cadmium에 내성을 보이는 5개의 균주를 동정하고, 그 특성을 조사한 결과는 다음과 같다. 동정 결과 AD1균주는 Alcaligenes paradoxus로, ED1 균주는 Escherichia coli로, 그리고 AD2, BD1, FD1 균주는 Pseudomonas aeruginosa로 동정하였다. 5개 균주의 cadmium에 대한 내성 범위를 조사하였더니, AD1과 ED1은 0.5mM 이상의 농도에서는 약하게 자랐고, AD2, BD1, FD1은 1mM의 농도에서도 자랐다. Cadmium에 내성이 강한 균주들은 CdCl2의 농도가 증가함에 따라 lag phase가 길어졌다. 동정된 균주들의 중금속에 대한 내성 여부를 조사해본 결과 AD2, BD1 및 ED1은 사용한 중금속 모두에 내성을 나타내었고, AD1은 Cr에 대해 그리고 FD1은 Cu와 Ni에 대해 내성이 없었다. 또한 항생제에 대한 내성 조사 결과 BD1은 ampicillin, chloramphenicol, kanamycin, streptomycin, tetracycline 모두에 내성을 나타냈고, AD1은 ampicillin, chloramphenicol, streptomycin에 대해 약하게 내성을 보였다. AD2는 ampicillin에만 내성을 보였으며, ED1과 FD1은 사용한 항생제에 대해 내성이 없었다. cadmium 내성을 나타내는 균 중 BD1에만 plasmid 1개가 확인되었다. 그래서 BD1의 plasmid를 제거하기 위해 MIC를 측정한 결과 acridine orange에는 2ppm, mitomycin-C에는 0.1ppm보다 훨씬 낮은 농도, 그리고 SDS는 0.04%보다 낮은 농도로 나타났다. cadmium 내성과 이 plasmid의 관련성을 입증하기 위해 curing과 형질 전환 실험을 실시했지만 관련되어 있다는 증거를 발견할 수 없었다. 따라서 Pseudomonas aeruginosa BD1이 나타내는 cadmium 내성은 plasmid가 아닌 chromosome상에 존재하는 유전자 때문으로 판단된다. The Identification and characterization of five strains which were resistant to cadmium were studied. The results were as follows. AD1 strain was identified as Alcaligenes paradoxus, ED1 strain was Escherichia coli and AD2, BD1 and FD1 strains were Pseudomonas aeruginosa. AD2, BD1 and FD1 showed growth at the 1mM cadmium concentration but AD1 and ED1 showed weak growth when the cadmium concentration was 0.5mM or over. The strains which were resistant to cadmium showed delayed lag phase as the concentration of cadmium increased. AD2, BD1 and ED1 strains were resistant to all kind of heavy metals studied, and AD1 was not resistant to Cr, and FD1 was not to Cu and Ni. BD1 strain was resistant to ampicillin, chloramphenicol, kanamycin, streptomycin, tetracycline, and AD1 showed weak resistance to ampicillin, chlormphenicol and streptomycin. AD2 strain was reisistant to ampicillin only, but ED1 and FD1 strains showed no resistance to antibiotics studied. BDI strain harvored one plasmid. For the plasmid curing, MIC of BD1 strain to each curing agent was measured. MIC of acridine orange, mitomycin-C, and SDS were 2ppm, lower than 0.1ppm, and 0.04% respectively. To confirm the relationship between the plasmid and cadmium resistance of BD1 strain curing and transformation test were performed, but there was no evidence of relation. The resistance to cadmium of BD1 strain was due to not plasmid but chromosome gene.
