      • Enhanced Maturation and Function of Dendritic Cells Using Hydrogel Coated Plate and Antigen Electroporation

        <P>Dendritic cells (DCs) are potent antigen-presenting cells that can be matured <I>in vitro</I> from immature dendritic cells (iDCs) in the presence of several biological agents such as cytokine cocktail, CD40L, TNF-a and antigen loading, which are necessary and achieved using various protocols, such as lipofection, passive pulse or electroporation. However, these DCs maturation protocols may cause with a significant loss of cells because of cellular attachment and spreading during culturing. Some biomaterials that influence adhesion and development of cells have been used in cell culture techniques, and it was thought that they might be applied on the culture of DCs. In this study, we used polyHEMA, which is a hydrogel coating biomaterial that prevents DCs from adherence, and investigated whether hydrogel coating affects the maturation of iDCs. The efficiency in the generation of mDCs was improved through hydrogel coating procedure and a dendritic cell maturation marker, CD83, was significantly increased in hydrogel-coated culture condition. The antigen-loaded mDCs from electroporation were further expressed the CD83. The mDCs generated in the hydrogel-coated culture condition showed more, longer and thicker dendrites, and produced more amounts of cytokines such as IL-12 and IFN-γ. Therefore, it was suggested that the hydrogel-coated culture condition could improve function of mDCs.<fn><P>Cheol-Hun Son and Jae-Ho Bae contributed equally to this work.</P></fn></P>

        사례보고 : 하수오 복용 후 발생한 재발성 독성 간염 1예

        Toxic hepatitis has been reported as a major cause of acute hepatitis, but its potential induction by herbal remedies and/or health foods is usually neglected. We experienced a case of toxic hepatitis associated with Polygoni multiflori, a Chinese herb commonly known as Ho-Shou-Wu. A 54-year-old woman consumed Ho-Shou-Wu for 1 month, after which she experienced fatigue and overall weakness. A diagnosis of toxic hepatitis was made based on her clinical history, the findings for viral markers and other laboratory data, and ultrasonography. Her condition improved considerably after she stopped taking Ho-Shou-Wu. However, she resumed taking Ho-Shou-Wu immediately after discharge from hospital, which aggravated her symptoms and liver function. She was immediately readmitted and stopped taking Ho-Shou-Wu. Her relapse into hepatitis immediate after resuming consumption of the herb is strongly indicative of the validity of Koch`s postulate in this case.

        열공형과 비열공형 피질하 혈관성 치매에서 위험인자의 차이에 관한 비교 연구

        Backgrounds and Objectives: Vascular dementia is a group of dementing disoders arising from various stroke syndrome. Among these. subcortical ischemic vascular dementia (SIVD) is regarded as a relatively distinct clinical entity. However, MRI patterns of SIVD are not homogenous. In some patients, lacunes are dominant, and in others, subcortical white matter changes are. This study was designed to compare risk factor profiles between SIVD with and without multiple lacunes. Methods: We divided 47 subjects (22 males, mean age. 68 years) recruited from VADAPET (Multicenter Trial For Evaluation Of The Changes In the PET Images Of Subcortical Vascular Dementia Patient) study into two groups one with more than 5 lacunes in deep gray matter (lacune group) and the other with 5 or less(non-lacune group) Clinical characteristics and laboratory findings of two groups were compared. Results: Nineteen of 47 patients (40%) belonged to the lacune group. The lacune and non-lacune groups d d not differ in the following variables: age, hypertension, diabetes mellitus, hyperlipidemia heart disease, history of stroke or TIA, history of trauma or major surgery, family history of hypertension stroke, or dementia, age at diagnosis of dementia, body mass index, white blood cell count, ESR, CRP, fibrinogen, hemoglobin A1C, total cholesterol. LDL cholesterol creatinine, proteinuria, glucosuria, and microhematuria. However, male sex, smoking alcohol. hemoglobin, and HDL cholesterol were possibly associated more with lacune group SIVD than with non-lacune group (p<0 1) Multivariate analyses revealed that smoking, hemoglobin, and HDL cholesterol were independent predictors of SIVD with multiple lacunes Conclusion: Our study suggests that SIVD with multiple lacunes may be significantly different in smoking habits hemoglobin, and HDL cholesterol from SIVD without multiple lacunes.

