Bone morphogenetic protein-2(BMP-2) was widely used in orthopedics and dental implant surgery but there are a variety of side effects including unwished bone formation, cancerous signaling process. To solve these problems, novel synthetic peptide (osteogenic peptide-5, OP-5) derived from BMP-2 was developed for clinical use. The OP-5 influence was evaluated by in vitro and in vivo assay compared with BMP-2. To optimize the osteogenic effects, it was conjugated onto electrospun fibers fabricated from poly(L-lactide). The peptide tightly attached to nanofibers and it was maintained at least 28 days, in vivo mouse calvarial defect model showed that OP-5 conjugated nanofibers could increase the bone regeneration and mechanical property similar to BMP-2 coated PLLA. Our results suggested that OP-5 can be used as new therapeutic agents for bone regeneration and OP-5 conjugated nanofibers might be a feasible method as guided bone regeneration for a dental and orthopeadic treatment.
RE-ORGA to Alleviate DHT-induced Male Baldness in Human Dermal Papilla Cells

Testosterone and dihydrotestosterone(DHT) that are affect hair loss. 5α -reductases are converting testosterone into DHT. In hair loss processes, TGF-β1, DKK-1, DHT/DHT-AR(androgen receptor) complex are key factors. RE-ORGA was extracted from natural ingredient to alleviate hair loss with less adverse effects. This study showed that RE-ORGA can induce proliferation of dermal papilla and regulate molecular signaling pathway. Western blot analysis treated with RE-ORGA showed lowered expression of 5α-reductases. From RNA-Seq their expression was also examined to be decreased in transcriptional levels. Log-scaled fold changes of SRD5A1,3 and Bax were -0.518, -0.253, -0.758. Androgen receptor and TFGB1I1, co-activator for androgen receptor, were also down-regulated. On the other hand Ptk2b that is negative androgen receptor regulator showed over-expression. These results showed that RE-ORGA could regulates hair loss induced by testosterone and DHT.

The most widely used chymopapain for chemonucleolysis was withdrawn from the market due to its anaphylactic reactions. To overcome these complications, the development of alternative without side-effects is a recent challenge in the field of bioengineering to treat back pain accompanying disc degeneration. In this study, Thermomatrix (Winnova Co. Ltd, Korea) was developed by hydrophobically modifying glycol chitosan, which has thermosensitive sol-gel transition, to reduce hernia symptoms. Lumbar disc segments were retrieved from sacrificed miniature porcine. After disc degeneration with the injection of chondroitinase ABC to porcine discs, hydrogel mixture accompanied with barium sulfate and ethyl alcohol was injected into the pre-defected site. After 4 week cultivation, discs were characterized with radiographic images and histological analysis

The 1st generation of scaffolds for spine fusion demands only `supporting` between vertebral bodies. Recently, it is needed that implants for spinal surgery have biocompatibility and functional properties such as bone regeneration. Besides, many studies reported non-union among the causes of the failure in bone fusion between implant and vertebra. In this study, new scaffold was introduced to commercial spinal interbody disk cage (PEEK). New bone integrity induced by PEEK implant incorporated with sponge-typed scaffold loaded with BMP-2. The implant prototype production is composed of disk case incorporated with sponge typed scaffold loaded with BMP-2 which is scheduled to preclinical trials using mini-porcine.

Disc herniation is one of the most common intervertebrate disc (IVD) diseases. The use of injectable hydrogel formulations have been suggested as a promising strategy for the treatment of degenerative disc disease that can both restore the biomechanical function and reduce the low back pain. Here, a new thermo-sensitive injectable N-hexanoyl glycol chitosan (HGC) hydrogel was fabricated with tunable thermosensitivity and high stability. Various HGCs that show thermo-sensitive sol-gel transition properties were synthesized and their physicochemical properties were characterized by <sup>1</sup>H NMR, FT-IR and rheological measurements. HGC hydrogel was injected into the degenerated IVD ex vivo model and evaluated by macroscopic observation, micro-computed tomography examination and histological analysis. The defected IVD was fully filled with HGC hydrogel, which was stable even at 28 days. These results suggested that HGC hydrogel has great potential for percutaneous treatment of disc herniation.

A series of thermo-sensitive N-acyl glycol chitosans were synthesized by a simple one-step N-acylation of glycol chitosan with acetic anhydrides and hexanoic anhydrides. N-acyl glycol chitosans with different molecular weights of hydrophobic groups and degree of acylation (DA) were synthesized to improve their hydrophobicity and stability. The chemical structures and viscoelastic properties of N-acylation of glycol chitosans were characterized by FT-IR, <sup>1</sup>H-NMR and rheometer. The influence of molecular weight of hydrophobic group and degree of acetylation was studied by tube inverting method on the thermo-sensitive sol-gel transition properties of N-acyl glycol chitosans. Biocompatible and thermo-sensitive N-acyl glycol chitosans hold great promise serving as new in-situ thermogelling platforms for various biomedical applications.

천연물로부터 유용성분을 추출하는 방법으로 열수추출법, 용매추출법 및 기계적 압착법 등 전통적인 방법이 주고 사용되고 있다. 어성초, 감초, 녹차 등 다양한 천연물을 포함하고 있는 BH-ORGA41의 유효성분을 추출하기 위하여 증기추출법을 이용하였다. 본 연구는 두피 탈모 개선과 예방을 위한 방법으로 BH-ORGA41의 성분의 효능을 알아보고자 한다. 먼저 세포독성을 확인하기위하여 MTT assay를 진행하였다. 또한 BH-ORGA41이 테스토스테론(testosterone)이 디하이드로테스토스테론(dihydrotestosterone, DHT)으로 변화하는 과정에 작용하는 효소인 5α-환원효소(5α-reductase)을 억제하는 효과를 가지고 있는지 확인하였다. 그 결과 천연복합물인 BH-ORGA41은 독성을 나타내지 않으며 탈모 개선에 효과가 있는 것으로 보여진다.

In recent years, the use of orthopedic implants for spine impairments has been rapidly increased and become prevalent for lumbar interbody fusion. The PEEK has a much lower stiffness than other metal materials. When using it for spinal fusion cage, the stress in shielding bone graft and the slow-down fusion rate were observed for the PEEK clinical applications. We have developed to evaluate the osteocompatibility of the PEEK device filled with rhBMP-2 on sponge composed of collagen and thermosensitive hydrogel that was manipulated to release slowly of rhBMP-2. Osteogenesis induced by scaffold was evaluated by assay of ALP activity, PCR and immunoblotting to determine the degree of osteogenesis in vitro. Then, the in vivo effective bone regeneration was achieved with the rhBMP-2 loaded on scaffold higher bone mass and mineral density than that of the control showing the images of u-CT and H&E on the mouse calvarial critical size defect model and porcin disc implant model.
