        CAUGHT IN THE ACT: STRONG, ACTIVE RAM PRESSURE STRIPPING IN VIRGO CLUSTER SPIRAL NGC 4330

        <P>We present a multi-wavelength study of NGC 4330, a highly inclined spiral galaxy in the Virgo Cluster which is a clear example of strong, ongoing intracluster medium-interstellar medium (ICM-ISM) ram pressure stripping. The H <SPAN CLASS='sml'>I</SPAN> has been removed from well within the undisturbed old stellar disk, to 50%-65% of R<SUB>25</SUB>. Multi-wavelength data (WIYN BVR-Hα, Very Large Array 21 cm H <SPAN CLASS='sml'>I</SPAN> and radio continuum, and Galaxy Evolution Explorer NUV and FUV) reveal several one-sided extraplanar features likely caused by ram pressure at an intermediate disk-wind angle. At the leading edge of the interaction, the Hα and dust extinction curve sharply out of the disk in a remarkable and distinctive 'upturn' feature that may be generally useful as a diagnostic indicator of active ram pressure. On the trailing side, the ISM is stretched out in a long tail which contains 10% of the galaxy's total H <SPAN CLASS='sml'>I</SPAN> emission, 6%-9% of its NUV-FUV emission, but only 2% of the Hα. The centroid of the H <SPAN CLASS='sml'>I</SPAN> tail is downwind of the UV/Hα tail, suggesting that the ICM wind has shifted most of the ISM downwind over the course of the past 10-300 Myr. Along the major axis, the disk is highly asymmetric in the UV, but more symmetric in Hα and H <SPAN CLASS='sml'>I</SPAN>, also implying recent changes in the distributions of gas and star formation. The UV-optical colors indicate very different star formation histories for the leading and trailing sides of the galaxy. On the leading side, a strong gradient in the UV-optical colors of the gas-stripped disk suggests that it has taken 200-400 Myr to strip the gas from a radius of >8 to 5 kpc, but on the trailing side there is no age gradient. All our data suggest a scenario in which NGC 4330 is falling into the cluster center for the first time and has experienced a significant increase in ram pressure over the last 200-400 Myr. Many of the UV-bright stars that form outside the thin disk due to ram pressure will ultimately produce stellar thick disk and halo components with characteristic morphologies and age distributions distinct from those produced by gravitational interactions.</P>

        Synthesis and biological evaluation of C-ring truncated deguelin derivatives as heat shock protein 90 (HSP90) inhibitors

        Based on the lead compound L-80 (compound 2), a potent heat shock protein 90 (HSP90) inhibitor, a series of C-ring truncated deguelin analogs were designed, synthesized and evaluated for Hypoxia Inducible Factor-1α (HIF-1α) inhibition as a primary screening method. Their structure-activity relationship was investigated in a systematic manner by varying the A/B ring, linker and D/E ring, respectively. Among the synthesized inhibitors, compound 5 exhibited potent HIF-1α inhibition in a dose-dependent manner and significant antitumor activity in human non-small cell lung carcinoma (H1299), with better activities than L-80. It also inhibited in vitro hypoxia-mediated angiogenic processes in human retinal microvascular endothelial cells (HRMEC). The docking study of 5 showed a similar binding mode as L-80: it occupied the C-terminal ATP-binding pocket of HSP90, indicating that the anticancer and antiangiogenic activities of 5 were derived from HIF-1α destabilization by inhibiting the C-terminal ATP-binding site of hHSP90.

        As the demand for saving energies has been getting higher all over the world the development for high-voltage and high capacity inverters has been achieved to reduce energy by controling the consumption patterns of industrial electric equipments. H-Bridge Multi-Level high-voltage inverter being researched is the single phase H-Bridge inverter by using low-voltage power semiconductor devices. It can make high-voltages close to a sine wave by connecting composed cells on series. This is a topology without filters because of the low input and output harmonic, and usages of it have been expanded in metal, oil, natural gas and electricity generation industries. The optimal operation conditions on these kinds of high-voltage inverters are varied by running environment conditions, and the performance tests for thermal stability of inner electric semiconductors changes are partially impossible because of facilities. In this research, the junction temperature of power electronics devices, distribution of surface temperature on heat sink and inner temperature and velocity of cells and systems have been drawn as the results by using ANSYS ICEPAK specialized in CFD simulation of power electronics equipments to secure the thermal reliability of cells and systems for 1500kW high-voltage inverters. Through these outcomes the ventilation structures and an effective arrangement of heat components of cell-based inverters could be applied to the plan for a uniform flow distribution of a high-voltage inverter system. Moreover, the consistency of thermal-flow simulation was proven by comparison of actual temperature through the cell-based performance tests.

        Vancomycin 약동학적 자문결과 분석

        The results of therapeutic drug monitoring (TDM) service were evaluated. Total 78 cases of peak and trough concentration of vancomycin were obtained on steady state. The 54 cases of all were obtained prior to TDM service. Statistically significant differences were observed between population vancomycin clearance by CAPCIL® computer program and Bayesian clearance by Bayesian method. Only 48.1% of the 54 cases given empirical dose of vancomycin were within therapeutic range. After TDM service, 23% (18/78 cases) were recommended dose reduction and 66.7% (12/18 cases) were accepted. Drug concentration were obtained from 15 cases who received modified dose through TDM service and 13 cases of them were within expected range. The total drug saving were 257 vials in 9 patients who were administered reduction dose (9/54 cases) and the calculated mean cost saving per case was 321,907 won (range : 78,911~473,466). In conclusion, TDM service for vancomycin is necessary to obtain optimal therapeutic drug level and for the cost saving. Furthermore the vancomycin dosing nomogram in Korean population should be developed.

