ASCL2 Gene Expression Analysis and Its Association with Carcass Traits in Pigs

Achaete-scute like 2 (ASCL2) gene encodes a member of the basic helix-loop-helix transcription factor which is essential for the maintenance of proliferating trophoblasts during placental development. ASCL2 gene preferentially expresses the maternal allele in the mouse. However, it escapes genomic imprinting in the human. In this study, the complete open reading frame consisting of 193 amino acids of ASCL2 gene was obtained. Sequence analysis indicated that a C-G mutation existed in the 3' region between Meishan and Large White pigs. The polymorphism was used to determine the monoallelic or biallelic expression with RT-PCR-RFLP in pigs of Large $White{\times}Meishan$ $F_1$ hybrids. Imprinting analysis indicated that the ASCL2 gene expression was biallelic in all the tested tissues (heart, liver, spleen, lung, kidney, stomach, small intestine, skeletal muscle, fat, uterus, ovary and pituitary). PCR-RFLP was used to detect the polymorphism in 270 pigs of the "$Large\;White{\times}Meishan$" $F_2$ resource population. The statistical results showed highly significant associations of the genotypes and fat meat percentage (FMP), lean meat percentage (LMP) and ratio of lean to fat (RLF) (p<0.01), and significant associations of the genotypes and loin eye area (LEA) and internal fat rate (IFR) (p<0.05).

Association Analyses with Carcass Traits in the Porcine KIAA1717 and HUMMLC2B Genes

By screening a subtracted cDNA library constructed with mRNA obtained from the longissimus dorsi muscles of F1 hybrids Landrace${\times}$Yorkshire and their Yorkshire female parents, we isolated two partial sequences coding for the H3-K4-specific methyltransferase (KIAA1717) and skeletal muscle myosin regulatory light chain (HUMMLC2B) genes. In the present work we investigated two SNPs, one (C1354T) at the 3' untranslated region (UTR) of KIAA1717 and one (A345G) at the SINE (PRE-1) element of HUMMLC2B, in a resource population derived from crossing Chinese Meishan and Large White pig. The selected pigs were genotyped by means of a PCR-RFLP protocol. Significant associations were observed for the KIAA1717 C1354T polymorphic site with thorax-waist backfat thickness (p<0.05), buttock backfat thickness (p<0.05), average backfat thickness (p<0.05), loin eye height (p<0.05), loin eye area (p<0.05), carcass length to 1$^{st}$ spondyle (p<0.01) and carcass length to 1st rib (p<0.01). HUMMLC2B A345G were significantly associated with loin eye width (p<0.05), loin eye area (p<0.05). Further studies are needed to confirm these preliminary results.

Effect of Genistein on Antioxidative Defence System and Membrane Fluidity in Chick Skeletal Muscle Cells

This study was conducted to investigate the protective effect of genistein on the antioxidative defence system and membrane fluidity in chick skeletal muscle cells after supplementation with 0, 20, 40, and $80{\mu}mol/L$ genistein in $50{\mu}mol/L$ $FeSO_4/H_2O_2$ treated cells for 24 h. Genistein supplementation recovered the decreased activity of total superoxide dismutase induced by $FeSO_4/H_2O_2$, significantly increased glutathione peroxidase activity (p<0.05) and decreased malondialdehyde production (p<0.05). The treatment of 80 mol/L genistein in $FeSO_4/H_2O_2$ treated cells decreased the secretion of creatine kinase (p<0.05). Fluorescence polarization values and microviscosities observed with $FeSO_4/H_2O_2$ treated cells were significantly higher than those observed with no $FeSO_4/H_2O_2$ treated cells. The addition of $80{\mu}mol/L$ genistein improved the increased fluorescence polarization value (p<0.05) caused by $FeSO_4/H_2O_2$ treatment. The microviscosity value was significantly decreased by adding genistein (p<0.05). In conclusion, genistein protected skeletal muscle cells from oxidative damage by improving antioxidative status and membrane fluidity.

Effects of Feeding Purified Zearalenone Contaminated Diets with or without Clay Enterosorbent on Growth, Nutrient Availability, and Genital Organs in Post-weaning Female Pigs

The effects of different levels of natural clay enterosorbent on the growth, nutrient availability, and genital organs of post-weaning female pigs fed with an addition of zearalenone (ZEA) were investigated in the study. A total of thirty-five post-weaning gilts ($L{\times}Y{\times}D$) with an average body weight of 12.36${\pm}$1.46 kg were used in the test. The gilts were raised individually in metabolism cages and fed a corn-soybean meal-whey basal diet with an addition of 0 or 1 mg/kg of ZEA for 24 d with four levels of natural clay enterosorbent added in the feed. The treatments were: i) control; ii) control+2.5 g/kg clay; iii) control+1 mg/kg ZEA; iv) control+1 mg/kg ZEA+1.25 g/kg clay; v) control+1 mg/kg ZEA+2.5 g/kg clay; vi) control+1 mg/kg ZEA+5.0 g/kg clay; vii) control+1 mg/kg ZEA +10 g/kg clay. Pigs fed diets contaminated with additional purified ZEA had significantly reduced apparent digestibility of crude protein (CP), gross energy (GE) and apparent metabolic rate of GE (ME/GE, p<0.05) without changes of net protein utilization (NPU, p>0.05). Final body weight, average daily gain (ADG), average daily feed intake (ADFI), vulva length, vulva width, vulva area, relative weights of genital organ and proliferative changes of the ovary tissues in gilts fed ZEA-contaminated diet were increased (p<0.05) compared to the gilts fed the control diet. Addition of natural clay enterosorbent in the ZEA-contaminated diet showed a positive protection effect on ZEA feeding, and the protection was increased linearly or quadratically as clay content increased. However, in pigs fed a diet with clay alone at 2.5 g/kg level there was no significant impact (p>0.05) on all the parameters as compared to the control. It is suggested that feeding ZEA at about 1.0 mg/kg for 24 days might result in a deleterious effect in pigs, and addition of 5 or 10 g clay enterosorbent per kg diet can effectively neutralize the detrimental effects of the ZEA feeding.

