 differentially expressed protein spots, 28 nonredundant spots were shown to be upregulated in the NESs; these protein spots included neurogenesis-related proteins (TAF1, SEPT2, NPH3, and CRABP), as expected. Interestingly, 6 of these 28 protein spots were cytoskeleton-associated proteins (CSAP) such as Fascin-1, Cofilin-1, and Stathmin-1. Western-blot analyses confirmed the increased levels of these proteins in the NESs. Furthermore, immunostaining analysis showed that both Fascin-1 and Stathmin-1 were preferentially expressed in the inner rims of neural rosettes, which are characteristic features of neuroprogenitors in culture. We also confirmed prominent expression of Fascin-1 in (sub-)ventricular zone in E15.5 mouse fetal brain. Our results suggest that, in addition to the induction of those genes involved in neural development, hESC differentiation into the NES is associated with a marked reorganization of the cellular cytoskeleton.</P>

        Diagnostic accuracy and safety of 16α-[<sup>18</sup>F]fluoro-17β-oestradiol PET-CT for the assessment of oestrogen receptor status in recurrent or metastatic lesions in patients with breast cancer: a prospective cohort study

        <P><B>Summary</B></P> <P><B>Background</B></P> <P>A biopsy of first recurrence or metastatic disease is recommended to re-evaluate oestrogen receptor status in patients with breast cancer and to select appropriate treatment. However, retesting for oestrogen receptor status with rebiopsy is not always feasible, depending on lesion location and the risk associated with biopsy, and in these cases clinicians continue to treat patients according to the oestrogen receptor status of the primary tumour. Consequently suboptimal therapy might be offered to these patients. We assessed the diagnostic accuracy and safety of 16α-[<SUP>18</SUP>F]fluoro-17β-oestradiol (<SUP>18</SUP>F-FES) PET-CT to assess oestrogen receptor status in patients with recurrent or metastatic breast cancer.</P> <P><B>Methods</B></P> <P>We did a prospective cohort study at the Asan Medical Center, Seoul, South Korea. Eligible patients had breast cancer, with first recurrence or metastatic disease at presentation, were 19 years or older, and had an Eastern Cooperative Oncology Group performance status of 0–2. The primary objective was to show the agreement between qualitative <SUP>18</SUP>F-FES PET-CT interpretation and the results of oestrogen receptor expression by immunohistochemical assay, a non-reference standard test. Whole-body <SUP>18</SUP>F-FES PET-CT imaging was done after intravenous injection of 111–222 MBq of <SUP>18</SUP>F-FES, with dosing primarily determined by radiation dosimetry analysis. <SUP>18</SUP>F-FES uptake above background intensity was interpreted as positive. Efficacy was assessed in all patients with histologically confirmed recurrent or metastatic breast cancer who received <SUP>18</SUP>F-FES and had PET-CT images available (intention-to-diagnose analysis), and safety was assessed in all patients who received <SUP>18</SUP>F-FES. This study is registered with ClinicalTrials.gov, number NCT01986569.</P> <P><B>Findings</B></P> <P>Between Nov 27, 2013, and Nov 10, 2016, 93 patients were enrolled. Of the 85 patients included in the efficacy analysis, 47 (55%) were oestrogen receptor-positive and 38 (45%) were oestrogen receptor-negative. Positive status percent agreement between the <SUP>18</SUP>F-FES PET-CT results and oestrogen receptor status by immunohistochemical assay was 76·6% (95% CI 62·0–87·7) and the negative status percent agreement was 100·0% (90·8–100·0). Patients who were oestrogen receptor-positive and had a positive <SUP>18</SUP>F-FES PET-CT result had a significantly higher progesterone receptor expression than those who were oestrogen receptor-positive and had a negative <SUP>18</SUP>F-FES PET-CT result (23 [68%] of 34 patients <I>vs</I> 0 of 11 patients; p<0·0001). The most common adverse event was procedural pain in nine (10%) of 90 patients injected with <SUP>18</SUP>F-FES. No adverse events were related to the study drug except injection site pain in one (1%) patient. No serious adverse events were recorded.</P> <P><B>Interpretation</B></P> <P>The high negative percent agreement between <SUP>18</SUP>F-FES PET-CT and oestrogen receptor status by immunohistochemical assay in this cohort suggests that positive <SUP>18</SUP>F-FES uptake by recurrent or metastatic oestrogen receptor-positive breast cancer lesions could be an alternative to oestrogen receptor assays in this setting. Staging assessment should include <SUP>18</SUP>F-FES PET-CT when retesting oestrogen receptor status is not feasible.</P> <P><B>Funding</B></P> <P>Asan Institute for Life Sciences, Ministry of Health and Welfare, South Korea.</P>

