        This study investigated the effect of porcelain primer on bonding of composite resin to porcelain surface. In order to test the bond strength between porcelain and composite resin, porcelain cylinders were embedded in acrylic resin, and polished with 240grit silicone carbide paper. The specimens were divided into twelve groups. All specimens were treated with three porcelain primers and bonded with five composite resins. All test groups were stored in 37℃ distilled water for 48hours. Shear bond strengths were measured with Instron(Model 4201) at a cross-head speed of 1㎜/min. The obtained results were as follows : 1. Scotchprime/SiluxⅡ group and BISCO Porcelain Primer/Bisfill group showed significant higher bond strengths than Clearfil Porcelain Primer/Photo clearfil Bright group(p<0.05). And there was no significant differences in bond strengths between Scotchprime/SiluxⅡ group and BISCO Primer/Bisfill group(p>0.05). 2. When composite resins were used with Scotchprime, the bond strengths were decresed SiluxⅡ(16.68±3.35 MPa), Bisfil(16.23±4.54 MPa), Poly-Fill(14.74±4.08 MPa), Photo Clearfil Bright(13.75±2.89 MPa) and Pekalux(14.74±4.08 MPa) in order, but there was no statistical significance(p>0.05). 3. When composite resins were used with BISCO Porcelain Primer, the bond strength were decreased Bisfil(16.17±1.60 MPa), SiluxⅡ(12.13±2.37 MPa), Poly-Fill(10.78±1.99 MPa), Photo Clearfil Bright(9.91±4.59 MPa) and Pekalux(7.36±2.16 MPa) in order, but there was no statistical significance(p>0.05). 4. SiluxⅡ, Photo Clearfil Bright and Poly-Fill used with Scotchprime showed significant higher bond strengths than BISCO Porcelain Primer(p<0.05).

        Epigenetic Modulation of Gene Expression during Keratinocyte Differentiation

        Background: Epigenetic modulation of gene expression occurs by various methods, including DNA methylation and histone modification. DNA methylation of specific genes may affect the chromatin structure, preventing access by the transcriptional machinery. Although gene expression is dramatically changed during keratinocyte differentiation, there is no evidence of epigenetic modulation during the process of epidermal stratification. Objective: We investigated whether epigenetic modulation is involved in keratinocyte differentiation-specific gene regulation. Methods: We used trypsin to produce epidermal fragmentation (named T1-T4) and performed a morphological analysis using hematoxylin-eosin stain and cytokeratin expression based on reverse transcription polymerase chain reaction. We then constructed a DNA methylation microarray. Results: Each epidermal fragment showed morphological features of the epithelial layer. T1 represented the basal layer, T2 was the spinous layer, T3 was the granular layer, and T4 was the cornified layer. The level of the K14 proliferation marker was increased in the T1 fraction, and the level of K10 differentiation marker was increased in the T2-T4 fractions. Using a methylation microarray with the T1 and T4 fractions, we obtained many hypermethylated and hypomethylated genes from differentiated keratinocytes. Conclusion: The importance of epigenetic modulation in target gene expression during keratinocyte differentiation is identified.

        급성 미만성 원형 탈모증 14례

        Background: Acute diffuse alopecia areata (AA) was described as a unique AA which mimics anagen effluvium, or in which the initial hair loss is diffuse and followed by total denudation of scalp within several weeks or months. In spite of its peculiar clinical feature, there was neither further characterization nor known effective treatment for this form of AA. Objective: To evaluate the characteristic clinical findings of acute diffuse AA and confirm the effect of high dose methyl prednisolone therapy. Methods: The medical records of 13 patients with acute diffuse AA between January 2002 and April 2006 at the Department of Dermatology, Chungnam National University Hospital were reviewed. All patients were treated with high dose methylprednisolone therapy. Results: Of the 13 patients who completed the study, 8 patients (61.5%) were male and 5 patients (38.5%) were female. The mean age was 29.6 years old. The progress of hair loss stopped 2.4 weeks on average after initial treatment and newly emerging hairs were recognized 4.1 weeks on average after initial treatment. 84.6% (11/13) of patients showed terminal hair growth, and 46.2% (6/13) of patients completely responded to this therapy. Conclusion: Acute diffuse alopecia areata can occur in male as well as in female patients. High dose methyl-prednisolone therapy appears to be effective in patients with rapidly progressing acute diffuse alopecia areata to prevent the progression of the disease.         The inhibitory effect of A20 on the inflammatory reaction of epidermal keratinocytes