      • Perovskite형 복합산화물 촉매를 이용한 희박연소 엔진의 NOx 배기가스처리

        Perovskite type mixed metal oxide which widely used in superconductor, semiconductor, and magnetic material was adapted lean burn deNOx catalyst by its own oxygen vacancy and electromagnetic character. Prepared perovskite type catalysts were divided roughly ?? and Lanthanide base perovsite(??). After characterization whether its structure was perovskite or not, then NOx removal activity was tested. And, in order to test NOx removal activity at real automotive exhaust condition, we accompanied engine dynamomter test. Most perovskite superior to zeolite which was being researched for lean burn deNOx catalyst. Moreover we could identify B site partial substituted catalyst has more high activity and lowest ??. Different from the porous catalyst, oxygen vacancy which origin from catalystic activity of perovskite catalyst was evaluated and associated with NOx removal activity. Then we could find out they have linearized corelationship. In kinetical consideration, CO adsorption order was similar between fresh catalyst and aged catalyst. But, fresh catalyst in 2.4 times CO adsorption rate constant than aged catalyst.

      • C6 glia 세포에서 유도성 Nitric Oxide Synthase 유전자 발현조절에 관한 연구

        To investigate transcriptional regulation of iNOS gene by LPS and cytokines, the production of NO, expression of iNOS mRNA and protein, binding activity of nuclear factor-kappa B(NF-kB), and promoter activity of iNOS gene were examined in rat C6 glial cells. LPS, interferon-gamma(IFN-γ), and tumor necrosis factor-alpha (TNF-α) stimulated the production of NO, which was increased synergistically by co-treatment. By the treatment of LPS, iNOS mRNA expression was initiated at 1 h, markedly increased by 3 h, and decreased gradually afterward. iNOS mRNA expression was markedly enhanced by mixture of LPS, IFN-γ and TNF-α. iNOS protein synthesis was increased by the treatment of mixture LPS and cytokine mixture. Treatment of LPS stimulated NF-kB activation, and the activation reached to the maximum level at 30 min, and the treatment of mixture of LPS and cytokines increased the activation. To determine the effect of NF-kB binding activity on iNOS promoter activation, CAT assay was performed. iNOS promoter activity was increased by the treatment with LPS for 5.5 h, and further increased by the combined treatment with LPS and cytokines. These results suggest that NF-kB activation by LPS and cytokines may play a significant role in the induction of the iNOS gene.

        목적: 림프관종은 흔한 질환이 아니므로 보고된 례들도 많지 않아 그 임상적 특성이 매우 다양하게 보고되어 있다. 저자들은 최근 2년 6개월간 14례의 림프관종을 경험하였기에 림프관종의 임상적 특성을 분석하는 것이 의의가 있을 것으로 판단하였다. 방법: 1993년 9월부터 1996년 3월까지 이화여자대학교 의과대학부속 목동병원에서 치료받았던 14례의 림프관종에 대하여 임상기록을 통한 후향적 분석을 하였다. 임상적 특성은 증상, 종양의 위치, 지난방법, 종양의 크기, 치료방법 그리고 합병증 및 재발여부에 관하여 분석을 하였다. 결과: 1) 림프관종 발견 연령분포는 4주 이내가 4명, 4주 이상 2세 이하가 4명이었고 2세 이상 10세 이하가 3명이었으며 남자가 6명, 여자가 8명으로서 남녀비가 1 : 1.3 이었다. 2) 증상으로는 신체 각 부위의 종괴로 나타나는 경우가 71.4%로서 가장 많았다. 3) 발생부위는 전흉벽과 액와부에 가장 많이 발생하였다. 4) 림프관종의 진단방법으로는 초음파가 유용한 진단적 가치가 있었다. 5) 림프관종의 크기는 5cm 이하가 6례(42.9%), 5cm 이상 10cm 이하가 6례(42.9%), 10cm 이상 15cm 이하가 2례(14.2%)였다. 6) 치료방법의 주류는 완전절제이며 수술후 합병증이나 재발의 례는 없었다. 결론: 림프관종은 종괴의 감별진단에서 꼭 염두를 두어야 하며 특히 소아의 종괴일 경우 림프관종을 감별하여야 한다. Objectives : Lymphangioma is an infrequent disease. Few cases of lymphangioma has been reported, so its clinical characteristics are variale. We had experienced fourteen patients of lymphangioma during relatively short period, two and half years. It was worth to analyze the clinical characteristics of the lymphangioma. Methods : A clinical analysis was made in fourteen patients with lymphangioma who had been admitted to Ewha Womans University Mokdong Hospital from September, 1993 to March, 1996 retrospectively by the review of clinical records. The characteristics were about the symptoms, locations, diagnostic methods, sized of tumor, treatment, and complications. Results : were summerized as following 1) The sex ratio was 1 to 1.3(male to female) and the most prevalent age group was less than 10 years(78.6%) 2) The most common presenting symptom was a mass(71.4%) 3) The locations were trunk and axillary(35.7%), retroperitoneum(21.4%), extremity(21.4%), neck and mesentery in decreasing order. 4) The ultrasonogram was a useful disgnostic method of lymphangioma(71.4%). 5) The size distribution of tumors were 42.9% in less than 5cm, 42.9% in 5 to 10cm, and 14.2% in 10.1 to 15cm. 6) The treatment of choice was complete excision. There was no complication, or recurrence. Conclusion : lymphangioma should be considered as differential diagnosis in patients with mass, especially children.

