      • Catalytic Properties of Aldehyde Dehydrogenase

        The effect of several modifiers including disulfiram and phenylmethanesulfonylfluoride and metal ions on the dehydrogenase and esterase activities of several aldehyde dehydrogenase (ALDH) isozymes isolated from rat liver and brain, bovine brain and baker's yeast was investigated to see whether or not the both reactions share a cmomon active site. Several lines of experimental evidence suggested that ALDH might have a common active site for both reactions, but with different microenvironmental situations for dehydrogenase and esterase reactions. 쥐의 간과 뇌, 소의 뇌, 효모균에서 추출 정제한 몇 가지 aldehyde dehydrogenase(ALDH) isozyme에 미치는 disulfiram, phenylmethanesulfonyl-fluoride 등과 같은 modifier와 금속이온 등의 영향을 관찰하고 ALDH의 dehydrogenase 반응과 esterase 반응의 활성부위의 동일성 여부를 추구한 결과 ALDH의 dehydrogenase 반응과 esterase 반응의 활성부위는 같으나 주변의 미세환경에 차이가 있는 것으로 생각된다.

        계배발생에 미치는 5 - hydroxytryptamine ( serotonin ) 의 영향에 관한 세포생물학적 연구

        Effect of 5-hydroxytryptamine(5-HT) on the proliferaton of brain and somite parts of 4 day old chick embryo have been investigated. When the brain cells were cultured in Dulbeco`s modified essential medium containing 5-HT (10^(-5)-10^(-3)M), a significant necrosis occured and the number of survived cells were only a half that of control group. Somite cells were also the case as seen in brain cells. RNA and protein synthesis were also greatly impaired when the cells were cultured in the medium containing 5-HT (10^(-5)-10^(-3)M). Among the protein bands appeared on SDS-polyacrylamide gel electrophoretogram of total protein extract, band 2 was found to be PAS staining positive glycoprotein and its molecular weight was identified being 74,000 dalton by SDS-polyacrylamide gel electrophoresis. SDS-polyacrylamide gel electrophoretogram of NaCl/EDTA extract showed that this was brain cellular peripheral glycoprotein. This glycoprotein was not bound to concanavalin A-lectin suggesting that the sugar might be linked to protein moiety through O-glycosidic bond, but the exact structure must be elucidated. It was also found that the mount of this glycoprotein decreased when the brain cells were cultured in the medium containing 5-HT. 5-HT may interfere the biosynthesis of peripheral glycoproteins which may play significant roles in various cellular phenomena. From the previous work and present results, it seems that the intracellular 5-HT level must be very improtant to control cellular metabolism. Externally supplied 5-HT might interfere the intracellular level of 5-HT resulting in abnormal metabolism. Possibly, 5-HT may have a potential function in cell proliferation and in early phase of embryogenesis as well as in adult central nervous system.
