      • Aspergillus awamori와 Zymomonas mobilis로 구성된 고정화혼합 배양계의 에탄올 생산에 미치는 Triton, PVA 및 PEG의 영향

        The effects of triton-X 100, PVA and PEG on the ethanol production and raw starch hydrolysis by co-immobilized A. awamori and Z. mobilis (A-Z system) were investigated. When gel beads containing A. awamori(1.25 × 10 exp (9) spores/ℓ - gel) and Z. mobilis(0.5g cell/ℓ - gel) were cultured in 2% raw starch solution at 30℃ for 120 hours, ethanol concentration, ethanol yield (Y_p/s) and pH of culture broth were 2.8g/ℓ, 0.16 and 3.8~4.0, respectively. Ethanol production and starch hydrolysis by addition of PVA was efficient, compared to that of PEG and ethanol concentration in culture broth with 0.2% PVA was 3.56g/ℓ. At A-Z 36 culture system with changing silicon check valve for cotton plug at 36 hours in A-Z culture system, ethanol yield on glucose, Y_p/s = 0.36, showed 2 times higher than that of control culture system(cotton plug culture). Ethanol production and productivity by cultivating 85g gel beads at 30℃ in a 1ℓ jar fermentor(220rpm) of medium 500㎖ were 32g/ℓ and 1.8g/ℓ/day, respectively. After 12 hours of cultivation, gel beads were distinguished oxygen-rich surface for A. awamori from oxygen-deficient central part for Z. mobilis.

      • Xanthomonas sp. EPS-1의 Exopolysaccharide생산을 위한 무기염류의 영향

        For the production of high viscosity exopolysaccharide from Xanthomonas sp. EPS-1 which isolated and identified from soils, optimum concentration of inorganic salts were investigated : 0.2% KH_2PO_4, 0.05% MgSO_4·7H_2O, 0.3% NaCl, 0.005% CaCO_3, 0.007% FeSO_4·7H_2O, 0.005% MnSO_4·7H_2O. When EPS-1 strain was cultivated at 30℃ for 72 hours in a jar fermentor (impeller speed 300 rpm, aeration rate 3 vvm, 2 liter), polymer content, product rate and yield(Y_p/s) was 18.4g/ℓ, 0.26g/ℓ/h and 37%, respectively.

        미용업 종사자들의 피부, 호흡기 및 근골격계 자각증상에 관한 유병률

        Objectives: Present study was conducted to evaluate work-related symptom prevalence among hairdressers. Methods: Exposed group comprised 184 employee employed 73 hair salons in 6 district of Pusan city, and non-exposed group comprised 119 people living recent apartments. A trained interviewer interviewed them with organised questionnaire which included dermatologic, respiratory and musculo-skeletal symptoms. Results : Prevalence of hand eczema was 28.3% in exposed group, and 5.8% in non-exposed group. Adjusted odds ratio for age and atopy history was 4.30(2.34-7.93). Prevalence of respiratory symptom (coughing) in exposed group was 22.1%, and 9.4% for non-exposed group. Adjusted odds ratio for coughing which was adjusted for age, smoking and atopy history was 2.76(1.32∼5.78). Prevalences of musculo-skeletal symptoms among exposed group were neck(59.9%), shoulder(76.6%), upper back(41.2%), lower back(72.2%), arm and elbow(31.3%), wrist(44.2%), finger(35.0%), leg(71.1%). Adjusted Odds Ratios for musculo-skeletal symptoms which was adjusted for age were neck 2.13(1.29∼3.51), shoulder 2.52(1.50∼4.24), upper back 1.71(1.01∼2.88), lower back1.78(1.06∼2.99), arm and elbow 3.10(1.62∼5.94), wrist 2.09(1.23∼3.57), finger 4.83(2.41∼9.68), leg 3.46(2.07∼5.79). Conclusions : These results show that employees in hair salon are likely to have high risk for work-related dermatologic, respiratory and musculo-skeletal symptoms and diseases. Hence, prevention methods from those work-related diseases are required to be developed. Also, the scope of occupational and environmental medicine should be expanded to service area including hairdressers.

