Authors experienced a 13-day-old boy who had suffered from walnut sized hard erythematous nodules on left axilla, anterior and posterior aspect of the neck which began to develop 9 days after birth. Histopathologic findings revealed subcutaneous fat necrosis and needle-shaped cleft in several fat cells. The skin lesions spontaneously resolved at 2 months of age. The clinical pattern, the skin manifestations, and the histologic appearance of the lesions were diagnostic of subcutaneous fat necrosis of newborn.

The epidermis, which is composed mainly of keratinocyt;es, undergoes continual proliferation and differentiation, To study keratinocyte differentiation, it is necessary to separate the keratinocytes at their sucessive stages of maturation. We separate the keratinocytes by layers by using Percoll and obtain the relatively pure fractions of basal cells, spinous cells, and granular cells, 76. 8%, 67. 2%, 30%. respectively.

Fox-Fordyce disease is a chronic pruritic papular ei*uption, usually appearing in women and presenting a strict localization to the areas bearing apocrine sweat glands. More than 90g, of the cases were seen in women, the ma.jority being in the age group of 13 to 35 years. The present report deals with unusual case of 14-year-old boy who developed Fox-Fordyce disease 6 years ago. To our knowledge, it is extremely rare case in Korea.

Background : Dermal papilla cells, which are mesenchyral components of the hair bulb are considered to play a fundamental role in the inductior hair growth. Objective : We studied how to establish a method of cuku e of dermal papilla cells. Methods : With human scalp tissue, we cultured dermal pipilla cells and identified them under the double-contrat microscopy and observed the epassion of eactin. Results : In vitro they spread slowly, initially as a moayer, and eventually formed multilayered paralled anys. At the edges of expanding colonies the cells were large and flattened and showed a tendency to form clumps. They expressed a-actin which is thought to be a marker of papilla cells, typeIII, typeIV colagen and laminin. Conclusion : These results suggest that using the female papilla cells, specially differentiated fibroblast, may be useful for the study of hau growth. (Kor J Dermatol 1995; 33(1); 28-32)

Juvenile xanthogranuloma is a benign disseminated xanthomatous and granulomatous condition of the skin and other organs. The skin lesions usually disappear spontaneously within 1 to 5 years. Our case was 4 month-old boy who had have match head to pea sized scattered yellowish papules and nodules on scrotum since age of 1 month and then these skin lesions gradually spread to trunk, face and scalp without subjective symptoms. Histopathologically, biopsy specimen of the trunk revealed massive accumulation of histiocytes at dermis. With fat stain, fat substance is demonstrated within the foamy histiocytes. The skull x-ray revealed coin sized, well circumscribed osteolytic lesion on the left parietal bone. By the age of two and half, most of the skin lesions and bone defect on skull x-ray disappeared spontaneously.

We evaluate the water content, hygroscopicity and water holding capacity in 40 healthy individuals and 17 leprosy patients by skin surface hydrometer(IBS Inc, Japan), and the correlation between the nerve destruction and the hydration state. In Eiealthy individuals the measuring sites were the flexor and extensor surfaces of the arm, forearm, thigh and leg, In leprosy patients we measured on the extensor surface of forearm with shows abnormal sensation, and opposite ar neighboring skin which shows normal sensation. The results are as follows: 1. The water content of healthy individuals is highest in the flexor surface af arm (49. 50 + 18.38ug ) and lowest in the extensor surface of leg(30.75 -12.09ug) 2. The hygrosccpicity of healthy individuals is highest in the flexor surface of arm(296. 30 + 70.75ug ) and lowest in the extensor surface of leg(24$. Q7+5) 5ppg) 3. The water content of the extensor surface of lorearm in leprosy patients, which .hows abnormal sensation, is decreased(16. 35 +20. 82 ug) and it is statisti calli significant when compared with healthy individuals(p<0.005). 4. The hygroscopicity of the extensor surface of forearm in leprosy patients. which shows abnormal sensation, is decreased(94. 29 + 71. 62 ug) and it is statistically significant when compared with opposite or neiphboring skin and healthy individuals(p<0. 005)
각질형성세포에서 HLA - DR 항원의 발현이 IL - 1 생산에 미치는 영향에 관한 연구

