      • Studies on Histones from Normal and Myelocytic Leukemia Leucocytes

        정상인의 백혈구와 골수성 백혈병 환자의 백혈구에서 histone을 추출하여 비교 관찰하였다. Johns씨 법에 의해 5분획의 histone을 얻어 polyacrylamide gel electrophoresis로 그 순도와 이동도를 확인하였다. 정상 및 골수성 백혈병 환자의 백혈구에서 추출한 histone의 총량은 각각 37.08 mg/g 및 58.64 mg/g packed cells 이었다. 각 histone 분획의 상대적 함유량은 골수성 백혈병 환자의 빽혈구에서는 정상인의 백혈구에 비해 f2b 분획이 증가하고 f1 및 f3는 감소하였다. 아미노산 조성을 보면 정상인 백혈구의 것에 비해 골수성 백혈병 환자의 백혈구의 것은 (1) f1 분획에서는 phenylalanine이 증가하고 arginine과 glycine은 감소하며 산성 아미노산 대 염기성 아마노산의 비율은 같으나 arginine 대 glycine의 비율은 4.05에서 11.85로 증가하였으며 (2) f2a1과 f2a2는 아마노산 조성이 거의 같으며 (3) f2b에서는 isoleucine의 양이 적고 (4) f3에서는 histidine의 양이 증가하였다. $\epsilon$-N-mono 및 $\epsilon$-N-dimethyllysine이 정상인의 백혈구에서는 보이지 않으나 골수성 백혈병 환자의 백혈구의 f3 분획에서 발견되었다. 일반적으로 암 조직에서 methylase 활성이 높다는 사실을 고려하면 골수성 백혈병 백혈구의 histone 분획내의 methylated derivatives의 존재는 흥미 있는 사실일 것이다. Comparative studies on histone from normal leukocyte and chronic myelocytic leukemic granulocyte were done. Five main fractions of histone were obtained by the method of Johns. And each of the fractions was confirmed of its homogeneity and its distinct electrophoretic mobilities on polyacrylamide gel electrophoresis. Total amounts of histone isolated from normal and leukemic cells were 37.08 mg and 58.64 mg per one gram of packed cells, respectively. Relative yields of histone fractions of leukemic cells revealed marked increment of f2b fraction and concurrent decrements of f1 and f3 fractions as compared to those of normal WBCs. Phenylalanine was increased, and arginine and glycine were decreased in f1 fraction of leukemic cells as compared to those of normal WBCs. Ratio of acidic to basic amino acids was not changed but the ratio of arginine to lysine was changed from 4.05 in f1 of normal WBCs to 11.85 in that of leukemic cells. F2b of leukemic cells contained lesser amount of isoleucine, and f3 of leukemic cells contained higher amount of histidine, as compared to that of normal WBCs, respectively. The composition of amino acids of f2a1 and f2a2 from leukemic granulocyte was similar to that of normal WBCs. $\epsilon$-N-mono- and $\epsilon$-N-dimethyllysine were observed in f3 of leukemic cells, which was not observed in normal WBCs. As methylase activity is increased in cancerous cell in general, the presence of methylated derivatives of lysine in leukemic cells in contrast to the absence of methylated derivatives in normal WBCs may have significant meanings.