        Aspergillus niger SFN-416으로부터 생산한 Xylanase 2의 분리정제 및 특성

        Aspergillus niger SFN-416으로부터 생성된 xylanase Ⅱ를 분리·정제하여 특성을 조사하였다. Aspergillus niger SFN-416의 배양액을 ethanol(70%) 침전, (NH_4)_2SO_4(30~90%) 침전, Sephadex G-100 chromatography 및 DEAE-Sephacel chromatography 등의 정제과정을 거친 결과, specific activity가 38.2 units/ mg protein으로 4.3배 정제되었고, 정제효소의 최적 활성온도는 50℃였다. 최적 pH는 5.5이었고, pH 4.5~6.0 사이에서 비교적 안정하였다. 또한 금속이온에 대한 효소의 활성은 대부분 억제를 보였고, 특히 Hg^2+에서는 3.7%로 강하게 효소활성이 저해됨을 보였고, Fe^2+에서는 100%의 상대활성을 나타내었다. 정제 효소의 분자량은 SDS-PAGE에 의하여 42,000 daltons이었으며, 유기용매에 대한 활성은 10%의 methanol, ethanol, isopropanol 및 1-butanol에 대하여 모두 낮은 활성을 나타었다. Xylanase (EC 3.2.1.8) was purified approximately 4,3 fold from Aspergillus niger SFN-416 by ammonium sulfate fractionation, Sephadex G-100 gel filtration and DEAE-Sephacel ion exchange chromatography. Molecular weight of the enzyme was approximately 42,000 daltons. The optimum pH and temperature of the enzyme activity were 5.5 and 50℃, respectively. The enzyme activity was enhanced by Fe^2+, and inhibited by Hg^2+. The activity was decreased by addition of methanol, ethanol, isopropanol and 1-butanol at a concentration of 10%(v/v).

        고주파유도 가열에 의한 나노구조 WC와 WC-TiAl 급속소결 및 기계적 성질

        In the case of cemented WC, Ni or Co is added as a binder for the formation of composite structures. However, the high cost and the low hardness of Ni or Co, and the low corrosion resistance of the WC-Ni and WC-Co cermets have generated interest in alternative binder phases recently. In this study, TiAl was used as a novel binder and consolidated by the high frequency induction heated sintering (HFIHS) method. Nanopowders of WC and TiAl were made by high energy ball milling. Highly dense WC and WC-10 vol% TiAl with a relative density of up to 99 % were obtained within one min by HFIHS under a pressure of 80 MPa. The addition of TiAl to WC enhanced the toughness without great decrease of hardness due to the crack deflection and nanostructured phase.

        펄스전류 활성 소결에 의한 나노구조 TiC-TiAl 제조 및 기계적 성질

        TiC-Co or TiC-Ni hard materials have been used for cutting tools. However, the high cost and the low hardness of Ni or Co, and the low corrosion resistance of the TiC-Ni and TiC-Co cermets have generated interest in recent years in using them as alternative binder phases. In this study, TiAl was used as a novel binder and consolidated by the pulsed current activated sintering(PCAS) method. Nanopowders of TiC and TiAl were fabricated using high energy ball milling. Highly dense TiC-TiAl hard materials with a relative density of up to 99.5% were sintered within three min by PCAS. Not only the hardness but also the fracture toughness of the TiC-10 vol%TiAl were better than those of TiC-10 vol%Ni or TiC-10 vol%Co. (Received December 14, 2015; Accepted March 20, 2016)