N/A There is increasing evidence that keratinocytes may parcipitate in the inflammatory processes and immune response of skin. Keratinocytes can be induced to express functional immunocompetent molecules such as HLA-DR antigen. In additior, they secrete a wide range of infiammatory cytokines including IL-1. In order to better understand the immunologic role of keratinocyte, it is necessary to quantitate the amount of IL-1α produced by cultured human keratinocytes and evalute the effect of HLA-DR expression on the production of IL-1α by keratinocytes. In this study, we employed the radioimmunoassay to measure the amount of IL-1α produced by cultured human keratinocyte. Human keratinocytes were isolated from neonatal foreskins and grown in MCDB 153 medium. HLA-DR expression was induced by 30 U/㎖ of r-IFN-?. The supernatants were assayed for IL-1α. The results were as follows : 1. When culture medium was not replaced until 72 hours, cell numbers of kerantinocytes without treatment of r-IFN-? were 0.80±0.26×10^6, 1.16±0.21×10^6, 1.33±0.21×10^6 at 24, 48 and 72 hours, respectively. When culture medium was replaced every 48 hours, cell numbers of keratinocytes with treatment of r-IFN-? were 0.72±0.29×10^6, 1.06±0.26×10^6 and 1.06±0.25×10^6 at 24, 48 and 72 hours, respectively. Antiproliferative effect of r-IFN-? was significant at 48 and 72 hours(p=0.046, p=0.003). When culture medium was replaced every 48 hours, cell numbers of keratinocytes without treatment of r-IFN-? were 1.16±0.21×10^6, 1.69±0.27×10^6, 2.01±0.18×10^6 at 48, 96 and 144 hours, respectively. Cell numbers of keratinocytes with treatment of r-IFN-? were 1.06±0.26×10^6, 1.51±0.20×10^6 and 1.67±0.17×10^6 at 48, 96 and 144 hours, respectively. A?tiproliferative effect of r-IFN-? was significant at 48 and 144 hours(p=0.046, p=0.0001). 2. Relatively lower concentration of IFN-?(30 U/㎖) can induce HLA-DR expression on keratinocytes. The percent of HLA-DR positive keratinocytes were 42.15±4.87% by 48 hours. Then it decreased to 14.26±2.85% by 144 hours. 3. When culture medium was not replaced, the levels of IL-1α from supernatants of cultured keratinocytes were 1.513±0.774, 0.778±0.299 and 0.720±0.274 fmol/10^7cells at 24, 48 and 72 hours, respectively. The level is highest at 24 hours and then i? decreased. 4. When the percentage of HLA-DR positive keratinocytes were relatively high 42.15±4.87% and 38.59±3.74% at 48 and 72 hours, respectively, the levels of IL-1α were 1.009±0.320 and 1.002±0.301 fmol/10^7cells, respectively. They were significantly higher than r-IFN-? untreated groups, of which IL-1α levels were 0.778±0.299 and 0.720±0.274 fmol/10^7cells, respectively(p=0.059, p=0.007). When the percentage of HLA-DR positive keratinocytes were decreased, the differences of IL-1αlevel between two groups were not significant. In summary, the results indicate that the amount of IL-1α produced by keratinocytes are relatively low and keratinocytes may participate in the immune process of the skin, and that the immunologic role of HLA-DR^+ keratinocytes might be media?ed via an increased production of IL-1α from HLA-DR^+ keratinocytes.

The authors investigated the antiproliferative effect and expression of HLA-DR an- tigen by recombinant gamma-interferon (r-IFN-y) on cultured human keratinocytes (KC). The results were as follows, 1. From 10l.J/ml of r-1FN-p exposure, the proliferation of KC decreased in a concentration dependent fashion. But there was little difference of antiproliferative effect above 30U/ml of r-IFN-y exposure. 2. The expression of HLA-DR antigen on KC increased in a concentration and time dependent fashion of r-IFN-p exposure. E3ut t,here was little difference of HLA-DR antigen expression on KC above 30tJ/ml and most of HLA-DR antigen were expressed within 48hr. 3. The opt,imal condition for HLA-DR antigen induction on KC by r-IFN-p was likely t,hat HLA-DR KC was observed at 48hr under the our exposure of 30U/ml of r-IFN p. 4. After 4hr exposure of 30U/ml of r-IFN-p, KC expresed HLA-BR. antigen, reaching a maximum intensity at 3 days. At, 7 days, the loss of HI A-DR KC showed over 90% of maximum intensity.
