To elucidate the preventive effect of oriental medicine Gungchi-hwadamsun (GH) on stress, we investigated the physiological change of rats which were applied immobilization stress. For immobilization stress, rats were placed in restrainer for 12 hours a day for 3 days. During application of stress, body weight of rats was measured. After sacrifice, 8 organs were taken for measurement of organ weight. Brain was sectioned into 4 parts that are Frontal Cortex, Corpus Striatum, Hypothalamus and Hippocampus. Each part was homogenated and its catecholamine and serotonin contents were measured with HPLC. In our study, stress mainly induced increase of concentration of neurotransmitters in brain, but had mild effect on other physical function of rats. GH inhibited stress induced changes of neurotransmitter content in brain.

In oriental medicine, it is well known that Angelica gigantis tonifies and harmonizes the blood, regulates the menses, and moistens the intestines. This herb has been used for deficient blood pattern, menstural problem, abdominal pain, and traumatic injuries in oriental medicine. A major component of Angelica gigantic consist of a various coumarines, decursin, schooltime, umbelliferone, safrole, 3-butylphalide sistesterol. In this study, we aimed to measure the contents of decursin, one of major coumarine derivative, to determine the standardization of Angelica gigantis.
대식세포(大食細胞) oxLDL 생성(生成)에 미치는 독활기생탕(獨活寄生湯)의 영향(影響)

The oxidative modification of low density lipoprotein(LDL) has been implicated in the development of atherosclerosis . Oxidized LDL(oxLDL) are found in macrophage foam cell , and it can induce an macrophage proliferation in atherosclerotic plaque. In this study, we investigated the hypothesis that Tokhwalkisaengtang(TK) may reduce atherosclerosis by lowering the oxidizability of LDL, To achieve this goal, we examined the effect of TK on LDL oxidation, nitric oxide production in murine macrophage cell line , and the effect of TK on cupuric sulfate-induced cytotoxicity. LDH release, and macrophage activity TK inhibited the generation of oxLDL from native LDL in macrophage cell culture, and decreased the release of LDH from cupric sulfate-stimulated macrophage. In other experiments, TK activated macrophase cell, and increased the survival rate, and enhanced nitric oxide production in macrophage.

In oriental medicine, Artemisiae asiatica (AA) has been used as analgesic, antiinflammatory agent, and coagulatory agent. Furthermore, eupatilin, a kind of flavonoids, is known as the active principle component of AA. This study was undertaken to determine the gastric antiulceration of AA and to elucidate its mechanism. AA showed the inhibitory effect on gastric ulceration induced by EtOH/HCl and aspirin. To elucidate its mechanism, the effect of AA on lipidperoxide level in gastric mucosa, microsome lipidperoxidation, iron -dependent lipidperoxidation, and neutrophil activation were examined. It is suggested that the antiperoxidative and antineutrophil activity of AA play important roles as a possible mechanism. These results suggest that AA might have gastric antiulceration activity due to antilipidperoxdative and antineutrophil activity.

It is well known that Dioscoreae Rhizoma tonifies and benefits the Spleen and Stomach, benefits the Lung and nourishes the Kidney. A major component isolated from this herb consist of dioscin, diosgenin, allantoin, starch, alanin, mucilage, choline, amylase, and glycoprotein. In this study, we aimed to measure the content of allantoin, one of major component, to determine the quality of Dioscoreae Rhizoma.

It is well known that Poncirus trifoliata harmonizes the Stomach dissipates the Nodules of Liver and manages Vitality. This herb has been used for stomachache, vomitting, nausea, abdominal distension and swelling of testis in the Oriental Medicine. A major components of Poncirus trifoliata consist of a various flavonoids such as poncirin, naringin, hesperidin, neohesperidin, rhoifolin, poncimarin, poncitrin. Generally pharmacological activities of an oriental medicinal drug may be different due to diverse origin, harvest season and storage Period. In this study, we aimed to measure the contents of poncirin and naringin, major flavonoid of this herb, to determine the quality of Poncirus trifoliata.

Osteoporosis is characterized by bone loss and mobidity with osteoporotic fracture. This study was performed to evaluate the effect of on the bone mass and its related factors in estrogen-deficient animal model. The model rats of osteoporsis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 14th day after ovariectomy-surgery, rats were administered with DBE, extract of Dioscorea batatas, per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of DBE. Osteoporetic rats showed lower serum estrogen level, higher body weight than normal rats, and showed atrophy of uterine horns. DBE showed inhibitory effect on bone loss in osteoporetic condition, and reduced the increase of ALP activity and osteocalcin level in serum, and reduced the increase of OH-proline level in urine. But, DBE had no effect on cell proliferation and ALP activity in rat calvarial cell culture.
가미소요산(加味逍遙散)이 지단백산화(脂蛋白酸化)에 따른 RAW 264.7 활성(活性)에 미치는 영향(影響)

In this study, we investigated the hypothesis that gamisoyosan(GS) may reduce atherosclerosis by lowering the oxidiazability of LDL, To achive this goal, we examined the effect of GS on LDL oxidation, nitric oxide production in mouse macrophage cell line, RAW264.7, and the effect of GS on cupuric sulfate-induced cytotoxicity, LDH release, and macrophage activity. GS inhibited the generation of oxidized LDL from native LDL in RAW264.7 cell culture, and decreased the release of LDH from cupric sulfate-stimulated RAW264.7 cell. In other experiments, GS activated RAW264.7 cell, and prolonged the survival time, and increased nitric oxide production in Raw 264.7 cells.
