Objectives: The aim of this study is to identify temporal trends in urinary cotinine levels and exposure factors among Non-smokers in Korean adults. Methods: Biological samples and questionnaire data were collected from representative Korean adults recruited in the Korean National Environmental Health Survey from 2009 to 2017. Multiple regression analysis was performed to determine the factors affecting urinary cotinine concentrations of non-smokers. Results: The urinary cotinine levels of Korean non-smokers decreased in Cycle 2 (1.04 µg/L, 2012-2014) compared to Cycle 1 (1.93 µg/L, 2009-2011) but increased slightly in Cycle 3 (1.37 µg/L, 2015-2017). Among the diverse variables, the main factors of cotinine exposure were secondhand smoke exposure and the presence of smokers in the family. Conclusions: The results of this study identified temporal trends in cotinine exposure among non-smokers in the Korean adult population. These findings will be used to develop further assessment and environmental health policies on secondhand smoke exposure.

Objectives : Osteoporosis is a systemic skeletal disease that decreases bone density and increases the risk of fractures. Bisphosphonates and SERMs are mainly used to treat osteoporosis, but, long-term use increases the risk of side effects such as jaw bone necrosis and breast cancer. Therefore, it is necessary to develop a therapeutic agent for a natural product with few side effects. Water extract of Lonicerae Japonicae Flos (wLF) was mainly found to have anti-cancer and anti-inflammatory effects. However, the effect of wLF on osteoporosis has not been elucidated. Therefore, this experiment investigated the effect of wLF on osteoclasts, osteoblasts and osteoporosis models. Methods : In order to study the effect of wLF on osteoporosis, the OVX-induced rat model was used for in vivo study. After 8 weeks, we measured body weight, uterine weight, liver weight, femur weight, bone density, trabecular area and tibia ash weight. To determine the effect of wLF on osteoclast differentiation, we measured the number of TRAP-positive cells and TRAP activity. To examine the effect of wLF on the expression of osteoblast-related genes, we measured the mRNA expression of alkaline phosphatase (ALP, Alpl) and osteocalcin (OCN, Bglap2). Results : In vivo experiment, wLF inhibited the reduction of femur weight, trabecular area, bone density and tibia ash weight. In vitro experiment, wLF had no significant effect on osteoclast differentiation. However, wLF increased the mRNA expression of Alpl and Bglap2 in MC3T3-E1 cell. Conclusions : This result suggested that wLF may be used for the treatment and prevention of postmenopausal osteoporosis.

A selective screening method based on a competitive enzyme-linked aptamer assay (ELAA) for dopamine (DA) in urine has been optimized and validated. Generally electroactive DA, uric acid (UA) and ascorbic acid (AA) are co-present in biological fluids such as blood and urine. Thus, it is important to develop a technique to selectively detect DA in the presence of AA and UA. In this paper, we report advantageous sensitivity and specificity of aptamer assays as compared to the existing antibody based-immunoassay. The RNA aptamer (67 mer) was immobilized via site-directed immobilization with biotin both at the 3’-end on aptamer and at neutravidin plate. Assay was performed with 0.01 μg mL-1 of aptamer and 1.205 × 10-7 M DA-HRP conjugate using the optimized method. A dose-response curve was constructed, and the limit of detection and a dynamic range for the DA were determined as 2.5 × 10-11 M and 4.0 × 10-7 M ~ 1.0 × 10-10 M, respectively. Competitive ELAA showed no matrix effect about AA and UA, which found in urine. Therefore, this paper describes usefulness of the aptamer assay in monitoring DA in urine.