      • NOTCH1 intracellular domain negatively regulates PAK1 signaling pathway through direct interaction

        p21-Activated kinase 1 (PAK1) is a serine/threonine protein kinase implicated in cytoskeletal remodeling and cell motility. Recent studies have shown that it also promotes cell proliferation, regulates apoptosis, and increases cell transformation and invasion. In this study, we showed that NOTCH1 intracellular domain (NOTCH1-IC) negatively regulated PAK1 signaling pathway. We found a novel interaction between NOTCH1-IC and PAK1. Overexpression of NOTCH1-IC decreased PAK1-induced integrin-linked kinase 1 (ILK1) phosphorylation, whereas inhibition of NOTCH1 signaling increased PAK1-induced ILK1 phosphorylation. Notably, ILK1 phosphorylation was higher in PS1,2<SUP>-/-</SUP> cells than in PS1,2<SUP>+/+</SUP> cells. As expected, overexpression of NOTCH1-IC decreased ILK1-induced phosphorylation of glycogen synthase kinase 3 beta (GSK-3beta). Furthermore, NOTCH1-IC disrupted the interaction of PAK1 with ILK1 and altered PAK1 localization by directly interacting with it. This inhibitory effect of NOTCH1-IC on the PAK1 signaling pathway was mediated by the binding of NOTCH1-IC to PAK1 and by the alteration of PAK1 localization. Together, these results suggest that NOTCH1-IC is a new regulator of the PAK1 signaling pathway that directly interacts with PAK1 and regulates its shuttling between the nucleus and the cytoplasm.

      • LRRK2 functions as a scaffolding kinase of ASK1-mediated neuronal cell death

        Leucine-rich repeat kinase 2 (LRRK2), a multi-domain protein, is a key causative factor in Parkinson's disease (PD). Identification of novel substrates and the molecular mechanisms underlying the effects of LRRK2 are essential for understanding the pathogenesis of PD. In this study, we showed that LRRK2 played an important role in neuronal cell death by directly phosphorylating and activating apoptosis signal-regulating kinase 1 (ASK1). LRRK2 phosphorylated ASK1 at Thr832 that is adjacent to Thr845, which serves as an autophosphorylation site. Moreover, results of binding and kinase assays showed that LRRK2 acted as a scaffolding protein by interacting with each components of the ASK1-MKK3/6-p38 MAPK pathway through its specific domains and increasing the proximity to downstream targets. Furthermore, LRRK2-induced apoptosis was suppressed by ASK1 inhibition in neuronal stem cells derived from patients with PD. These results clearly indicate that LRRK2 acts as an upstream kinase in the ASK1 pathway and plays an important role in the pathogenesis of PD.

      • JIP1 binding to RBP-Jk mediates cross-talk between the Notch1 and JIP1-JNK signaling pathway

        Notch1 signaling has a critical function in maintaining a balance among cell proliferation, differentiation, and apoptosis. Our earlier work showed that the Notch1 intracellular domain interferes with the scaffolding function of c-Jun N-terminal kinase (JNK)-interacting protein-1 (JIP1), yet the effect of JIP1 for Notch1–recombining binding protein suppressor of hairless (RBP-Jk) signaling remains unknown. Here, we show that JIP1 suppresses Notch1 activity. JIP1 was found to physically associate with either intracellular domain of Notch1 or RBP-Jk and interfere with the interaction between them. Furthermore, we ascertained that JIP1 caused the cytoplasmic retention of RBP-Jk through an interaction between the C-terminal region of JIP1 including Src homology 3 domain and the proline-rich domain of RBP-Jk. We also found that RBP-Jk inhibits JIP1-mediated activation of the JNK1 signaling cascade and cell death. Our results suggest that direct protein–protein interactions coordinate cross-talk between the Notch1–RBP-Jk and JIP1-JNK pathways.

      • Prediabetes is not a risk factor for subclinical coronary atherosclerosis

        Background: There are limited data regarding the influence of glycemic status on the risk of subclinical coronary atherosclerosis on coronary computed tomographic angiography (CCTA) in asymptomatic individuals. Methods: We analyzed 6434 asymptomatic individuals who underwent CCTA. The degree and extent of subclinical coronary atherosclerosis were assessed by CCTA, and ≥50% diameter stenosis was defined as significant. Of study participants, 2197 (34.1%), 3122 (48.5%), and 1115 (17.3%) were categorized as normal, prediabetic and diabetic individuals, respectively. Results: Compared with normal individuals, there were no statistically differences in the adjusted odds ratios of prediabetic individuals for significant coronary artery stenosis (0.98, 95% confidence interval [CI] 0.80-1.22, p=0.888), any plaque (0.96, 95% CI 0.86-1.07, p=0.483), calcified plaque (0.90, 95% CI 0.79-1.01, p=0.080), non-calcified plaque (1.02, 95% CI 0.88-1.17, p=0.803), and mixed plaque (1.00, 95% CI 0.82-1.22, p=0.983). However, adjusted odds ratios for significant coronary artery stenosis (1.71, 95% CI 1.34-2.19, p<0.001), any plaque (1.45, 95% CI 1.26-1.68, p<0.001), calcified plaque (1.35, 95% CI 1.15-1.57, p<0.001), non-calcified plaque (1.33, 95% CI 1.11-1.59, p=0.002), and mixed plaque (1.64, 95% CI 1.30-2.07, p<0.001) of diabetic individuals were significantly higher than those of the normal individuals. Conclusion: In asymptomatic individuals, diabetic individuals had a higher risk for subclinical coronary atherosclerosis, but prediabetic individuals were not associated with an increased risk of subclinical coronary atherosclerosis.

        We introduce a semidiscrete mixed finite element approximation for the single-phase linear Stefan problem and show the unique existence of the approximation. And the optimal rate of convergence in $L^2$ and $H^1$ norms are derived.
