This study was conducted for SNPs in the 5'-regions of estrogen receptor-${\alpha}$ (ESR-${\alpha}$), and association with calving interval (CI), service per conception (SPC) and 305 days milk yield in Hanwoo and Holstein dairy cattle. The genetic improvement was incurred low reproduction performance. The objective of this study was to investigate connections between single nucleotide polymorphisms (SNP) of Estrogen receptor-${\alpha}$ (ESR-${\alpha}$) with reproduction performance (calving interval, service per conception, and 305 d milk yield) in Hanwoo and Holstein dairy cattle. Hanwoo and Holstein blood samples were collected from 183 and 124 dam of breeding farms and DNA was extracted. Primer design was based on NCBI GenBank (Accession No. AY340579). The PCR-RFLP method with Bgl I was used to genotype the cattle. The result showed two variants of the ESR-${\alpha}$ gene. The Bgl I cut the 492 bp amplification product into 322 bp and 170 bp fragments for allele G, while allele A remained uncut, resulting in two restriction fragments for homozygote G/G and three fragments for heterozygote A/G. We found two of different genotypes in these breeds, A/G and G/G. In Hanwoo, the A/G genotype frequency was 0.13, and G/G was 0.87. The CI of A/G was $382.18{\pm}10.03$ days, and G/G was $381.69{\pm}5.22$ days. The SPC of A/G was $1.62{\pm}0.16$, and G/G was $1.32{\pm}0.04$. While CI showed no significance difference, SPC exhibited significant difference (p<0.05). In Holstein cattle, the frequency of genotype A/G was 0.02 and G/G was 0.98. The 305 days milk yield of A/G was $7,253.00{\pm}936.00kg$ and of G/G was $8,747.51{\pm}204.88kg$, showing no significant difference.

As a nursing practice involves nurses' actions in a specific context of health care, this study has focused on exploring the espoused theories in nursing practice within the action science perspectives, Espoused theories are the belief, principles, and rationale expressed by the practitioner as guiding her/his actions in a situation of practice. The data were analysed qualitatively and 25 elements of espoused theories of nursing action were identified and clustered into 6 categories. The 25 elements of espoused theories are as follows: The clinical nurse worked in wholistic and individual nursing, focussed on the patient's needed, comfort and supportive nursing (5 theories of nursing goal) ; excellent skills, knowledge based, assessment and data collection, explaining, educating or a scientific basis(6 theories of nursing intervention): advocacy, value oriented, treatment, account- ability and commitment(4 theories of nursing ethics) ; human respect. partnership, trust(3 theories of patient-nurse relationship) : knowledgable, accumulated clinical experiences and personally lived experiences. positive perspectives(4 theories of nurse), role of intervention. rewarding peer relationship(3 theories of situations) The above mentioned espoused theories are similar to that of nursing textbooks which students learned through basic nursing education and almost the same as the Acts ofa Nurse in Korean. However, we are doubtful whether nurses actually do as they think. Therefore. it is recommended to review the theories-in-use in order to and any discrepancies between the espoused theories and the reality of nursing actions
Litsenolide A2: The major anti-inflammatory activity compound in <i>Litsea japonica</i> fruit

<P><B>Abstract</B></P> <P>In this study, we investigated the anti-inflammatory effect of litsenolide A2 (LNA2) isolated from <I>L. japonica</I> fruit and its mechanism of action in LPS-induced RAW 264.7 cells. LPS-induced production of anti-inflammatory mediators and cytokines such as NO, PGE<SUB>2</SUB>, iNOS, COX-2, TNF-α, and IL-6 was significantly inhibited by LNA2. Next, western blot experiments were performed to investigate the mechanism of action of the anti-inflammatory effect of LNA2. The results indicated that LNA2 markedly reduced the LPS-induced activation of NF-κB and the nuclear translocation of NF-κB p65. Furthermore, LNA2 also inhibited the phosphorylation of MAPK by LPS. LNA2 was the major component in the ethanol extract of <I>L. japonica</I> at 59 mg/g according to high performance liquid chromatography (HPLC). These results indicate that LNA2 can be used as a functional and marker compound for standardization of the manufacturing process when <I>L. japonica</I> fruit is used as a functional food.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We study the anti-inflammatory effect of LNA2 in LPS-activated RAW 264.7 cells. </LI> <LI> LNA2 inhibited production of pro-inflammatory mediator and cytokines. </LI> <LI> LNA2 exhibited anti-inflammatory properties by suppression of NF-κB and MPAKs. </LI> <LI> LNA2 was the major component in the ethanol extract of <I>L. japonica</I> at 59 mg/g. </LI> </UL> </P>

