The purpose of this in vitro study was to evaluate contraction of the human gingival and periodontal ligament fibroblast in the collagen matrix. The cell cultures were prepared using human gingiva and tooth. The collagen gels were observed daily with an inverted microscope and their area was determined by measuring the diameters of the collagen gel. The configurations of human gingival and periodontal ligament fibroblasts were done by SEM observation. The results as follows; 1. The contraction was found to be proportional to seeding cell without seeding cells, the contraction was not occured. 2. Periodontal ligament fibroblast was shown more fast contraction than gingival fibroblast. 3. The collagen matrix was translucent initially, but with fully contraction, the structure become opaque. 4. The collagen matrix with fibroblast were aggregated to form bundles and reorganized collagen fiber.

만성 정신 분열병 환자에서 Risperidone교체 전후의 인지기능의 변화

Objectives : The aim of this study is to evaluate the effect of risperidone on cognitive functions in chronic schizophrenic patients after 8 weeks of treatments and determine whether any improvement on cognitive function relates to improvement in psychopathology. Methods : The subjects were 20 chronic schizophrenic outpatients, who participated in eight-week crossover study from conventional antipsychotics to risperidone. Clinical symptoms were assessed by Positive and Negative Syndrome Scale, Clinical Global Impression and Extrapyramidal Symptom Rating Scale, while they were receiving conventional antipsychotic treatment. Cognitive functions were assessed by K-WAIS digit span and digit symbol substitution test, and vigilance, continuous attention, reaction unit, corsi-block-tapping test. Patients were then reassessed after changing to a new treatment of risperidone. Results : After crossover from conventional antipsychotics to risperidone, positive and negative symptoms and the general psychopathology were significantly improved. The only item of Vienna tests significantly improved after risperidone treatment was corsi-block-tapping test. However, this improvement was not related to the improvement in psychopathology. Backward digit span score, digit span total score, digit symbol substitution score were significantly improved after risperidone treatment. But this improvement was not related to the improvement in psychopathology, except digit span total score. Digit span total score was related to the improvement in negative symptom score and PANNS total score. Conclusions : Treatment with risperidone appeared to exert a favorable effect on visuospatial memory and working memory, but long-term and larger sample replication study is necessary. 연구 목적: 정형 항정신병 약물을 복용중인 만성 정신 분열병 환자에서 risperidone 의 8주간의 치료가 신경인지기능에 미치는 영향을 파악하고, 이것이 임상증상의 변화와 연관되어있는지 여부를 알아보는 것이다. 방법 : 연구 대상은 정형 항정신병 약물을 복용하면서 안정적으로 생활하고 있는 만성 정신 분열병환자 20명이었다. 임상 증상은 치료 전후로 양성 및 음성 증후군 척도, 전반적 임상 인상 척도 및 추체외로 증상 평가 척도를 사용하여 평가하였다. 교체 투약 전후로 숫자 외우기 검사와 바꿔쓰기 검사, 경계력 검사, 지속적 주의력 검사, 반응 결정력 검사, Corsi 토막 두드리기 검사를 실시하였다. 결과 : 정형 항정신병 약물에서 Risperidone으로 교체 투약 8주후 임상증상은 양성척도, 음성척도, 일반 정신병리 척도에서 모두 유의한 호전을 보였다. 교체 투약후의 Vienna 검사 중 corsi 토막 두드리기 검사에서만 유의한 호전을 보였으나, 임상증상의 호전정도와 상관성을 보이지 않았다. 숫자 거꾸로 외우기 점수, 숫자 외우기 총 점수, 바꿔쓰기 점수가 유의한 호전을 보였으며, 숫자 외우기 총 점수만이 음성증상, PANNS 총 점수와 상관성을 보였다. 교체투약후 추체외로 증상 정도와 상관성을 보이는 인지기능 검사 항목은 없었다. 결론 : 만성 정신 분열병 환자에서 Risperidone 투약이 시공간 기억 및 작동기억에 우월한 효능이 있음이 제시되었으며, 장기간의 투여에 의한 연구가 필요할 것으로 생각된다