      • Comparative Genomics of T-complex protein 10 like in Humans and Chimpanzees

        Comparing 231 genes on chimpanzee chromosome 22 with their orthologous on human chromosome 21, we have found that 15 orthologs have indels within their coding sequences. It was rather surprising that significant number of genes have changed by indel, despite the shorter time since their divergence and led us hypothesize that indels and structural changes may represent one of the major mechanism of proteome evolution in the higher primates. Human T-complex protein 10 like (TCP 10L) is a representative having indel within its coding sequence. Gene structure of human TCP10L compared with chimpanzee TCP10L gene showed 16 base pair difference in genomic DNA. As a result of the indel, frame shift mutation occurs in coding sequence (CDS) and human TCP10L express longer polypeptide of 21 amino acid residues than that of chimpanzee. Our prediction found that the indel may affect to dramatic change of secondary protein structure between human and chimpanzee TCP10L. Especially, the structural changes in the C-terminal region of TCP10L protein may affect on the interacting potential to other proteins rather than DNA binding function of the protein. Through these changes, TCP10L might influence gene expression profiles in liver and testis and subsequently influence the physiological changes required in primate evolution.

        BDNF Enhancement of Neuronal Differentiation with Adipose-Derived Stem Cells(ADSCs)

        Adipose-derived stem cells(ADSCs) are multipotent cells that can be manipulated to differentiate into different cell lines and are considered a potential cell source for regenerative medicine and tissue engineering. The brain derived neurotrophic factor (BDNF) is known to stimulate stem cells to differentiate into neuronal cells as well as promote plasticity. The purpose of this study was to evaluate the effects of BDNF on the neuronal differentiation of ADSCs. The proliferation of the ADSCs was increased by BDNF in a dose-dependent manner. The expression of neuronal markers including Nestin and microtubule-associated protein(MAP2) were highly upregulated by BDNF. In addition, BDNF considerably suppressed the expression of glial fibrillary acidic protein(GFAP), a preventative glial cell or astrocyte marker. Using immunocytochemistry, we confirmed the results by real time-PCR. An increased production of Nestin and MAP2 by BDNF, but not GFAP was observed. These data show that supplementation of the ADSCs with BDNF effectively maintained cell survival, proliferation and differentiation of these cells into neuronal precursor cells. In conclusion, specific neuronal differentiation of ADSCs after treatment with BDNF increased the generation of a mature neuronal phenotype.

        The Role of BF-7 on Neuroprotection and Enhancement of Cognitive Function

        Amyloid ${\beta}-peptide\;(A{\beta})$ contributes to the pathogenesis of Alzheimer's disease (AD), causing neuronal death through apoptosis. In this study, the neuroprotective role of BF-7, extracted form sericultural product, was examined against $A{\beta}-induced$ toxicity in cultured human neuronal cell SKN-SH. In order to know if the BF-7 has positive role on the cognition and memory in human, the mixture of BF-7, DHA and EPA (BDE) was examined using Rey Kim and K-WAIS test with 50 healthy high school student. We report here that BDE significantly attenuated $A{\beta}-induced$ apoptosis through the reduction of ROS accumulation, and diminished caspase-like protease activity. Moreover, the memory index and memory preservation, and attentative concentration of BDE treated group for 1 month were significantly improved, in contrast to the case of placebo control treated with DHA and EPA. This result represent that the BF-7 play significant positive role on learning memory. Taken together, our result suggested the natural product BF-7 is a good substance for the brain functionally and physiologically.