The ubiquitin conjugating enzyme E2 (UBC E2) mediates selective ubiquitination, acting with E1 and E3 enzymes to designate specific proteins for subsequent degradation. In the present study, we characterized the function of the mung bean VrUBC1 gene (Vigna radiata UBC 1). RNA gel-blot analysis showed that VrUBC1 mRNA expression was induced by either dehydration, high salinity or by the exogenous abscisic acid (ABA), but not by low temperature or wounding. Biochemical studies of VrUBC1 recombinant protein and complementation of yeast ubc4/5 by VrUBC1 revealed that VrUBC1 encodes a functional UBC E2. To understand the function of this gene in development and plant responses to osmotic stresses, we overexpressed VrUBC1 in Arabidopsis (Arabidopsis thaliana). The VrUBC1-overexpressing plants displayed highly sensitive responses to ABA and osmotic stress during germination, enhanced ABA- or salt-induced stomatal closing, and increased drought stress tolerance. The expression levels of a number of key ABA signaling genes were increased in VrUBC1-overexpressing plants compared to the wild-type plants. Yeast two-hybrid and bimolecular fluorescence complementation demonstrated that VrUBC1 interacts with AtVBP1 (A. thaliana VrUBC1 Binding Partner 1), a C3HC4-type RING E3 ligase. Overall, these results demonstrate that VrUBC1 plays a positive role in osmotic stress tolerance through transcriptional regulation of ABA-related genes and possibly through interaction with a novel RING E3 ligase.
The Difference of the Changes of Images on Ultrasound Scanner Setting Parameters

The setting parameters of ultrasound scanner give influences to change of image. Sonographers have used a Matlab program to make Low Contrast Sensitivity(LCS) value and compared original images in order to evaluate the use of the supersonic diagnosis machinery. We confirmed the change of image in Grayscale values using Photoshop program. Experiment equipment of our research used A Medison Accuvix V10, A Multi-Tissue Ultrasound Phantom(040 GSE) of CHRIS Company, A Adobe Photoshop CS4 Program, A Convex Probe, A USB memory stick, A Probe Fixation Equipment. The method used Gain, Dynamic Range(DR) of the setting parameters of ultrasound scanner and researched Gain and DR was set to 10 dB. We changed the different settings to see the changes of images using Grayscale values of a Photoshop program about tissue images of a phantom. This study evaluated DR and Gain whether it is an image controller to get the optimum contrast to produce an image to see the how effect on the images. We did not use Gateway in supersonic diagnosis machinery. We can easily open to open the files through Photoshop program before we get Digital Imaging and Communications in Medicine(DICOM) files use USB memory stick in supersonic diagnosis machinery. When we diagnosed the lesion of the patient with ultrasound, the contrast and the Gray scale value of image are very important. In this research, we determined the optimum setting parameters that provided useful information to diagnose disease and evaluated the change of improved images.

<P><B>Abstract</B></P> <P>We synthesized a planar pDPPTTi-OD polymer based on diketopyrrolopyrrole (DPP) and thieno [2,3-<I>b</I>]thiophene (TTi) and investigated the electrical properties of the pDPPTTi-OD polymer. pDPPTTi-OD films displayed a low optical bandgap of 1.57 eV, and HOMO and LUMO levels of −5.40 and −3.74 eV, respectively. The 150°C-annealed pDPPTTi-OD films showed a high hole mobility of 0.16 cm<SUP>2</SUP>V<SUP>−1</SUP>s<SUP>−1</SUP> in organic thin-film transistor (OTFT) devices. The photovoltaic properties of polymer solar cells (PSCs) incorporating the pDPPTTi-OD were also measured. A pDPPTTi-OD:PC<SUB>71</SUB>BM blend film was spin-coated at 25, 70 and 90 °C. High-temperature processing significantly improved the power conversion efficiency of PSCs by effectively reducing the PC<SUB>71</SUB>BM domain sizes, which improved the miscibility between pDPPTTi-OD and PC<SUB>71</SUB>BM. The $Na^+/H^+$ exchanger is responsible for maintaining the acidic tumor microenvironment through its promotion of the reabsorption of extracellular $Na^+$ and the extrusion of intracellular $H^+$. The resultant increase in the extracellular acidity contributes to the chemoresistance of malignant tumors. In this study, the chemosensitizing effects of cariporide, a potent $Na^+/H^+-exchange$ inhibitor, were evaluated in human malignant mesothelioma H-2452 cells preadapted with lactic acid. A higher basal level of phosphorylated (p)-AKT protein was found in the acid-tolerable H-2452AcT cells compared with their parental acid-sensitive H-2452 cells. When introduced in H-2452AcT cells with a concentration that shows only a slight toxicity in H-2452 cells, cariporide exhibited growth-suppressive and apoptosis-promoting activities, as demonstrated by an increase in the cells with pyknotic and fragmented nuclei, annexin V-PE(+) staining, a $sub-G_0/G_1$ peak, and a $G_2/M$ phase-transition delay in the cell cycle. Preceding these changes, a cariporide-induced p-AKT down-regulation, a p53 up-regulation, an ROS accumulation, and the depolarization of the mitochondrial-membrane potential were observed. A pretreatment with the phosphatidylinositol-3-kinase (PI3K) inhibitor LY294002 markedly augmented the DNA damage caused by the cariporide, as indicated by a much greater extent of comet tails and a tail moment with increased levels of the p-histone H2A.X, $p-ATM^{Ser1981}$, $p-ATR^{Ser428}$, $p-CHK1^{Ser345}$, and $p-CHK2^{Thr68}$, as well as a series of pro-apoptotic events. The data suggest that an inhibition of the PI3K/AKT signaling is necessary to enhance the cytotoxicity toward the acidtolerable H-2452AcT cells, and it underlines the significance of proton-pump targeting as a potential therapeutic strategy to overcome the acidic-microenvironment-associated chemotherapeutic resistance.
