On-Chip Lipid Extraction Using Superabsorbent Polymers for Mass Spectrometry

<P>Pretreatment of samples is one of the most important steps in analytical methods for efficient and accurate results. Typically, an extraction method used for lipid analysis with mass spectrometry is accompanied by complex liquid–liquid extraction. We have devised a simple, rapid, and efficient lipid extraction method using superabsorbent polymers (SAPs) and developed a high-throughput lipid extraction platform based on a microfluidic system. Since SAPs can rapidly absorb an aqueous solution from a raw sample and convert it into the gel, the lipid extraction process can be remarkably simplified. The hydrophobic lipid components were captured into the fibrous SAP gel and then solubilized and eluted directly into the organic solvent without significant interference by this polymer. The small-scale lipid extraction process minimizes the liquid handling and unnecessary centrifugation steps, thereby enabling the implementation of a SAP-integrated microfluidic lipid extraction platform. The SAP method successfully induced reproducible extraction and high recovery rates (95–100%) compared to the conventional Folch method in several lipid classes. We also demonstrated the feasibility of the SAP method for the analysis of lipids in complex biological samples, such as the brain and liver, as well as <I>Escherichia coli</I>. 성폭력 상담사의 대리외상에 대한 주관성 인식 유형 연구

본 연구는 성폭력 상담사들의 대리외상에 대한 주관성을 유형화하고 유형별 특성을 도출하여 상담사의 상담 후 후유증 및 스트레스 관리에 대한 지지전략과 문제해결에 기초자료를 제시하는 데 목적이 있다. 연구방법은 Q방법론을 적용하여 성폭력 상담사에 대한 심층면접과 대리외상과 관련된 문헌고찰을 종합적으로 분석하여 33개의 Q표본을 선정하였고, 성폭력 상담사 총 25명을 P표본으로 표집하여 분류하도록 하였다. 분석 결과, 상담사의 대리외상에 대한 인식은 4가지로 유형화되었다. 첫째, 상담과정에 정서적객관성을 유지하는 데 어려움을 느끼는 내담자 정서 과몰입형, 둘째, 개인의 가치관이부정적으로 변화하여 대인관계의 축소를 초래하는 가치관 변화형, 셋째, 상담사례를 통해 개인 및 주변 사람들의 안전을 염려하는 안전 중시형, 마지막으로 상담 후유증이 상담사의 신체와 정서의 전반에 영향을 미치고 있는 심신 민감형으로 인식유형의 특성이나타났다. 이상의 연구결과를 통하여, 성폭력 상담사의 유형화된 대리외상의 특성을 바탕으로 대리외상을 완화하기 위한 서비스 지원과 제도방안에 대해 논의하였다. This study proposes that grouped to subjectivity regarding vicarious traumatization of sexual abuse counselors and suggest basic data to support strategy and problem solving to aftereffects and stress management after consultation of sexual abuse counselor as derive from a characteristic of each group. The study utilizes Q methodology, and selected 33 sample Q from analyzed in-depth interviews with sexual abuse counselors within the literature review. The study also divided P sample into sampling 25 sexual abuse counselors. The analyzed result indicates grouping by following four categories: type of emotional involvement for clients, type of change of identity, type of emphasis on safety, and type of sensitive to the physical and mental. Based on the results, the aforementioned study examined support to relieve vicarious traumatization and arrange a systematic plan according to each group of sexual abuse counselors.

본 연구는 사회복지 실무자의 권위주의적 성격, 사회적 소수자에 대한 편견, 문화적 역량 간의 관계에 대한 모형을 가정하고 검증하기 위한 연구로, 409명의 조사 자료를 분석하였다. 권위주의적 성격을 독립변수, 사회적 소수자에 대한 편견을 매개변수, 문화적 역량을 종속변수로 하여 모형을 구성하였으며, 가설분석을 위해 AMOS 18.0을 활용하였다. 분석 결과 첫째, 권위주의적 성격과 문화적 역량 사이에 사회적 소수자에 대한 편견의 매개효과에 대해 독립변수의 직접경로가 포함되지 않은 완전매개모형의 적합도가 높은 것으로 나타났다. 둘째, 권위주의적 성격은 문화적 역량에 직접 효과는 통계적 유의성이 없었으며, 사회적 소수자에 대한 편견을 매개로 한 간접효과가 검증되었다. 본 연구 결과는 향후 문화적 역량강화를 위한 교육에 실무자의 자기인식의 중요성을 강조하는 근거로 활용될 것으로 기대된다. This study intends to investigate the main and mediating effects which social prejudice against minorities on cultural competence of social service workers considering the relationship with authoritarian personality. The survey was conducted upon 409 social service workers. Collected data were analyzed by SEM (structural equation modeling) method using AMOS 18.0 The summary of the result is as follows. First, the model fit of full mediation model which is elimination direct path of authoritarian personality to cultural competence is better than partial mediation model. Second, only the indirect effect of authoritarian personality to cultural competence is statically significant but direct effect. Therefore, social prejudice against minorities was main factor to mediate between authoritarian personality and cultural competence. In conclusion, we could confirm the necessity of the program about social prejudice against minorities to social service workers especially who has authoritarian personality tendency.

Porphyromonas gingivalis, a gram-negative anaerobic oral bacterium, causes periodontal disease by binding to saliva-coated oral surfaces. The FimA protein from P. gingivalis is a crucial pathogenic component of the bacterium and a target for vaccine development against periodontal disease. Complementary DNAs encoding the heavy and light chains of two monoclonal antibodies that bind specifically to the FimA protein were cloned into a plant expression vector under the control of the duplicated Cauliflower Mosaic Virus 35S promoter, and agroinfiltration was used to allow the vectors to infiltrate tobacco plants. The expressions of the heavy and light chains in the leaf tissue were detected using antibodies specific to each antibody chain. Western blot analysis showed the specific binding of the plant-derived monoclonal antibodies to the native FimA protein purified from P. gingivalis. Our finding that plant-derived monoclonal antibodies bound specifically to the native FimA protein indicates that plantderived monoclonal antibodies can protect against P. gingivalis invasion.