Controlling the initial conditions of precursor powders and its effects on the phase evolution and $J_c$ properties of Bi-2223/Ag tapes

By varying fabricating process, precursor powders with different initial conditions were prepared. Subsequently, Bi-2223/Ag tapes were made through these powders. The effects of precursor powders on the phase evolution and Jr properties of Bi-2223/Ag tapes were studied along with several thermomechanical cycles. Our results showed that the initial conditions of precursor powders could strongly influence the phase formation rate and $J_{c}$ value in final tapes. The factors of precursor powders that influence the phase formation and $J_{c}$ of Bi-2223/Ag tapes must be studied and optimized in combination.ion.

Synthesis and evaluation of a novel chiral derivatization reagent for resolution of carboxylic acid enantiomers by RP-HPLC

<P>A novel derivatization reagent, N-[1-Oxo-5-(triphenylphosphonium)pentyl]-(S)-3-aminopyrrolidine (OTPA), with triphenylphosphine (TPP) as a basic structure carrying a permanent positive charge was developed for the enantiomeric separation of chiral carboxylic acids by high-performance liquid chromatography (HPLC). OTPA reacted with the carboxylic acids at 40 degrees C within 90 min in the presence of 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and 1-hydroxybenzotriazole (HOBt). The degree of epimerization (racemization) during the derivatization reaction was negligible. The separability of the diastereomers was evaluated in terms of their resolution value (Rs). The Rs values of the derivatives of non-steroidal anti-inflammatory drugs (NSAIDs), which were selected as the representative carboxylic acids, were completely separated by reversed-phase chromatography using an ODS (4.6 mm x 150 mm I.D., 5.0 mu m) column. The resolution Rs values were 1.54-2.23 for the evaluated carboxylic acids and the OTPA-derivatization was also effective for the enantiomeric separation of chiral carboxylic acids. The calibration curves (r(2) > 0.9971) were linear over the concentration range of 0.0125-1.25 mM for each enantiomer of ketoprofen (KET), and naproxen (NAP), 0.05-1.0 mM for each enantiomer of ibuprofen (IBU), 2-phenylpropionic acid (PPA), and loxoprofen (LOX), and 0.05-1.25 mM for each enantiomer of PBA. The limit of detection (S/N = 3) for each of the enantiomers of the NSAIDs and chiral carboxylic acid enantiomers was 1.4-7.6 mu mol/L. The inter-day and intra-day assay precisions were all <6.77% and the mean recoveries (%) of the NSAIDs and chiral carboxylic acids from the spiked human plasma were 95.27-101.12%. Use of negative capacitance to simulate the electrical characteristics in double-gate ferroelectric field-effect transistors

The surface potential and drain current of double-gate metal-ferroelectric-insulator-semiconductor (MFIS) field-effect transistor were investigated by using the ferroelectric negative capacitance. The derived results demonstrated that the up-converted semiconductor surface potential and low subthreshold swing S = 34 (<60 mV/dec) can be realized with appropriate thicknesses of ferroelectric thin film and insulator layer at room temperature. What’s more, a reduction gate voltage about 260 mV can be reached if the ON-state current is fixed to 600 mA/mm. It is expected that the derived results can offer useful guidelines for the application of low power dissipation in ongoing scaling of FETs.

With the onging trend of weight saving in automobiles, the application of light alloys is increasing. Recently, aluminum powder metallurgy has been the subject of renewed attention due to the combination of lightweight of aluminium and the efficient material utilisation of the powder metallurgical process, which offer attractive benefits to potential end-users. This study is to explore the use of warm compaction process to aluminium powder metallurgy. This paper presents a detailed study of the effect of warm compression and sintering conditions on the resultant microstructures and mechanical properties of Al-Cu-Mg-Si PM blend.

Associations of T→A Mutation in the Promoter Region of Myostatin Gene with Birth Weight in Yorkshire Pigs

A T$\longrightarrow$A mutation in the promoter region of porcine myostatin (MSTN) gene has been identified in our previous work. This study analyzed the associations of the myostatin genotypes (TT, TA) caused by this mutation with birth weigh in Yorkshire pigs. Data from 211 unrelated individuals were collected three times from one breeding farm. Detections of the mutation were carried out by PCR-RFLPs approach. The effects of MSTN genotypes (TT and TA) on birth weight were compared by least square means. The results showed that for birth weight of Yorkshire pigs, individuals with TA genotype were significantly higher than those with TT genotype (p<0.05), and the birth weight for pigs with TA genotype were 1.37 kg in average but only 1.25 kg for pigs with TT genotype, indicating a positive effect of birth weight for A allele.

Reconfigurable all-optical code translation in spectrally phase-coded O-CDMA

Reconfigurable all-optical code translation are demonstrated in a spectrally phase-coded optical-code-division multiple-access (O-CDMA) testbed with an interference user. For both one-stage and two-stage code translations, less than 0.9-dB power penalties are induced at each code translation. Multistage code translations are investigated via simulation and experimental emulation in a loop pulse shaper to show the potential application of the proposed method for up to several tens of code translations.
