DSS(Dextran Sodium Sulfate)로 유발된 마우스 대장염에서 WK-20이 갖는 치료효과

염증성 장질환은 장관 내 비정상적인 만성 염증이 호전과 재발을 반복하는 질환으로 아직까지 명확한 발병기전은 밝혀져 있지 않다. 그 중 궤양성 대장염은 염증 및 혈변, 설사 심한 경우에는 발열을 동반한다. 현재 치료제로는 설파살라진, 메살라민과 같은 항염 증제, 항생제 등을 사용하고 있지만, 부작용으로 두통, 빈혈 등의 있고 드물게는 간염, 폐렴 등이 유발되기 때문에 보다 안전한 방법이 요구되는 실정이다. 본 실험은 5주령의 수컷 마우스를 일주일간 적응기간 후, CON군 DSS군, 저농도군, 중농도군으로 나누었다. CON군을 제외한 모든 군에 DSS를 일주일간 투여해 장염을 유발한 후 2주간 WK-20을 투여한 후 검사를 한 결과 저농도, 중농도군에서 대장염의 증상이 유의적으로 완화된 것으로 보였다. 따라서, WK-20은 DSS를 통해 유발되는 대장염에서 치료적 효과를 보이지만 MTT 검사를 통해 농도의 조절이 필요한 것으로 사료된다.
db/db마우스에서 Aspergillus awamori로 발효한 황기 추출물의 항당뇨 및 지질개선 효능

Background: This study investigated the anti-diabetic effects of Astragalus membranaceus (Fisch.) Bunge fermented by Aspergillus awarmori in db/db mice. Methods and Results: Astragalus membranaceus roots were fermented with A. awarmori for 4 days and extracted with hot water. The male mice were divided into five groups: db/m (non-diabetic db/+mice), db/db (C57BLKs/J db/db mice), MT (db/db + 1% metformin), AM (db/db + 1% A. membranaceus extract), and FAM (db/db + 1% fermented A. membranaceus extract). After 10 weeks of treatment, we measured the body weight, food intake, and blood glucose, hemoglobin A1C (HbA1c), plasma insulin, adiponectin, and transaminase levels. Oral glucose tolerance test was performed and serum lipid profile was determined. As expected, the AM and FAM groups had significantly reduced body weight, fasting glucose, lipid content, and HbA1c, transaminase, and serum insulin levels, and increased serum adiponectin levels. Conclusions: Therefore, based on these research results, A. membranaceus extract and fermented A. membranaceus extract can be used to prevent and treat type 2 diabetes by reducing blood glucose levels and lipid content.

This study sought to investigate the antioxidant and immune-stimulatory effects of different Curcuma longa (C. longa) extracts based on their color (random, light, medium, dark orange), the soil of cultivation (clay, granite), drying temperature (50∼80℃), and extraction methods (water, ethanol). The antioxidant factors of C. longa were determined by the total phenolic content and DPPH and ABTS radical scavenging activities. The nitric oxide (NO) production of the macrophage cell line (RAW264.7) and the proliferation of splenocytes were investigated to assess the immune-enhancing effects of C. longa. The antioxidant activity was higher in the dark color and the ethanol extracts. All the types of extracts were nontoxic and enhanced the production of NO in RAW264.7 at less than 250 μg/mL of C. longa. When mouse splenocytes were exposed to various concentrations of hot water or ethanol extracts of C. longa with media, concanavalin A (Con A), lipopolysaccharide (LPS), and cyclophosphamide (CPA), their proliferation increased. Specifically, the 70% ethanol extract of C. longa showed the highest levels of splenocyte proliferation among all samples. These results suggest that the antioxidant and immune-stimulating activities of C. longa were most affected by the extracting solvent rather than color, soil in which it was cultured, and drying temperature. Appropriate extraction methods for C. longa should be researched further to develop good healthy foods with high antioxidant and immune-enhancing activities.
