        In Silico identification of dengue NS5 phytochemical inhibitors as potential antiviral drug compounds via molecular docking

        Dengue fever is a viral disease caused by the dengue flavivirus, transmitted through the bites of infected mosquitoes. The endeavor to combat dengue has led many researchers to develop antiviral drugs. Several medicinal plants, containing diverse phytochemicals, exhibit potential for inhibiting pathogenic proteins and are employed in ongoing research and therapeutic advancement. This has piqued researchers’ interest in identifying potential plantderived molecule inhibitors 』of the dengue virus Non- Structural Protein 5 (NS5) and analyzing their subsequent interactions. This study employed candidate selection and molecular docking using an in silico approach. We used the Protein Data Bank database and performed National Center for Biotechnology Information (NCBI) blastp analysis to gather and compare the best protein structures of the two NS5 serotypes. In addition, we identified ligand molecules previously reported to exhibit potential inhibitory effects against NS5 by retrieving their 3D structures from NCBI PubChem. Moreover, we utilized PyRx and PyMOL to perform molecular docking. The findings revealed a conspicuous prevalence of interactions between the MTase domain in NS5 and phytochemical compounds, notably Kaempferol and 6-Shogaol. Based on the evidence presented in this study, we propose that investigating the NS5 protein in flavivirus is necessary for dengue fever prevention, as these proteins play a vital role in the replication of the virus. Our findings provide valuable insights beneficial for advancing research into antiviral medication. We suggest further investigation into other medicinal plants that may inhibit dengue and the additional scrutiny of Kaempferol and 6-Shogaol, the compounds that yielded significant results in this study.

        High Histologic Grade and High Ki-67 Expression Predict Phenotypic Alterations in Node Metastasis in Primary Breast Cancers

        Purpose: Several studies have shown that estrogen receptor (ER) and progesterone receptor (PR) expression and human epidermal growth factor receptor 2 (HER2) expression may vary during tumoral progression. We aimed to describe and compare ER, PR, and HER2 expressions in primary breast tumors and synchronic axillary nodal metastases, and evaluate phenotypic correlations between them. Methods: Patients were identified prospectively through surgical procedures between September 2013 and July 2016. The status of ER, PR, HER2, and Ki-67 were pathologically analyzed in breast cancers and axillary nodal metastases; these patients were classified based on the breast cancer phenotypes into five subgroups. Results: Synchronic axillary nodal metastases were observed in 127 patients. In breast cancers and nodal metastases, correlation analyses of ER, PR, and Ki-67 expression showed a statistical dependence and concordance between these samples was unambiguously demonstrated through Bland-Altman plots for each determination. Primary breast tumors were classified as follows: luminal A, 41.6%; luminal B, 40.0%; luminal B/HER2, 9.6%; HER2, 2.4%; triple negative, 6.4%. Alterations in phenotype were observed in 28% of patients. The most frequent phenotypic alteration was from luminal B to A (36.4%). Ten cases (30.3%) showed alterations with therapeutic implications; six gained HER2 overexpression, and four, hormonal receptor (HR) expression. A moderate strength of agreement (Cohen’s κ coefficient, 0.59; 95% confidence interval, 0.48−0.71) was observed. In multivariate analyses, high histologic grade (odds ratio [OR], 2.79; p<0.047) and high Ki-67 expression (OR, 1.05; p<0.037) were independent factors predictive of phenotypic alterations. Conclusion: Strong correlations were observed in HR and Ki-67 expressions between primary breast tumors and axillary nodal metastases, and a moderate concordance was observed in their phenotypical characteristics. Nevertheless, alterations did exist, and one-third of these changes may have therapeutic implications. The nodal metastases of tumors with high grade and high Ki-67 expression may need to be analyzed, to obtain complete therapeutic information.

        Total Maxillary Reconstruction Using a Double-Barreled and Double Skin Paddle Fibular Flap after Total Maxillectomy

        Chondroblastomas are rare entities accounting for approximately 1% of all primary bone tumors. We describe a case of a 7-year-old girl with a giant chondroblastoma of the maxilla, treated with bilateral class III maxillectomy and reconstruction with a double-barreled and double skin paddle fibular free flap. We show evidence of an excellent aesthetic outcome at 6 months’ follow up with no evidence of tumor recurrence.

        Growth performance and health of nursing lambs supplemented with inulin and Lactobacillus casei

        Objective: This experiment was designed to evaluate the effects of Agave tequilana inulin and Lactobacillus casei (L. casei) on growth performace, hematological variables, serum metabolites, and total coliforms in nursing lambs. Methods: The experimental design was completely randomized; treatments were T1, control (pre-starter concentrate, PC), T2: T1+2% inulin, and T3: T1+2% inulin+L. casei; treatments were compared with Tukey test (p≤0.05); and 45 new born Kathadin×Dorset lambs (4.8±0.8 kg birth weight) were the experimental units (15 per treatment). The variables were daily weight gain (DWG), dry matter intake and diarrheas incidence (%) during 56 d. Twenty-four hours after birth and at the end of the experiment, blood samples were collected to evaluate hematological variables and serum metabolites. Besides, the populations of total coliforms and lactobacilli were estimated in fecal samples. Results: Addition of agave inulin and L. casei increased (p≤0.05) DWG 356, 384, and 415 g/d, weaning weight 24.92, 26.18, and 28.07 kg, as well as lactobacilli population 5.79, 6.32, and 6.48 Log10 cfu/g, for T1, T2, and T3, respectively. Lambs fed L. casei had decreased (p≤0.05) populations of total coliforms (T1 = 6.18, T2 = 5.77, and T3 = 5.07 Log10 cfu/g), diarrheas incidence (T1 = 11.67%, T2 = 8.33%, and T3 = 5.0%), and serum cholesterol concentration (11% in T2 and 13% in T3, compared to control). Conclusion: The combination of Agave tequilana inulin and L. casei increases weight gain and improves intestinal health by reducing coliforms and diarrheas incidence in Katahdin× Dorset lambs during the pre-weaning period.
