De-novo Hybrid Protein Design for Biodegradation of Organophosphate Pesticides

In the present investigation, we attempted to design a protocol to develop a hybrid protein with better bioremediation capacity. Using in silico approaches, a Hybrid Open Reading Frame (Hybrid ORF) is developed targeting the genes of microorganisms known for degradation of organophosphates. Out of 21 genes identified through BLAST search, 8 structurally similar genes (opdA, opd, opaA, pte RO, pdeA, parC, mpd and phnE) involved in biodegradation were screened. Gene conservational analysis categorizes these organophosphates degrading 8 genes into 4 super families i.e., Metallo-dependent hydrolases, Lactamase B, MPP and TM_PBP2 superfamily. Hybrid protein structure was modeled using multi-template homology modeling (3S07_A; 99%, 1P9E_A; 98%, 2ZO9_B; 33%, 2DXL_A; 33%) by $Schr{\ddot{o}}dinger$ software suit version 10.4.018. Structural verification of protein models was done using Ramachandran plot, it was showing 96.0% residue in the favored region, which was verified using RAMPAGE. The phosphotriesterase protein was showing the highest structural similarity with hybrid protein having raw score 984. The 5 binding sites of hybrid protein were identified through binding site prediction. The docking study shows that hybrid protein potentially interacts with 10 different organophosphates. The study results indicate that the hybrid protein designed has the capability of degrading a wide range of organophosphate compounds.

Zinc oxide (ZnO) nano flowers uniformly attached to the surface of Molybdenum disulfide (MoS<SUB>2</SUB>) sheet were successfully prepared via a simple facile one pot hydrothermal method. The morphology and microstructure of the prepared samples was characterized using field - emission scanning electron microscopy (FE-SEM), Transmission electron microscopy (TEM), X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FT-IR), and photoluminescence (PL) spectroscopy. The Brunauer-Emmett-Teller (BET) surface area was determined using physical and chemical analyzer. The characterization results showed that the flowers like ZnO micro-structures were uniformly attached to the surface of MoS<SUB>2</SUB>. The as-prepared MoS<SUB>2</SUB>/ZnO composite showed enhanced photocatalytic activity and considerable stability for the photo degradation of phenol red as compared with pure MoS<SUB>2</SUB> and ZnO under both UV and visible light irradiation. Such an enhancement of photo degradation capability was ascribed to the synergistic effect between the flowers-shaped ZnO particles and sheet-like structure of MoS<SUB>2</SUB>, which boost the migration efficiency of photo-induced electrons and hinder electron-hole recombination rate. It was also found that the amount of MoS<SUB>2</SUB> in the composite has a significant effect on the degradation activity. Moreover, the antibacterial activity test towards the gram negative Escherichia-coli showed that the MoS<SUB>2</SUB>/ZnO composite has better antibacterial properties compared to pure ZnO and MoS<SUB>2</SUB>.

Objectives This objective of the study was to report the prevalence of internet addiction, sleep quality, depression, anxiety, and stress in undergraduate medical students. Methods A cross-sectional, questionnaire-based study was conducted among 221 undergraduate medical students at Government Doon Medical College, Dehradun, Uttarakhand. Data pertaining to internet addiction, sleep quality, and depressive symptoms were also collected using validated and reliable questionnaires (Young Internet Addiction Test, Pittsburgh Sleep Quality Index, and Depression Anxiety Stress Scale 21). Results The prevalence of poor sleep quality, severe anxiety, and severe depression was 33.9%, 7.3% and 3.6%, respectively. The place of residence was significantly associated (p = 0.006) with internet addiction. The mean Young Internet Addiction Test score was higher in students residing in hostels compared with students staying with families. Stress was associated with age. The mean stress score was higher in the age group 17–20 compared with the 21–24 age group. Stress and depression were independent predictors of sleep quality. Conclusion Quality sleep is the key for good health. Based on limited samples, this study showed that poor sleep quality was associated with stress and depression. Hence, continuous counselling is suggested for supporting students managing their stress and depression.

