A high cell density culture system for the anchorage dependent CHO cells was developed based on the combination of in removal of ammonium ion and microcarrier culture system, and semi-fed-batch feeding of glucose and glutamine was employed to the developed culture system. The glass bead was selected as an optimum microcarrier in terms of cell growth. An ammonium ion selective zeolite, Phillipsite-Gismondine, was packed in a dialysis menium ion. The semi-fed-batch operation was employer to the novel culture system for the high density cell culture, and the results showed the cell growth was improved by 32% and tPA productivity by 250%.

<P><B>Abstract</B></P> <P>We isolated the novel vasoactive marine natural products, (5<I>E</I>,10<I>E</I>)-14-hydroxy-2,6,10-trimethylpentadeca-5,10-dien-4-one (<B>4</B>) and sargachromenol D (<B>5</B>), from Sargassum siliquastrum collected from the coast of the East Sea in South Korea by using activity-guided HPLC purification. The compounds effectively dilated depolarization (50mMK<SUP>+</SUP>)-induced basilar artery contraction with EC<SUB>50</SUB> values of 3.52±0.42 and 1.62±0.63μM, respectively, but only sargachromenol D (<B>5</B>) showed a vasodilatory effect on endothelin-1 (ET-1)-induced basilar artery contraction (EC<SUB>50</SUB> =9.8±0.6μM). These results indicated that sargachromenol D (<B>5</B>) could act as a dual antagonist of <SMALL>L</SMALL>-type Ca<SUP>2+</SUP> channel and endothelin A/B<SUB>2</SUB> receptors. Moreover, sargachromenol D (<B>5</B>) lowered blood pressure in spontaneous hypertensive rats (SHRs) 2h after oral treatment at a dose of 80mg/kg dose and the effect was maintained for 24h. Based on our ex vivo and <I>in vivo</I> experiments, we propose that sargachromenol D (<B>5</B>) is a strong candidate for the treatment of hypertension that is not controlled by conventional drugs, in particular, severe-, type II diabetes-, salt-sensitive, and metabolic disease-induced hypertension.</P> <P><B>Graphical abstract</B></P> <P>Sargachromenol D (<B>5</B>) acts as a dual antagonist of L-type Ca<SUP>2+</SUP> channel and endothelin A/B<SUB>2</SUB> receptors.</P> <P>[DISPLAY OMISSION]</P>
폐결핵 환자의 활동성 판정에 대한 67 Gallium 폐주사와 HRCT 의 진단적 가치비교

Objective: The activity of pulmonary tuberculosis has been usually detarmined by smear & culture of AFB in sputum and serial chest X ray. However it is sorntimes difficult to determine the activity of pulmonary tuberculosis by either. We have studied Gallium lung scan 4 HRCT in the patients with pulmonary tuberculosis to determine its activity and compared their diagnostic significance. Methods: 67Gallium lung scan & HRCT were examined in 63 patients with pulmonary tuberculosis suspected by chest X ray and were compared each other with their possible diagnostic significance in determination of activity. The active pulmonary tuberculosis was defined by positive AFB smear and/ or culture in sputum and changes of findings in serial chest X ray. Results: 1) Of 63 patients with pulmonary tuberculosis, 37 patients has been diagnosed active 9 patients were diagnosed by positive AFB smear in sputum, 20 patients by positive AFB culture in sputum and 8 patients by changes of findings in serial chest X ray. 2) In the 37 patients with active pulmonary tuberculosis, chest X ray revealed active pulmonary tuberculosis findings in 28 patients, inactive in 4 patients and undetermined in 5 patients. On the contrary, In the 26 patients with inactive pulmonary tuberculosis, the chest X ray revealed active pulmonary tuberculosis findings in 6 patients, inactive in 16 patients and undetermined in 4 patients. 3) 67Gallium lung scan showed positive uptake in 34 patients of the 37 patients with active pulmonary tuberculosis, but did not show any uptake at all in 26 patients with inactive pulmonary tuberculosis. 4) HRCT showed active findings in 36 patients of the 37 patients with pulmonary tuberculosis. And it showed active findings in 4 patients of the 26 patients with inactive pulmonary tuberculosis. Conclusion: The Gallium lung scan showed 82% of sensitivity and 8% of false negativity. And HRCT showed 97% of sensitivity and 15% of false positivity. In the aspect of laboratory examinations for the determination of activity in pulmonary tuberculosis the 67Gallium lung scan and the HRCT appeared useful.