        Under lean burn condition region, main hazard exhaust components are NOx and abundant hydrocarbons. Therefore, NOx abatement using hydrocarbon reductant was most desirable. During last twenty years, NO direct decomposition reaction by Cu-ZSM-5 was suggested with strong possibilities. However, at actual engine exhaust condition(i.e. hydrothermal atmosphere) that wasn't work its own function. In this paper, we tried in order to overcome these disadvantages, other precious metal, especially Pt, Ir, Rh ion exchanged zeolite catalysts were used for lean deNOx catalysis.

        치아이동에 의한 백서 삼차신경감각핵군내 c-Fos의 발현

        이 연구의 목적은 말초조직에 유해 자극을 가하였을 때 중추 신경계내 이차 신경 세포체내에 발현되어 Neuronal marker로 사용되고 있는 c-fos를 사용한 면역 조직화학법으로 치아이동시 동반되는 동통의 투사경로의 이해에 도움을 주고자하는 것이다. 생후 9 주령의 210gm내외의 Sprague-Dawley계 웅성 백서 21마리를 교정력을 가하지 않고 마취만을 시행한 정상대조군과 교정력 적용 시간 경과에 따라 1시간, 3시간, 6시간, 12시간, 1일, 3일군으로 나누어 각 해당 시간동안 상악 우측 제1대구치와 상악 우측 측절사이에 Ni-Ti coil spring을 결찰하여 30gm내외의 지속적인 교정력을 가한 후 희생시켰다. 희생시킨 백서의 뇌간을 적출하여 토끼의 항체를 이용하여 면역화학 염색을 시행하였다. 삼차신경 감각핵군내 부위에 따른 c-fos 면역 반응 세포를 측정하여 교정력 적용 시간 경과에 따른 변화를 관찰하였다. · c-fos면역 반응 세포의 배측에서의 분포는 자극측 중위핵과 미측핵의 이행부위에서 시작하여 제1경추 척수 후각에까지 이어졌는데 가장 많은 분포를 보인 곳은 미측핵의 문측 부위였다. 그리고 주로 I층 과 II층에서 관찰되었다. · 복측에서의 c-fos 면역 반응 세포의 분포는 자극측 중위핵의 미측 부위에서 시작하여 미측핵의 중간부위에 까지 이어졌다. · 교정력 적용 3,6시간군에서 c-fos면역 반응 세포가 가장 많이 관찰 되었으며 12시간군에서 감소되기 시작하여 1일, 3일군에서는 현저히 감소 하였다. 위로 미루어 볼 때 지속적인 교정력에 의한 동통은 중위핵과 미측핵의 이행부위, 미측핵, 제1경추 척수후각에서 매개되는 것으로 생각된다. The c-fos is known as neuronal marker of second neurons which is activated by noxious peripheral stimulation. To investigate the changes of c-fos expression in the trigeminal nucleus complex during tooth movement, immunohistochemical study was performed. Experimental rats(9 weeks old, 210 gm, 21 rats) were divided into seven groups(normal, 1 hour group, 3 hour group, 6 hour group, 12 hour group, 1 day group, 3 day group). Rats in the normal group were anesthesized without orthodontic force. Rats in the experimental groups were applied orthodontic force (approximately 30 gm) to upper right maxillary molar. Frozen sections of brain stem were immunostained using rabbit antisera. The change of c-fos expression were observed with respect to rostrocaudal distribution, laminar organization, and duration of orthodontic force application. The study results were as follows. · The c-fos nuclei in the dorsal part were observed from ipsilateral transition zone of subnucleus interpolaris and subnucleus caudalis to C1 cervical dorsal horn rostrocaudally. The maixmal peak point was the rostral part of subnucleus caudalis. The greatest proportion of c-fos cells were located within lamina and I and II. · The c-fos nuclei in the dorsal part were observed from the most caudal part of subnuleus interpolaris to the middle part of the subnucleus caudalis. · The number of c-fos immunoreactive dot increased at 1 hour group, reached its maximum at the 3 and 6 hour groups, and showed a decreasing trend after 12 hours. These resutls imply that nociceptive stimulation caused by continuous orthodontic force might be modulated by transition zone of subnucleus interpolaris and subnucleus caudalis, subnucleus caudalis, C1 spinal dorsal horn.

