Mesenchymal stem cells (MSCs) are widely used in cartilage tissue engineering to repair articular cartilage defects. However, hypertrophy of chondrocytes derived from MSCs might hinder the stabilization of hyaline cartilage. Thus, it is very important to find a suitable way to maintain the chondrogenic phenotype of chondrocytes. It has been reported that cordycepin has anti-inflammatory and anti-tumor functions. However, the role of cordycepin in chondrocyte hypertrophy remains unclear. Therefore, the objective of this study was to determine the effect of cordycepin on chondrogenesis and chondrocyte hypertrophy in MSCs and ATDC5 cells. Cordycepin upregulated chondrogenic markers including Sox9 and collagen type II while down-regulated hypertrophic markers including Runx2 and collagen type X. Further exploration showed that cordycepin promoted chondrogenesis through inhibiting Nrf2 while activating BMP signaling. Besides, cordycepin suppressed chondrocyte hypertrophy through PI3K/Bapx1 pathway and Notch signaling. Our results indicated cordycepin had the potential to maintain chondrocyte phenotype and reconstruct engineered cartilage. [BMB Reports 2016; 49(10): 548-553]

Plant P450s play an important role in plant response to environmental stimuli, especially to pathogen challenge, besides leading to the biosynthesis of secondary metabolites. In this work, a Verticillium dahliae-induced cDNA sequence was cloned from wild eggplant Solanum torvum and designated StoCYP77A2. StoCYP77A2 encodes a 511-amino acid peptide that shows sequence homology to the plant CYP77 family. StoCYP77A2 is expressed in a tissue-specific manner with the highest expression level in stem. The expression level of StoCYP77A2 was increased in wild eggplants which were infected with Verticillium dahlia. We expressed StoCYP77A2 in V. dahliae-sensitive tobacco, and the transgenic tobacco plants were more resistant to V. dahliae infection than the wild type. Protein extraction of transgenic tobacco showed strong antifungal activity against V. dahliae, indicating that StoCYP77A2 may be involved in the synthesis of some antimicrobial compounds. These results suggest that StoCYP77A2 plays a role in plant defense to V. dahliae infection.

Since the first nuclear reactor first critical, nuclear systems has gone through four generations of history,and the fourth generation nuclear system will be truly realized in the near future. The notions of SVBRand lead-bismuth eutectic alloy coolant put forward by Russia were well received by the internationalnuclear science community. Lead-bismuth eutectic alloy with the ability of the better neutron economy,the low melting point, the high boiling point, the chemical inertness to water and air and other features,which was considered the most promising coolant for the 4th generation nuclear reactors. This studymainly focuses on the structural design optimization of the 4th-generation reactor coolant pump,including analysis of external characteristics, inner flow, and transient characteristic. It was found that:the reactor coolant pump with a central symmetrical dual-outlet volute structure has better radialdirectionbalance, the pump without guide vane has better hydraulic performance, and the pumpwith guide vanes has worse torsional vibration and pressure pulsation. This study serves as experienceaccumulation and technical support for the development of the 4th generation nuclear energy system.

In this paper, an adaptive radial basis function neural network (RBFNN) controller based on extraordinariness particle swarm optimization (EPSO) is proposed. To improve the trajectory tracking performance of robotic manipulators, the uncertainties of the manipulator dynamic equation are locally approximated using three RBFNNs with optimized hyperparameters. Besides, a robust control item is also considered in the controller to resist external disturbances. During hyperparameters optimization, the EPSO optimizer iteratively optimizes the hyperparameters of the RBFNN controller using the composite error of the system output. The stability of the control scheme is analyzed with the Lyapunov stability. Simulation results as well as the experimental verification prove the efficiency and applicability of the control scheme.

A novel polysaccharide (AGP-32) from the gonad of Haliotis discus hannai Ino was isolated using a protease-assisted process and successive ion-exchange and gel-filtration chromatography. The backbone of AGP-32 was determined using hydrolysis with trifluoroacetic acid. FTIR, NMR, and methylation analysis, and periodate oxidation and Smith degradation analysis revealed that the AGP-32 backbone mainly consisted of (1→6)-linked mannose, (1→3)-linked galactose, and (1→3)-linked glucose in a proportion of 2:3:1. An in vitro cell assay indicated that AGP-32 promoted mice splenic lymphocyte proliferation by 26% at a concentration of 50 μg/mL. AGP-32 had an effect on immune protection and is a candidate for consideration as a functional food.