        누에의 품종육성을 위한 유전적 기초정보를 얻고자 7개 품종을 이면교잡하여 얻은 F₁42개 조합을 재료로 각 형질에 대한 조합능력 및 형질상호간의 상관관계를 산출한 결과는 다음과 같다. 1. GCA와 SCA는 모든 형질에서 유의하였고, RCA는 암견층비율과 화용비율이 유의하지 않았다. 모든 형질은 GCA가 SCA나 RCA보다 높았다. 2. GCA 효과는 N74가 화용비율에서, S1이용체중에서, 잠 108이 5령경과, 암수단견중, 암수견층중, 암수견층비율, 발아일수에서, N63이화용비율과 발아일수에서, 잠 124가 전령경과, 5령경과, 암수단견중, 암수견층중, 암수견층비율, 수용체중에서 각각 正으로 높았다. 3. SCA효과는 형질 및 교배조합에 따라 다양하였으나 2×7(S1×잠 124)조합이 암수단견중, 암수견층중, 암수용체중에서, 3×4(잠 107×잠 108)조합이 암수단견중, 수견층중, 암수용체중, 화용비율에서, 3×7(잠 107×잠 124) 조합이 수단견중, 암수견층중, 암수견층비율, 수용체중, 화용비율에서 각각 正으로 높았다. 4. RCA효과는 1×5(N74×C51)조합이 전령경과, 5령경과에서, 3×4(잠 107×잠 108)조합이 암수견층중, 수견층비율에서, 6×7(N63×잠 124)조합이 수단견중, 암수견층중, 수용체중에서 각각 正으로 높았다. 5. 유전력은 전령경과, 5령경과, 암수단견중, 암수견층중, 암수견층비율이 85.79% 이상으로 높았다. 6. 상관관계는 전령경과와 수단견중 ·암수견층중 ·암수견층비율 ·발아일수에서, 암단견중과 수단견중 ·암수견층중 ·수견층비율 ·암수용체중에서, 수단견중과 암수견층중 ·암수견층비율 ·암수용체중에서, 암견층중과 수견층중 ·암수견층비율 ·암수용체중 ·발아일수에서, 수견층중과 암수견층비율 ·수용체중 ·발아일수에서, 암견층비율과 수견층비율 ·수용체중 ·발아일수에서, 수견층비율과 발아일수에서, 암용체중과 수용체중에서 正으로 유의하였다. Combining ability and correlation coefficient for F₁population form diallel cross in the silkworm were estimated for twelve quantitative characters i. e., total instar period, fifth instar period, female of cocoon weight, male of cocoon weight, female of cocoon layer weight, male of cocoon layer weight, female of cocoon layer ratio, male of cocoon layer ratio, female of pupal weight, male of pupal weight, percentage of pupation and eclosion period. The results of combining ability test, it was found that general combining ability (GCA) and specific combining ability(SCA) were significantly different among twelve quantitative characters, and the variance of the former was higher than that the latter. Variety Jam 108 and Jam 124 was found to be the useful parent based upon the GCA effect considering the cocoon yield and yield components. the crosses of S1×Jam 124, Jam 107×Jam124 showed the most promising SCA effects on cocoon yield and yield components. In reciprocal combining ability(RCA) effect, hybrids in Jam 107×Jam 108, N63×Jam 124 were exhibited positively high for yield component. The heritabilities of larval period, cocoon weight, cocoon layer weight and cocoon layer ratio were higher than those of pupal weight, percentage of pupation and eclosion period. There were positive coefficients; between the character of total instar period and the character of cocoon layer weight, or cocoon layer ratio ; between the characters of cocoon weight and the character of cocoon layer weight or cocoon layer ratio, or pupal weight ; between the character of cocoon layer weight and the character of cocoon layer ratio, or eclosion period. Moreover highly significant values were found between the character of cocoon yield and the character of cocoon layer weight or cocoon layer ratio, or pupal weight.

        급속 가열에 의한 나노구조의 AlCr2-Al2O3 복합재료 합성과 소결 및 기계적 특성

        A dense nanostuctured AlCr2-Al2O3 composite was sintered by pulsed current activated heating up to 1200 °C within 2 min from mechanically milled powders. The advantage of this process is that it allows very quick densification to near theoretical density and prohibition of grain growth in nanostructured materials. A highly dense AlCr2-Al2O3 composite with a relative density of up to 96% was produced under application of a 80 MPa pressure and a pulsed current of 2000 A. The fracture toughness and hardness of the AlCr2-Al2O3 composite in this study was better than those of previous studies due to the grain refinement. The microstructure and mechanical properties of the composite were investigated using field emission scanning electron microscopy and a Vickers hardness tester. (Received November 24, 2015)