      • 팔영산 삼림식생과 토양환경

        전라남도 고흥군 소재 팔영산 (608.6 m) 일대의 삼림군락에 대한 색생조사를 1993년 10월부터 1995년 7월까지 실시하였다. 조사한 자료는 Z-M 방식에 의해 종조성표를 작성하였다. 이에 따라 삼림군락은 신갈나무군락, 졸참나무군락, 굴참나무군락, 서어나무군락, 소나무군락등 5 개 군락으로 분류되었다. 입지와 환경과의 유기적 관계를 조사하기 위해 토양을 분석한 결과, 신갈나무군락은 토양의 비옥도가 높은 산의 능선 부위에 집중 분포하였으며, 굴참나무군락과 소나무군락은 토양의 비옥도가 낮고 비교적 건조한 고도 400 m 이하 지역에 주로 군락을 형성하고 있었다. 또한 졸참나무군락은 고도 300 - 550 m 이내에서 부분적으로 나타났으며, 서어나무군락은 토양의 비옥도가 양호한 고도 300 - 550 m 사이의 골짜기 주변에 국지적으로 분포하였다. 팔영산 삼림식생의 특징은 상록활엽수림이 형성될 수 있는 지역이나 후박나무, 동백나무, 사스레피나무 등의 몇몇 종을 제외하고는 상록활엽수의 분포를 확인할 수 없었으며, 일부 식재림을 제외한 대부분의 지역이 낙엽활엽수와 소나무의 혼효림으로 구성되어 있었다. The phytosociological study forest vegetation on Mt. Palyoung (608.6 m) was investigated from October 1993 to July 1995 in Koheug-Gun, Cheonnam province. According to Z-M method, the floristic composition table of the forest vegetation was divided into five community as follows; Quercus mongolica community, Quercus serrata community, Quercus variabilis community, Pinus densiflora community, Carpinus laxiflora community. The soil was analyzed to investigated the soil conditions and soil fertility. As a result, the organic matters which showing soil fertility were found to be low Quercus variabilis and Pinus densiflora community at comparatively xerophic conditions, below 400 m altitude, while they were increased in Quercus mongolica community on mountain ridge, and that Quercus serrata community was appeared 300-550 m altitude around mountain stream to moderate soilfertility. The forest vegetation of Mt. Palyoung area unconfirmed capable of evergreen broad-leaved forest, but appeared to Machilus thunbergii, Camellia japonica and Eurya japonica species etc. On the other hand, surveyed site's vegetation was distributed mixed forest of deciduous leaved forest and Pinus desiflora, except for some afforestation.