<P><B>Abstract</B></P> <P>Mesoporous architectures are remarkable electrode materials for energy storage system due to their large number of active sites and high surface area. Here we report, mesoporous MoS<SUB>2</SUB> particles (pore diameter 34.04 nm) well attached to the surface of thin layered reduced graphene oxide (rGO) via an ultrasonic chemical method for supercapacitor applications. The rGO not only increases the conductivity of MoS<SUB>2</SUB> but also provides a substrate for the attachment of MoS<SUB>2</SUB> with low aggregation. The porous MoS<SUB>2</SUB> provides a large surface area and sufficient way for the fast transport of electrolyte ions toward electrode materials. As a result, the synthesized MoS<SUB>2</SUB>/rGO composites exhibited excellent electrochemical performance with a specific capacitance 314.5 F/g in 2M KOH aqueous solution at a scan rate of 10 mV/s and excellent specific capacitance retention (80.02%) after 1000 cycles in a three electrode system for energy storage applications.</P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

여러가지 긴 사슬 지방산 하이드라지드는 방향족과 헤테로 고리 알데히드와 반응하여 알코올 용매하에서 2- hydroxy benzylidene와 새로운 모기 파라-페로몬인1H-indol-3-ylmethylene hydrazides을 얻었다. 마이크로파의 조사기 술과 마찬가지로 전통적인 방법에 의한 다양한 새로운 긴고리 지방산 하이드라지드 (2-hydroxy benzylidene와 1Hindol- 3-ylmethylene)의 합성방법도 보고한다. 이 화합물들의 구조는 FTIR, NMR & MS와 같은 분광학적 기법에 의해 증명되었다. 이 화합물의 전자충격질량스펙트럼 분쇄 패턴의 몇 가지 재미있는 특징도 논의했다. Various long chain aliphatic acid hydrazides react with aromatic and heterocyclic aldehydes in alcoholic medium in refluxing conditions to give corresponding 2-hydroxy benzylidene and 1H-indol-3-ylmethylene hydrazides, a newer class of mosquito para-pheromones. We describe here synthesis of various novel long chain aliphatic acid (2- hydroxy benzylidene and 1H-indol-3-ylmethylene) hydrazides by conventional as well as microwave irradiation techniques. The structures of these compounds have been confirmed by spectroscopic techniques (FTIR, NMR & MS). Some of the interesting features of the electron impact mass spectral fragmentation pattern of these compounds have also been discussed.

The vitamin D receptor (VDR) gene serves as a good candidate gene for susceptibility to essential hypertension. The gene regulates the renin angiotensin system by influencing blood pressure regulation. Around 3% of the human genome is regulated by the vitamin D endocrine system. Several studies have reported mixed results with respect to relationship of VDR gene and hypertension. Observational evidence supports the concept that vitamin D plays a role in the pathogenesis of cardiovascular disease and arterial hypertension which is further supported by meta-analysis and case control studies reporting how VDR polymorphism leads to the onset and development of hypertension. In this review, we summarize the existing literature on the link between VDR and hypertension, including mechanistic studies, observational data, and clinical trials showing relationship of vitamin D level and hypertension with a focus on recent findings related to genetic studies that showed the relationship of VDR gene polymorphism with vitamin D level in hypertensive and normotensive groups. As a result, determining the association of VDR polymorphisms with essential hypertension is expected to aid in the risk assessment for the condition.
Effect of sinusoidal cylindrical surface of PCM on melting performance

A numerical analysis was conducted to examine the effect of capsule shape on phase change material (PCM) melting speed. The surface parameters of the irregular capsule shape were amplitude a and the number of sinusoidal undulations n . Case studies were conducted: (1) with the same number of sinusoids but variable amplitude, (2) with the same amplitude but variable number of sinusoids, and (3) same PCM volume, i.e., the same irregular cylindrical capsule perimeter. Unlike observations with a convection-only inside capsule, the amplitude of irregularity in capsule shape a , rather than capsule area, was found to be the dominant factor in heat transfer enhancement. The capsule area was important only during the initial conduction-dominant period. The opposing influences of amplitude a and number of sinusoidal undulations n on the intensity of natural convection was observed. The result shows that only increasing the area does not increase the melting performance, it is the amplitude a which enhance the melting performance.