        <P>A20 is a negative regulator of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-kappa B) signaling, and has been implicated in the pathogenesis of psoriasis through genome-wide association study (GWAS). In the present study, we investigated the putative role of A20 in epidermal keratinocytes. Immunohistochemical analysis showed that A20 was expressed in all layers of the epidermis, with an increasing pattern in the upper layers. In our model of calcium-induced keratinocyte differentiation, A20 expression was increased in a time-dependent manner. To investigate whether A20 affected keratinocyte differentiation, we overexpressed A20 in cultured keratinocytes. As a result, we noted that A20 overexpression did not affect keratinocyte differentiation, suggesting that A20 is not a direct modulator of keratinocyte differentiation. Interestingly, we found that A20 levels were decreased in psoriatic lesional skin compared to non-lesional areas. To investigate whether A20 played a role in the innate immune response of keratinocytes, we overexpressed A20 and then examined poly(I:C)-induced cytokine expression. We noted that A20 significantly inhibited poly(I:C)-induced cytokine production, and this effect was related to the inhibition of NF-kappa B signaling. These results suggest that the downregulation of A20 increased the susceptibility of keratinocytes to external stimuli, thus contributing to the development of psoriasis.</P>

        도재표면처리제가 복합레진과 도재와의 결합에 미치는 영향

        신규 백금착물 항암제 KBP31705-C127 , KBP30603-901 의 Clsplatn 및 Carboplatin 과의 약동력학적 동태 비교

        The present study examined pharmacokinetic profiles of KBP31705-C127 and KBP30603-901, new platinum coordination complexes synthesized as anticancer candidates, in comparison with two well-known platinum-containing anticancer agents, cisplatin and carboplatin in rats. Under sodium pentobarbital anesthesia of male Sprague-Dawley rats, urinary bladder, and femoral artery and vein were catheterized for urine collection, blood sampling and drug injection, respectively. Following i.v, administration of cisplatin (2 ㎎/㎏), KBP31705-C127 (2 ㎎/㎏), carboplatin (20 ㎎/㎏) or KBP30603-901 (20 ㎎/㎏), blood samples were collected at 2, 4, 6, 8, 10, 15, 20, 30, 45, 60 and 120 minutes. Urine samples were collected at 1-hr interval for 4 hr. Platinum concentrations in plasma and urine were measured using an inductively coupled plasmamass spectrometer. The plasma concentration-time curves were biphasic for all drugs during the time period studied. Compared with cisplatin, KBP31705-C127 showed similar decay patterns in the alpha- and beta-phases with slightly lower plasma concentrations. Urinary platinum excretion for cisplatin and KBP31705-C 127 was 56 and 52% of the administered dose in 4 hr, respectively. With regard to carboplatin and KBP30603-901, a similar decay pattern was also observed in the alpha-phase. The half life of KBP30603-901 in the beta-phase, however, was much longer than that of carboplatin, which was consistent with the urinary excretion results that 46 and 59% of the administered dose were excreted in the urine in 4hr, respectively. The results suggest that platinum coordination complexes are primarily excreted via the renal route and KBP30603-901 can elicit longer duration of action due to slower renal excretion compared to carboplatin.

        일부 지역주민의 비만 수준과 비만 관련요인

        Objective: The aim of present study was to assess the distribution and correlates of obesity in a Korean rural people using both body mass index(BMI) and body far percent. Methods: A total of 1,243 participants were recruited using a two-staged stratified sampling. A structured questionnaire was used to ask their sociodemographics (gender, age, marital status, educational background, and etc.) and health-related behaviors (smoking, drinking, and regular exercise etc.). The data of weight-for height, and body fat percent were also collected by physical examination. POI' the estimation and analysis of con-elates of obesity, we used BMI(≥25kg/㎡) and body fat percent(male≥25%, female≥30) as a cut-point of obesity. All analyses were stratified to three age groups(<20,20-39,40≥). Results: The prevalence of obesity in this study was higher than that in previous studies. This study showed that 32.6% (male: 33.7%, Female: 31.7%) of participants according to BMI, and 45.6% (male: 43.8%, female: 47.4%) of them according to body fat percent were obese group. Logistic regression analysis showed that, in the criteria of BMI, while obesity was associated with female and low educational background under the age of 40, prevalence of obesity was higher in non smokers over the age of 40. In case of body fat percent crteria, single(unmarried, divorced, separated, widowed) were more likely to be obese in male aged 20 to 39 years. In the group aged 40 years and over, risk of obesity was higher in female than in male. Conclusion: This result suggests that obesity is common in Korean rural areas, especially among adolescents and female aged 40 years or over, and the risk factors for obesity were different by age. It is required that health management program focusing on obesity and its adverse outcomes should be developed in a community setting.
