This work describes the development of a numerical module with a multiphysics structure to simulatethe thermo-hydro-chemo-mechanical behavior of compacted bentonites. First, the conceptual model,based on the state-of-the-art formulation for clay-based engineered barriers in deep geological repositories,is described. Second, the advantages of multiphysics-based modules are highlighted. Then, theguidelines to develop a code using such tools are outlined, presenting an example of implementation. Finally, the simulation of three tests that illustrate the behavior of compacted bentonites assesses thescope of the developed code. The satisfactory results obtained, and the relative simplicity of implementation,show the opportunity of the modeling strategy proposed

Intrathecal drug delivery is an effective and safe option for the treatment of chronic pathology refractory to conventional pain therapies. Typical intrathecal administered drugs are opioids, baclofen, local anesthetics and adjuvant medications. Although knowledge about mechanisms of action of intrathecal drugs are every day more clear many doubt remain respect the correct location of intrathecal catheter in order to achieve the best therapeutic result. We analyze the factors that can affect drug distribution within the cerebrospinal fluid. Three categories of variables were identified: drug features, cerebrospinal fluid (CSF) dynamics and patients features. First category includes physicochemical properties and pharmacological features of intrathecal administered drugs with special attention to drug lipophilicity. In the second category, the variables in CSF flow, are considered that can modify the drug distribution within the CSF with special attention to the new theories of liquoral circulation. Last category try to explain inter-individual difference in baclofen response with difference that are specific for each patients such as the anatomical area to treat, patient posture or reaction to inflammatory stimulus. We conclude that a comprehensive evaluation of the patients, including imaging techniques to study the anatomy and physiology of intrathecal environment and CSF dynamics, could become essential in the future to the purpose of optimize the clinical outcome of intrathecal therapy. Background: Dysregulation of DNA methylation has been related with obesity. Although the association of the FTO-rs9939609-T>A polymorphism with obesity has been widely reported, few studies have analyzed the methylation patterns associated with this polymorphism. Moreover, replication of the individual fi nding is needed to increase the evidence level. We aimed to analyze the association between methylation levels in selected CpG island of the FTO gene and the rs9939609 polymorphism in two populations. Methods: We studied 195(98 men and 97 women) subjects participating in the PREDIMED-Valencia study, and 46 subjects (23 men and 23 women) of the general population participating in the OBENUTIC study.DNA was obtained from leucocytes. The rs9939609 polymorphism was determined and quantitative DNA methylation analysis of the FTO gene was undertaken by matrix-assisted laser desorption ionization-time of fi ight (MALDI-TOF) mass spectrometry. We evaluated methylation levels in 21 CpG sites (13 in amplicon A and 8 in amplicon B) surrounding the rs9939609 polymorphim in intron 1 the FTO(Chr16:53817859-53822719). Results: Participants in the PREDIMED were older than participants in the OBENUTIC study. We detected significant differences in the methylation of several CpG sites between these populations, with PREDIMED participants having higher methylation levels. Interestingly, we observed that the CpG site 8 in amplicon A was differentially methylated depending on the FTO-rs9939609 polymorphism, where homozygous for the obesity-risk allele (AA) had lower methylation levels in both populations (expressed as % of methylated C:PREDIMED:44.5±39.2% in TT,19.4±26.8% in TA and 4.9±11.7% in AA;P<0.001;OBENUTIC:12.9±17.3% in TT,4.2±8.3% in TA and 0.0±0.0% in AA;P=0.046). Conclusions: We detected differences in methylation of CpG sites of FTO gene in high cardiovascular risk subjects. We also replicated the association between the rs9939609 polymorphism and methylation levels in both populations.

<P>We present observational evidence for oscillations of magnetic flux density in the quiet areas of the Sun. The majority of magnetic fields on the solar surface have strengths of the order of or lower than the equipartition field (300-500 G). This results in a myriad of magnetic fields whose evolution is largely determined by the turbulent plasma motions. When granules evolve they squash the magnetic field lines together or pull them apart. Here, we report on the periodic deformation of the shapes of features in circular polarization observed at high resolution with SUNRISE. In particular, we note that the area of patches with a constant magnetic flux oscillates with time, which implies that the apparent magnetic field intensity oscillates in antiphase. The periods associated with this oscillatory pattern are compatible with the granular lifetime and change abruptly, which suggests that these oscillations might not correspond to characteristic oscillatory modes of magnetic structures, but to the forcing by granular motions. In one particular case, we find three patches around the same granule oscillating in phase, which means that the spatial coherence of these oscillations can reach 1600 km. Interestingly, the same kind of oscillatory phenomenon is also found in the upper photosphere.</P>
