Characteristics of Eggshell Powder as Carriers of Probiotics

계란 가공 부산물인 난각(ES: Eggshell)은 탄산칼슘 함량이 높아 사료에 첨가하여 칼슘원으로 이용되고 있다. 본 연구에서는 ES를 생균제의 부형제인 운반체로서 활용 가능성을 처음으로 시도하였다. L. plantarum을 대두박(SBM: Soybean meal), 난각조각(ESL: Eggshell powder with large particles), 난각미세분말(ESF: Eggshell powder with fine particles), 그리고 이들의 복합운반체인 SBM+ESL과 SBM+ESF에 생균제를 흡착시켜 그 부착상태를 주사전자현미경으로 확인하였다. 이 중 복합운반체인 SBM+ESF는 상온에서 4주 동안 pH 7~8을 유지하면서 L. plantarum의 가장 높은 생존율을 보였다. 본 연구에 사용한 모든 생균제들은 보존기간 동안 4℃에서는 높은 생존율을 보였다. 30℃에서는 유산균수는 크게 감소하였으나, B. licheniformis는 높은 생존율을 유지하였고 B. subtilis, B. amyloliquefaciens와 S. cerevisiae는 2 log10 (CFU/g)정도 감소하였다. 상기 연구결과는 사료의 칼슘원으로 이용되는 난각미세분말(ESF)을 대두박과 혼합하여 사용하면 B. licheniformis를 비롯한 일부 생균제의 생존성을 향상시켜 부형제(운반체)로도 사용할 수 있음을 밝혔다. Eggshell (ES) is a by-product of table eggs with high content of calcium carbonate which can be used as a calcium source in feed. In this study, we have first illuminated the potential application of ES as a novel carrier for probiotics. The carriers used in the study include a SBM (Soybean meal), ESL (Eggshell powder with large particles), ESF (Eggshell powder with fine particles), and the complex carriers (SBM+ESL, SBM+ESF). The structure of carriers absorbed by L. plantarum was confirmed by SEM image. Among these carriers, the complex carrier SBM+ESF showed the highest viability of L. plantarum with pH 7~8 during four weeks storage at room temperature. The SBM+ESF was further tested as a carrier for various probiotic strains at 4℃ or 30℃. All the probiotic strains showed high viability at 4℃ storage. However, a significant reduction of Lactobacillus cells was observed at 30℃ storage. B. lichenifomis maintained high viability whereas B. subtilis, B. amyloliquefaciens, and S. cerevisiae showed the reduction of 2 log10 (CFU/g). These results suggest that if the ESF as a calcium source in feed was mixed with SBM, it can be used as an effective complex carrier for improving the viability of some probiotics including B. licheniformis.

Six experimental diets for juvenile sea cucumbers Apostichopus japonicus were evaluated and compared with current commercial diets. Sea cucumbers (50 per tank; initial mean weight $2.3{\pm}0.1g$) in three replicates of seven groups were fed one of six experimental diets (ED1, 20% wild-harvested Sargassum thunbergii powder; ED2, 20% commercial S. thunbergii powder; ED3, 20% Undaria pinnatifida and Ascophyllum nodosum powders; ED4, 40% U. pinnatifida and A. nodosum powders; ED5, 10% brewer's yeast; and ED6, 10% sea shell powder) or a commercial diet (CD) for 28 weeks. The survival rate in all groups was 85-94%, with no significant difference among the experimental groups. The mean body weight of the sea cucumbers was significantly different among the experimental groups 6 weeks after the feeding trial. The final mean weight of the sea cucumbers was highest in ED1, ED3, and ED4, followed by ED6, ED5, ED2, and CD, in decreasing order. The results of this study suggest that U. pinnatifida and A. nodosum powders could provide a substitute for the S. thunbergii powder commonly used in commercial sea cucumber diets. Thus, the EP4 diet formulation, which contains no S. thunbergii, could be used as a practical feed for juvenile sea cucumbers.

By-products produced from medicinal plants (MP) were evaluated as feed additives on growth performance, blood chemistry, and immune responses of the juvenile red lip mullet Liza haematocheila. A commercial diet was used as the control. Four experimental diets were prepared by combining 0.5% of MPs such as Panax ginseng (PG), Schisandra chinensis (SC), Angelica gigas (AG), and Rehmannia glutinosa (RG), via water absorption, to the control diet. Triplicate groups of fish averaging 61.8 g were fed one of experimental diets to apparent satiation for 8 weeks. Dietary supplementation of MPs did not affect the growth performance, biological indices and whole body composition of fish compared to the control. The fish fed with AG diet had an increased weight gain, and specific growth rate compared to fish in SC group. Decreased level of plasma glucose was observed in fish fed diet containing MPs. Lysozyme activity was significantly increased in the fish fed AG diet compared to that of fish in PG group. These results indicate that dietary supplementation of Angelica gigas may enhance growth performance and immune responses in juvenile red lip mullet.

This study was conducted to investigate the effects of extrusion pressure and particle size of feed ingredients on the growth performance and plasma hormone activity in juvenile olive flounder Paralichthys olivaceus. Experimental diets were prepared with extrusion pressure manipulated by screw speed [low pressure (LP), 885 rpm/min; high pressure (HP), 708 rpm/min] and different dietary particle sizes [specific surface area: small (SS), 169.9 ㎡/kg; large (LS), 67.4 ㎡/kg] in a two-level factorial design. Four experimental diets (LP+SS, LP+LS, HP+SS, and HP+LS) were randomly assigned to 12 tanks (3 replicates) stocked with 20 fish (initial weight, 57 g) per tank. After a 4-week feeding trial, the observable trends of the main effects of extrusion pressure and particle size on growth performance showed that LP and SS enhanced fish weight gain. The plasma insulin-like growth factor-I level was significantly higher in fish fed the LP+SS diet than in fish fed the HP+SS diet. These results indicate that manipulation of the physical qualities of feed through adjustment of extrusion pressure and feed ingredient particle size may influence the growth performance of juvenile olive flounder, which should be considered in feed manufacture.

This study aimed to evaluate how fish meal (FM) replacement in diets with a mixture of animal and plant protein sources affect growth performance, feed utilization, hematological parameters and innate immunity of red seabream Pagrus major. A control FM diet was formulated to contain 65% FM (Con). Two other diets were prepared replacing FM in the control diet with a mixture of protein sources (wheat gluten, soy-protein concentrate, tankage meal, and poultry by-product meal) by 30 and 40% (FM30 and FM40, respectively). Total 300 red seabream (body weight, 77.6±0.3g) were distributed to 12 tanks (300 L) in 4 replicates per diet. The fish were fed the diets to apparent satiation for 19 weeks. After the feeding trial, no significant differences could be observed in growth performance, feed utilization, hematological parameters, innate immunity, and survivals among all the dietary treatments. This long-term feeding trial at low water temperature (13.8-17.5°C) indicates that a proper mixture ratio of wheat gluten, soy protein concentrate, tankage meal, and poultry by-product meal can replace FM up to 40% in red seabream diets.
